Los modelos de cultivo in vitro en 3D, que proporcionan características estructurales y morfológicas más similares a las de in vivo en comparación con los modelos de cultivo 2D convencionales, han sido reconocidos como sistemas prometedores en diversas aplicaciones biomédicas, como la ingeniería de tejidos, el modelado de enfermedades y el cribado de fármacos 1,2,3. Como un tipo de modelo de cultivo 3D, los esferoides celulares suelen referirse a la agregación celular, creando estructuras esferoidales 3D caracterizadas por interacciones mejoradas célula-célula y célula-matriz 4,5,6. Por lo tanto, la fabricación de esferoides celulares se ha convertido en una poderosa herramienta para permitir diversos estudios biológicos.
Para obtener esferoides se han desarrollado diversas técnicas, como la gota colgante7, las placas no adhesivas8 o los dispositivos de micropocillos9. En principio, estos métodos suelen facilitar el ensamblaje celular mediante la utilización de fuerzas físicas como la fuerza gravitacional, al tiempo que minimizan las interacciones entre las células y el sustrato. Sin embargo, a menudo implican procesos que requieren mucha mano de obra, tienen baja productividad y plantean desafíos para controlar el tamaño del esferoide10,11. Es importante destacar que la producción de esferoides con el tamaño y la uniformidad deseados en cantidad suficiente es de suma importancia para satisfacer aplicaciones biológicas específicas. A diferencia de los métodos mencionados anteriormente, las ondas acústicas, como un tipo de técnica impulsada por fuerzas externas 12,13,14, han mostrado potencial para la fabricación masiva de esferoides celulares con alta calidad y rendimiento, basados en el principio de mejorar la agregación celular a través de fuerzas externas 15,16,17,18 . A diferencia de las fuerzas electromagnéticas o magnéticas, las técnicas de manipulación celular basadas en la acústica no son invasivas y no contienen marcadores, lo que permite la formación de esferoides con una excelente biocompatibilidad19,20.
Comúnmente, se han desarrollado dispositivos basados en ondas acústicas de superficie estacionaria (SAW) y ondas acústicas masivas (BAW) para generar esferoides, utilizando los nodos acústicos (AN) producidos por los campos acústicos permanentes correspondientes 21,22,23. En particular, los dispositivos de ensamblaje acústico basados en BAW, con las ventajas de una fabricación conveniente, fácil operación y excelente escalabilidad, han ganado atención para la fabricación de esferoides celulares24,25. Recientemente hemos desarrollado un dispositivo de ensamblaje acústico fácil basado en BAWs con la capacidad de generar esferoides con alto rendimiento26. El dispositivo propuesto consiste en una cámara cuadrada de polimetacrilato de metilo (PMMA) con tres transductores de titanato de circonato de plomo (PZT) dispuestos respectivamente en el plano X/Y/Z. Esta disposición permite la creación de un patrón de matriz de puntos 3D de nodos acústicos levitados (LAN) para el ensamblaje de celdas de conducción. En comparación con los dispositivos basados en BAW o SAW informados anteriormente, que solo pueden crear una matriz 1D o 2D de ANs 27,28,29, el dispositivo actual permite una matriz de puntos 3D de LAN para la rápida formación de agregados celulares dentro de la solución de gelatina de metacriloilo (GelMA). Posteriormente, los agregados celulares maduraron en esferoides con alta viabilidad dentro de los andamios fotocurados GelMA después de tres días de cultivo. Por último, se obtuvo fácilmente un gran número de esferoides de tamaño uniforme a partir de los andamios GelMA para aplicaciones posteriores.

three-dimensional acoustic assembly device for mass manufacturing of cell spheroids

Los esferoides celulares son modelos tridimensionales (3D) prometedores que han adquirido amplias aplicaciones en muchos campos biológicos. Este protocolo presenta un método para fabricar esferoides celulares de alta calidad y alto rendimiento utilizando un dispositivo de ensamblaje acústico 3D a través de procedimientos maniobrables. El dispositivo de montaje acústico consta de tres transductores de titanato de circonato de plomo (PZT), cada uno dispuesto en el plano X/Y/Z de una cámara cuadrada de polimetacrilato de metilo (PMMA). 3D in vitro culture models, which provide more in vivo-like structural and morphological characteristics compared to conventional 2D culture models, have been recognized as promising systems in various biomedical applications such as tissue engineering, disease modeling, and drug screening1,2,3. As one type of 3D culture model, cell spheroids typically refer to cell aggregation, creating 3D spheroidal structures characterized by enhanced cell-cell and cell-matrix interactions4,5,6. Therefore, fabricating cell spheroids has become a powerful tool for enabling diverse biological studies.
Various techniques, including hanging drop7, non-adhesive plates8, or microwell devices9, have been developed to obtain spheroids. In principle, these methods commonly facilitate cell assembly by utilizing physical forces such as gravitational force while minimizing interactions between cells and the substrate. However, they often involve labor-intensive processes, have low productivity, and pose challenges for controlling spheroid size10,11. Importantly, the production of spheroids with the desired size and uniformity in sufficient quantity is of utmost importance to satisfy specific biological applications. In contrast to the above-mentioned methods, acoustic waves, as one type of external-force-driven technique12,13,14, have shown potential for mass manufacturing of cell spheroids with high quality and throughput, based on the principle of enhancing cell aggregation through external forces15,16,17,18. Unlike electromagnetic or magnetic forces, acoustic-based cell manipulation techniques are non-invasive and label-free, enabling spheroid formation with excellent biocompatibility19,20.
Commonly, standing surface acoustic waves (SAWs) and bulk acoustic waves (BAWs)-based devices have been developed to generate spheroids, utilizing the acoustic nodes (ANs) produced by corresponding standing acoustic fields21,22,23. Particularly, acoustic assembly devices based on BAWs, with the merits of convenient manufacture, easy operation, and excellent scalability, have gained attention for fabricating cell spheroids24,25. We have recently developed a facile BAWs-based acoustic assembly device with the ability to generate spheroids with high throughput26. The proposed device consists of a square polymethyl methacrylate (PMMA) chamber with three lead zirconate titanate (PZT) transducers arranged respectively in the X/Y/Z plane. This arrangement enables the creation of a 3D dot-array pattern of levitated acoustic nodes (LANs) for driving cell assembly. Compared to previously reported BAWs- or SAWs-based devices, which can only create a 1D or 2D array of ANs27,28,29, the present device enables a 3D dot-array of LANs for rapid cell aggregate formation within the gelatin methacryloyl (GelMA) solution. Subsequently, cell aggregates matured into spheroids with high viability within the photocured GelMA scaffolds after three days of cultivation. Finally, a large number of spheroids with uniform size were easily obtained from the GelMA scaffolds for downstream applications.

3D in vitro culture models, like cell spheroids, have been widely used in the fields of tissue engineering, disease modeling, and drug screening. Our research focuses on developing and invest per engineering method to fabricate spheroids or other 3D in vitro culture models with high quality for diverse biological studies. In the current protocol, the spheroids grew within the GelMA scaffold and are retrieved at the final layback, dissolving the scaffold, causing additional procedures.We will try to acoustically assemble cell aggregates in a cultural medium to enable them to grow within spheroids without scaffold. Compared to magnetic assembly techniques, acoustic waves can directly assemble cells in a label-free manner without potential toxic effects on cells. Besides, our protocol enables cell spheroids to be assembled in a three-dimensional array, substantially increasing throughput.
