<p class="jove_content">Optische beeldvorming (OI) is een eenvoudige, snelle en goedkope tool voor in vivo monitoring van nieuwe stamceltherapieën. De techniek is gebaseerd op ex vivo etikettering van stamcellen met een fluorescerende kleurstof, latere intraveneuze injectie van de gelabelde cellen en visualisatie van hun accumulatie in specifieke doelorganen of pathologieën. De gepresenteerde techniek laat zien hoe we menselijke mesenchymale stamcellen (hMSC) label door eenvoudige incubatie met de lipofiele fluorescerende kleurstof DiD (C67H103CIN2O3S) en hoe we menselijke embryonale stamcellen (hESC) label met de FDA goedgekeurde fluorescerende kleurstof Indocyanine Green (ICG). De opname mechanisme is via de hechting en de verspreiding van de lypophilic kleurstof over de fosfolipide celmembraan bilaag. De etikettering rendement is meestal verbeterd als de cellen worden geïncubeerd met de kleurstof in serum-vrije media, in tegenstelling tot incubatie in serum-bevattende media. Bovendien is de toevoeging van de transfectie-agent protaminesulfaat verbetert aanzienlijk contrastmiddel opname.</p>
