Single-cell elektroporatie (SCE) is een krachtig hulpmiddel voor het bepalen van gen-functie en het uitvoeren van gerichte genetische manipulatie. De transparantie van de albino kikkervisje en de toegankelijkheid van de hersenen maken dit model systeem bij uitstek geschikt voor het visualiseren van neuronale groei en de intracellulaire gebeurtenissen binnen een levend organisme. Single-cell elektroporatie (SCE) is een gespecialiseerde techniek waardoor de levering van DNA of andere macromoleculen in individuele cellen in intact weefsel, ook in vivo preparaten. Het duidelijk voordeel van deze techniek is dat de experimentele manipulaties kunnen worden uitgevoerd op individuele cellen terwijl het omliggende weefsel ongewijzigd, waardoor onderscheid cel-autonome effecten van die welke voortvloeien uit de wereldwijde behandelingen. In combinatie met geavanceerde in vivo imaging technieken, SCE van TL-markers maakt directe visualisatie van de cellulaire morfologie, de celgroei, en intracellulaire gebeurtenissen van tijdschalen variërend van seconden tot dagen. Hoewel deze techniek wordt gebruikt in een verscheidenheid van in vivo en ex vivo de voorbereidingen, hebben we deze techniek geoptimaliseerd voor gebruik in Xenopus laevis kikkervisjes. In deze video artikel hebben we detail de procedure voor SCE van een fluorescente kleurstof of plasmide DNA in neuronen in de intacte hersenen van de albino Xenopus kikkervisje. We bespreken ook methoden om rendement te optimaliseren, en tonen voorbeelden van levende twee-foton fluorescentie beeldvorming van neuronen fluorescent gelabeld door SCE.
