Dette prosjektet tar sikte på å gi en noelle-protokoll for storskala produksjoner av adeno-assosierte virusvektorer med høy titer og høy renhet ved bruk av en cellefabrikkplattform og optimaliserte kulturelle forhold. Den reviderte renseprosessen kombinerer AEV fra begge cellefraksjoner og ansettelsesmerket tofasebegjæring, etterfulgt av vår gradientartifisering som oppnår overlegent utbytte og renhet. Til tross for nylige fremskritt, gjenstår noen utfordringer i akademiske forskningslaboratorier.
Disse inkluderer optimalisering av skalerbare kulturforhold, bruk av andre celler, reduksjon av prosentandelen tomme kapsider og balansering av virusutbyttet med renhet. Sammenlignet med andre teknikker tilbyr protokollen vår skalerbarhet via cellefabrikker, økt utbytte med optimaliserte kulturforhold og forbedret adeno-assosiert virusvektorrenhet gjennom en noelle to-trinns renseprosess. Denne skalerbare protokollen med overlegen adeno-assosiert viral renhet banet vei for større prekliniske studier i de akademiske forskningslaboratoriene.
