Os gânglios cervical superior (GCS) de ratos são pequenos, brilhantes, em forma de amêndoa estruturas que contêm neurônios simpáticos. Esses neurônios fornecem inervação simpática para a cabeça e pescoço e constituem uma população bem caracterizados e relativamente homogêneo (4). Neurônios simpáticos são dependentes de fator de crescimento neural (NGF) para a diferenciação, sobrevivência e crescimento axonal ea propagação de grande disponibilidade de NGF facilita sua cultura e manipulação experimental (2, 3, 6). Por estas razões, cultivadas neurônios simpáticos têm sido utilizadas em uma ampla variedade de estudos, incluindo o desenvolvimento neuronal e diferenciação, os mecanismos de morte celular programada e patológicos, e transdução de sinal (1, 2, 5 e 6). Dissecando o SCG de ratos recém-nascidos e neurônios simpáticos cultura não é muito complicado e pode ser dominada com bastante rapidez. Neste artigo, iremos descrever em detalhes como dissecar o SCG de filhotes de rato recém-nascido e para usá-los para estabelecer culturas de neurônios simpáticos. O artigo também descreve as etapas preparatórias e os vários reagentes e equipamentos que são necessários para alcançar este objectivo.
