잠재적으로 심각한 임상적 영향에도 불구하고, 부분적으로 물류 및 기술적 한계로 인해 전이-전이 파종에 대해 알려진 바가 거의 없습니다. 사람을 대상으로 한 연구는 임상 샘플의 부족으로 인해 한계가 있습니다. 전이성 병변의 임상적 절제와 생검은 드물며, 겉보기에 건강해 보이는 장기의 생검도 드물며, 단일 파종된 종양 세포가 숨어 있을 수 있습니다. 즉, 인간 대상 연구는 일반적으로 원발성 종양이 아직 제자리에 있거나 이전에 절제되었지만 연구자가 여전히 사용할 수 있는 개인의 부검 샘플을 사용해서만 가능합니다. 이러한 샘플을 이용할 수 있는 경우, 암 진행에 대한 계통 분석은 염기서열 분석 방법14을 사용하여 수행되어야 한다. 그러나 일치하는 원발성 종양 및 전이의 대량 염기서열 분석은 포괄적인 계통 추적에 필요한 민감도를 가지고 있지 않습니다. 예를 들어, 하나의 병변에 대한 대량 염기서열 분석은 일치하는 병변 중 어느 것에서도 검출할 수 없는 서브클론을 드러낼 수 있습니다. 이 경우 이 서브클론의 기원을 확인할 수 없습니다. 원발성 종양 또는 다른 전이에서 검출 한계 이하의 빈도로 존재했을 수도 있고, 발견된 전이성 병변의 초기 집락화 이후에 발생했을 수도 있습니다. 단일 세포 염기서열분석은 향상된 감도를 제공하지만 높은 비용으로 인해 이 기술의 대규모 적용이 제한됩니다. 이러한 연구의 후향적 특성은 또한 일시적인 전이성 사건과 서로 다른 시점의 질병 환경에 대한 제한된 통찰력을 제공한다는 것을 의미합니다.

이러한 새로운 방법은 암 진행에 대한 상세한 고해상도 계통발생 지도를 생성할 수 있는 큰 잠재력을 가지고 있지만, 전이에서 전이로의 파종 사건의 정확한 시기와 이를 촉진하거나 예방하는 요인을 연구하는 사람들에게는 매우 비실용적입니다. 이러한 지식 격차를 해소하는 것은 전이성 암에 대한 이해와 치료를 개선하는 데 매우 중요하지만 이러한 연구를 촉진하는 기술이 눈에 띄게 부족합니다. 이러한 요구를 해결하기 위해 우리는 최근에 전이 부위(폐)에서 광변환을 통해 종양 세포를 특이적으로 표시한 후 3차 장기에서 재식별할 수 있는 새로운 기술을 개발하여 여기에 제시합니다. 이 기술을 사용하여 우리는 최근 유방암 세포가 폐 전이와 종자 3차 장기에서 재증식한다는 것을 보여주었다13. 이 기술은 또한 좁은 창 내에서 재확산 사건의 타이밍을 결정하고 재확산된 종양 세포를 정량화하는 데 사용할 수 있으며, 재확산된 세포의 유기 영양성 및 재확산을 촉진/방지하는 요인에 대한 연구를 용이하게 합니다.
하나의 형광 단백질을 다른 형광 단백질로 영구적으로 대체하는 광변환 및 국소 유도 cre/lox 시스템은종양 세포를 표시하고 추적하는 데 이전에 사용되었지만, 우리가 아는 한, 종양 세포의 시공간 표시에 대한 접근 방식은 가장 흔한 14가지 암 중 하나로 진단된 남성과 여성에게 가장 흔한 전이 부위 중 하나인 폐를 표적으로 삼도록 최적화되지 않았습니다24. 모든 암세포 유형과 폐 전이 생성을 위한 모든 프로토콜을 당사 절차와 함께 사용할 수 있으므로 전이 연구자에게 광범위하게 유용합니다. 폐 전이를 생성하는 데 사용되는 모든 암세포는 광전환 또는 광전환 가능한 단백질을 발현해야 하며, 연구자들은 특정 필요와 자원에 따라 사용할 단백질을 선택할 수 있습니다. 이 연구에서는 히스톤 H2B에 태그된 광변환 가능한 녹색-적색 형광 단백질 Dendra2(6DT1-Dendra2 세포)25를 안정적으로 발현하는 6DT1 유방암 세포를 사용했습니다. 5.0 × 104 6DT1-Dendra2 세포를 암컷 Rag2-/- 마우스의 네 번째 유방 지방 패드에 주입했습니다. 원발성 종양은 주사 후 12일에서 16일 사이에 만져질 수 있었고 실험 기간 동안 절제되지 않았습니다. 자발적 폐 전이는 종양 세포 주입 후 19일에서 26일 사이에 발생했습니다. 광전환 수술은 종양 세포 주입 후 26일에서 29일 사이에 수행되었습니다. 마우스는 폐 전이 부담으로 인해 수술 후 72시간 이내에 희생되었습니다.
