Den cerebellär cortex är en väl beskriven struktur som ger unika möjligheter att studera neuronala egenskaper och utveckling 1,2. Av de cerebellär neuronala typer (granulat celler, Purkinje celler och inhibitoriska interneuron), granulat nervceller är den i särklass mest talrika och är den vanligaste typen av nervceller i däggdjur hjärnan. Hos gnagare är cerebellär granulat nervceller genereras under de två första postnatala veckor från stamceller i det yttersta lagret av cerebellar hjärnbarken, den externa granulat lagret (EGL). Protokollet presenteras här beskriver tekniker för att berika och kultur nervceller granulat och progenitorceller från postnatal musen lillhjärnan. Vi kommer att beskriva procedurer för att få olika kulturer att öka renhet 3,4, som kan användas för att studera differentiering av förökar stamceller till granulat neuroner 5,6. När progenitorceller differentiera, kulturer också ge en homogen population av granulat nervceller för experimentell manipulation och karakterisering av fenomen som synaptogenesis, glutamat receptor funktion 7, samverkan med andra renade cerebellär celler 8,9 eller celldöd 7.
