using laser tweezers for manipulating isolated neurons in vitro

Neste artigo e vídeo, descrevemos os protocolos utilizados em nosso laboratório para estudar as preferências de segmentação dos processos celulares regeneração de neurônios da retina adulta in vitro. Procedimentos para a preparação de culturas de células da retina começam com a dissecção digestão, e trituração da retina, e terminam com o chapeamento de isoladas as células da retina em pratos feitos especialmente para uso com laser pinças. Estes pratos são divididos em meia célula adesiva e meia repelente celular. O lado adesivo célula é revestida com uma camada de Sal-1 anticorpos, que fornecem um substrato sobre o qual nossas células crescer. Outros substratos adesivo pode ser usado para outros tipos celulares. O lado repelente célula é revestida com uma fina camada de poly-HEMA.
