interview: hiv-1 proviral dna excision using an evolved recombinase

HIV-1 integreras i den mottagande kromosom av infekterade celler och kvarstår som en provirussekvenser flankeras av lång terminal upprepas. Aktuella behandlingsstrategier riktar främst virus enzymer eller virus-cellfusion, undertrycka virusets livscykel utan att utrota smittan. Eftersom den integrerade provirussekvenser inte är måltavla för dessa metoder, nya resistenta stammar av HIV-1 kan uppkomma. Här rapporterar vi att de tekniska recombinase Tre (se Molekylär evolution av Tre recombinase, Buchholz, F., Max Planck-institutet för cellbiologi och genetik, Dresden) effektivt punktskatter integrerat HIV-1 proviral DNA från genomet av infekterade celler. Vi producerade loxLTR innehåller virus pseudotypes och infekterade celler HeLa att undersöka om Tre recombinase kan punktskatt på provirussekvenser från genomet hos HIV-1 infekterade mänskliga celler. Ett virus partikel-releasing cellinje var klonade och transfererats med en plasmid som uttrycker Tre eller med en föräldrakontroll vektor. Recombinase aktivitet och virus produktionen övervakades. Alla analyser har visat en effektiv strykning av provirussekvenser från infekterade celler utan synliga cytotoxiska effekter. Dessa resultat utgör ett bevis på principen att det är möjligt att utveckla en recombinase att särskilt inrikta en HIV-1 LTR och att denna recombinase kan excising HIV-1 provirussekvenser från genomet av HIV-1-infekterade mänskliga celler. Innan en konstruerad recombinase kunde komma in i terapeutiska arenan dock betydande hinder måste övervinnas. Bland de mest kritiska frågorna, att vi står inför, är en effektiv och säker leverans till riktade celler och frånvaro av biverkningar.
