We maken gebruik van Sprague Dawley ratten voor onze culturen. Neem de tijd en zorg bij het schoonmaken van de ganglia. Verwijder alle vuil, bloedvaten en vet. Deze stap is belangrijk te zorgen voor een cultuur die is min of meer homogeen en vrij van vreemde celtypen. De toevoeging van uridine/5-FDU zal grotendeels te elimineren alle resterende niet-neuronale (mitotische) cellen besmetten van de cultuur of in ieder geval onderdrukken hun proliferatie. De superieure cervicale ganglia (SCG) bij ratten zijn kleine, glanzende, amandel-vormige structuren die sympathische neuronen bevatten. Deze neuronen zorgen sympathiek innervaties voor het hoofd en de nek regio en vormen zij een goed gekarakteriseerd en relatief homogene populatie (4). Sympathische neuronen zijn afhankelijk van zenuw groeifactor (NGF) om te overleven, differentiatie en axonale groei en de wijdverbreide beschikbaarheid van de NGF vergemakkelijkt hun cultuur en experimentele manipulatie (2, 3, 6). Om deze redenen hebben gekweekte sympathische neuronen is gebruikt in een breed scala van studies met inbegrip van neuronale ontwikkeling en differentiatie, mechanismen van geprogrammeerde celdood en pathologische, en signaaltransductie (1, 2, 5 en 6). Ontleden van de SCG van pasgeboren ratten en het kweken van sympathische neuronen is niet erg ingewikkeld en kan vrij snel onder de knie. In dit artikel zullen we in detail beschrijven hoe te ontleden het SCG van pasgeboren ratten pups en om ze te gebruiken om culturen van sympathische neuronen vast te stellen. Het artikel zal beschrijven ook de voorbereidende stappen en de verschillende reagentia en apparatuur die nodig zijn om dit te bereiken.
