The moss Physcomitrella patens is an attractive model system for plant biology and functional genome analysis. It shares many biological features with higher plants but has the unique advantage of an efficient homologous recombination system for its nuclear DNA. This allows precise genetic manipulations and targeted knockouts to study gene function, an approach that due to the very low frequency of targeted recombination events is not routinely possible in any higher plant.

Procedura di estrazione di proteine e il contenuto di agente riducente (DTT, Ditioeritritolo, tributyl fosfina e tris (2-carbossietil) fosfina (TCEP)) dei buffer sample e reidratazione sono stati ottimizzati per foglie di faggio europeo e le radici e gli aghi di abete rosso. Estrazione ottimale &egrave; stato ottenuto con 100 mM DTT per foglie e aghi e una miscela di 2 TCEP e 50 mM DTT per radici. Esecuzione IEF nei buffer contenente hydroxyethyldisulphide notevolmente migliorato la qualit&agrave; di separazione per tutte le proteine tranne le proteine acide radice, che ottimamente si concentravano nello stesso buffer come estratti.

Optimal extraction was achieved with 100 mM DTT for leaves and needles and a mixture of 2 mM TCEP and 50 mM DTT for roots. Performing IEF in buffers containing hydroxyethyldisulphide significantly enhanced the quality of separation for all proteins except for acidic root proteins, which were optimally focused in the same buffer as extracted.

Protein extraction from plant samples is usually challenging due to the low protein content and high level of contaminants. Therefore, the 2-DE pattern resolution is strongly influenced by the procedure of sample preparation. Efficient solubilization of proteins strictly depends on the chaotrope and detergent in the extraction buffer. Despite the large number of detergents that have been developed for the use in protein extraction and IEF, there is no single compound able to efficiently extract proteins from any source. Hence, optimization has to be performed for each type of sample. We tested several chaotrope/detergent combinations to achieve optimal solubilization and separation of proteins from Norway spruce [Picea abies (L.) H. Karst.] needles and European beech (Fagus sylvatica L.) leaves and roots. The same chaotrope mixture (7 M urea, 2 M thiourea) was found to be suitable for the extraction and separation of proteins from all samples. Nonetheless, the efficiency of the surfactants tested varied between samples so that optimal extraction and separation was achieved with different detergents or combination of detergents for each sample. The 2-DE separation of spruce needle proteins was optimal in a mixture of two zwitterionic detergents (2% CHAPS and 2% decyl dimethylammonio propanesulfonate). Beech proteins were best separated in buffers containing sugar-based detergents (2% n-octyl beta-D-glucopiranoside in the case of leaf samples and 2% dodecyl maltoside for the root samples). IEF was performed in buffers with the same composition as the extraction buffer except for the root proteins that were better focused in a buffer containing 2% CHAPS.

Malgr&eacute; le grand nombre des d&eacute;tergents qui ont &eacute;t&eacute; d&eacute;velopp&eacute;es pour l&#39;extraction des prot&eacute;ines et IEF, il n&#39;y a aucun compos&eacute; unique capable d&#39;extraire efficacement les prot&eacute;ines provenant d&#39;une source. Par cons&eacute;quent, optimisation doit &ecirc;tre ex&eacute;cut&eacute; pour chaque type d&#39;&eacute;chantillon. Nous avons test&eacute; plusieurs combinaisons spatiale/d&eacute;tergent pour atteindre la solubilisation optimale et la s&eacute;paration des prot&eacute;ines de l&#39;&eacute;pinette de Norv&egrave;ge [Picea abies (L.) Le m&ecirc;me m&eacute;lange de spatiale (ur&eacute;e 7 M, 2 M thiour&eacute;e) s&#39;est av&eacute;r&eacute; appropri&eacute; pour l&#39;extraction et la s&eacute;paration des prot&eacute;ines de tous les &eacute;chantillons. N&eacute;anmoins, l&#39;efficacit&eacute; des surfactants test&eacute;s vari&eacute; entre les &eacute;chantillons afin que la s&eacute;paration et l&#39;extraction optimale a &eacute;t&eacute; r&eacute;alis&eacute;e avec diff&eacute;rents d&eacute;tergents ou combinaison de d&eacute;tergents pour chaque &eacute;chantillon. 2-DE s&eacute;paration des prot&eacute;ines d&#39;aiguilles d&#39;&eacute;pinette est optimale dans un m&eacute;lange de deux d&eacute;tergents zwitterioniques (CHAPS 2 % et 2 % decyl dim&eacute;thylammonio propanesulfonate). Les prot&eacute;ines de h&ecirc;tre ont &eacute;t&eacute; mieux s&eacute;par&eacute;es dans des tampons contenant des d&eacute;tergents &agrave; base de sucre (2 % n-octyle b&ecirc;ta-D-glucopiranoside dans le cas de la feuille le maltoside dod&eacute;cyl &eacute;chantillons et 2 % pour les &eacute;chantillons de racines). IEF a &eacute;t&eacute; r&eacute;alis&eacute;e dans des tampons avec la m&ecirc;me composition que le tampon d&#39;extraction, &agrave; l&#39;exception des prot&eacute;ines de racine qui portaient mieux dans un tampon contenant 2 % CHAPS.

Protein-Extraktion aus Pflanzen Proben ist in der Regel schwierig aufgrund des niedrigen Eiwei&szlig;gehalts und high-Level von Verunreinigungen. Daher ist die 2-DE-Muster-Aufl&ouml;sung durch das Verfahren der Probenvorbereitung stark beeinflusst. Effiziente Solubilization Proteine h&auml;ngt streng Chaotrope und Reinigungsmittel in der Extraktion-Puffer. Trotz der gro&szlig;en Anzahl von Waschmitteln, die f&uuml;r den Einsatz in Protein-Extraktion und IEF entwickelt wurden, gibt es keine einzige Verbindung effizient Proteine aus jeder Quelle zu extrahieren. Daher hat die Optimierung f&uuml;r jede Art der Probe durchgef&uuml;hrt werden. Wir testeten mehrere Chaotrope/Waschmittel-Kombinationen um optimale Solubilization und Trennung der Proteine von Fichte zu erreichen [Picea Abies (L.) H. Karst.] Nadeln und Rotbuche (Fagus Sylvatica L.) Bl&auml;tter und Wurzeln. Die gleiche Chaotrope-Mischung (7 M Harnstoff, 2M Thioharnstoff) wurde festgestellt, dass f&uuml;r die Extraktion und Trennung der Proteine von allen Proben geeignet. Die Effizienz der Tenside getestet dennoch abwechslungsreiche zwischen Proben, damit optimale Extraktion und Trennung mit verschiedenen Waschmitteln oder Kombination von Reinigungsmitteln f&uuml;r jede Probe erreicht wurde. Die 2-DE-Trennung von Fichtennadel Proteine war optimal in einer Mischung aus zwei zwitterionischen Waschmittel (2 % CHAPS und 2 % Decyl Dimethylammonio Propanesulfonate). Buche-Proteine wurden am besten in Puffern mit Zucker basierenden Reinigungsmittel (2 % n-Octyl Beta-D-Glucopiranoside bei Blatt Proben und 2 % Natriumdodecylsulfat-Maltoside f&uuml;r die Stamm-Proben) getrennt. IEF erfolgte in Puffern mit der gleichen Zusammensetzung wie der Extraktion-Puffer mit Ausnahme der Wurzel-Proteine, die besser in einen Puffer mit 2 % CHAPS ausgerichtet waren.

Karst.] aghi e faggio europeo (Fagus sylvatica L.) foglie e radici. La stessa miscela di chaotrope (7 M urea, tiourea M 2) &egrave; stata trovata per essere adatti per l&#39;estrazione e la separazione delle proteine da tutti i campioni. Tuttavia, l&#39;efficienza dei tensioattivi testato varia tra i campioni che separazione ed estrazione ottimale &egrave; stato raggiunto con diversi detergenti o combinazione di detergenti per ogni campione. La 2-DE separazione delle proteine di aghi di abete rosso era ottima in una miscela di due zwitterionici detergenti (screpolature del 2% e 2% decyl dimethylammonio propanesulfonate). Faggio proteine migliori erano separate nei buffer contenente detergenti a base di zucchero (2% n-ottile beta-D-glucopiranoside nel caso di foglia campioni e 2% dodecil maltoside per i campioni di radice). IEF &egrave; stata eseguita nel buffer con la stessa composizione del buffer di estrazione fatta eccezione per le proteine di radice che meglio si sono concentrate in un buffer contenente il 2% CHAPS.

Extracci&oacute;n de prote&iacute;nas de muestras de la planta es generalmente dif&iacute;cil debido al contenido de prote&iacute;na bajo y alto nivel de contaminantes. Por lo tanto, la resoluci&oacute;n de 2 DE patr&oacute;n est&aacute; fuertemente influenciada por el procedimiento de preparaci&oacute;n de la muestra. Eficiente solubilizaci&oacute;n de prote&iacute;nas depende estrictamente el chaotrope y el detergente en el buffer de extracci&oacute;n. The model bryophyte Physcomitrella patens exhibits high frequencies of gene targeting when transformed with DNA constructs containing sequences homologous with genomic loci. 'Targeted gene replacement' (TGR) resulting from homologous recombination (HR) between each end of a targeting construct and the targeted locus occurs when either single or multiple targeting vectors are delivered. In the latter instance simultaneous, multiple, independent integration of different transgenes occurs at the targeted loci. In both single gene and 'batch' transformations, DNA can also be found to undergo 'targeted insertion' (TI), integrating at one end of the targeted locus by HR with one flanking sequence of the vector accompanied by an apparent non-homologous end-joining (NHEJ) event at the other. Untargeted integration at nonhomologous sites also occurs, but at a lower frequency. Molecular analysis of TI at a single locus shows that this occurs as a consequence of concatenation of the transforming DNA, in planta, prior to integration, followed by HR between a single site in the genomic target and two of its repeated homologues in the concatenated vector. This reinforces the view that HR is the major pathway by which transforming DNA is integrated in Physcomitrella.

In species with high gene flow and consequent low interpopulation differentiation over wide geographic ranges, differential gene expression along ecological gradients often reveals adaptive significance. We investigated potential differences in protein expression between Picea abies ecotypes adapted to contrasting altitude conditions. Protein expression patterns were compared between needles and roots of 2-month-old P. abies seedlings by means of 2-dimensional electrophoresis. Proteins exhibiting differential expression between the 2 ecotypes were analyzed by tandem mass spectrometry. A total of 19 proteins exhibited qualitative or quantitative polymorphism between the 2 populations. These proteins exhibited organ-specific expression, and the level of interpopulation protein polymorphism was organ dependent. Among differentially expressed proteins, we identified proteins involved in photosynthesis, photorespiration, root tracheary element differentiation, and transmitochondrial membrane transport. Our results show that P. abies seedlings from locally adapted ecotypes exhibit consistent differences in protein expression. The expression polymorphism of some of these proteins has potential adaptive significance.

Differentiell exprimierten Proteinen erkennbar wir Proteine Photosynthese, Photorespiration, Stamm Tracheary Elements Differenzierung und Transmitochondrial Membrane Transport beteiligt. Unsere Ergebnisse zeigen, dass p. Abies S&auml;mlinge aus lokal angepasste &Ouml;kotypen eindeutigen Unterschiede im Proteinausdruck aufweisen. Der Ausdruck-Polymorphismus einiger dieser Proteine hat potenzielle adaptive Bedeutung.

In specie con elevato flusso genico e conseguente bassa differenziazione evidenzia negli intervalli ampi geografici, espressione genica differenziale lungo gradienti ecologici spesso rivela il significato adattivo. Abbiamo studiato le differenze potenziali nell&#39;espressione della proteina tra Picea abies ecotipi adattati alle condizioni contrastanti di altitudine. I modelli di espressione della proteina sono stati confrontati tra gli aghi e le radici delle piantine di abies p. 2-mese-vecchio mediante elettroforesi bidimensionale. Proteine espositrici espressione differenziale tra le 2 ecotipi sono state analizzate dalla spettrometria di massa tandem. Un totale di 19 proteine esposte polimorfismo qualitativa o quantitativa tra le 2 popolazioni. Although tree species typically exhibit low genetic differentiation between populations, ecotypes adapted to different environmental conditions can vary in their capacity to withstand and recover from environmental stresses like heat stress. Two month old seedlings of a Picea abies ecotype adapted to high elevation showed lower level of thermotolerance and higher level of tolerance to oxidative stress relative to a low elevation ecotype. Protein expression patterns following exposure to severe heat stress of the two ecotypes were compared by means of 2-DE. Several proteins exhibiting ecotype and tissue specific expression were identified by MS/MS. Among them, small heat shock proteins of the HSP 20 family and proteins involved in protection from oxidative stress displayed qualitative and quantitative differences in expression between the ecotypes correlated with the observed phenotypic differences. On the basis of these results, it can be speculated that the observed interpopulation polymorphism of protein regulation in response to heat stress could underlie their different capacities to withstand and recover from heat stress. These local adaptations are potentially relevant for the species adaptation to the conditions predicted by the current models for climate change.

Anche se specie arboree tipicamente mostrano scarsa differenziazione genetica tra popolazioni, ecotipi adattati alle diverse condizioni ambientali possono variare nella loro capacit&agrave; di resistere e recuperare da stress ambientali come stress da calore. Due mesi di et&agrave; piantine di un Picea abies ecotipo adattato ad alta quota ha mostrato pi&ugrave; basso livello di thermotolerance e pi&ugrave; alto livello di tolleranza agli stress ossidativo rispetto un ecotipo di bassa elevazione. I modelli di espressione della proteina dopo esposizione a stress termico grave i due ecotipi sono stati confrontati mediante 2-DE. Di MS/MS sono state identificate diverse proteine espositrici ecotipo e tessuto specifica espressione. Fra loro, proteine di scossa di calore piccola della famiglia HSP 20 e proteine coinvolte nella protezione dallo stress ossidativo visualizzato differenze qualitative e quantitative di espressione tra gli ecotipi correlato con le differenze fenotipiche osservate. Plantones de dos meses de una Picea abies ecotipo adaptado a elevaciones altas mostraron menor grado de thermotolerance y alto nivel de tolerancia al estr&eacute;s oxidativo en relaci&oacute;n con un ecotipo de baja elevaci&oacute;n. Patrones de expresi&oacute;n de la prote&iacute;na despu&eacute;s de la exposici&oacute;n al estr&eacute;s de calor severo de los dos ecotipos se compararon por medio DE-2. Se identificaron varias prote&iacute;nas que exhibe la expresi&oacute;n espec&iacute;fica de tejido y ecotipo por MS/MS. Entre ellos, muestran diferencias cualitativas y cuantitativas en la expresi&oacute;n entre los ecotipos correlacionada con las diferencias fenot&iacute;picas observadas prote&iacute;nas de choque de calor peque&ntilde;o de la familia de HSP 20 y prote&iacute;nas implicadas en la protecci&oacute;n del estr&eacute;s oxidativo. En base a estos resultados, se puede especular que el polimorfismo observado interpoblacional de regulaci&oacute;n de la prote&iacute;na en respuesta a estr&eacute;s cal&oacute;rico podr&iacute;a subyacen en sus diferentes capacidades para resistir y recuperarse del estr&eacute;s por calor. Phytophthora species are major plant pathogens infecting herbaceous and woody plants including European beech, the dominant or co-dominant tree in temperate Europe and an economically important species. For the analysis of the interaction of Phytophthora citricola with Fagus sylvatica suppression subtractive hybridization was used to isolate transcripts induced during infection and 1,149 sequences were generated. Hybridizations with driver and tester populations demonstrated differential expression in infected roots as compared to controls and verify efficient enrichment of these cDNAs during subtraction. Up regulation of selected genes during pathogenesis demonstrated using RT-PCR is consistent with these results. Pathogenesis-related proteins formed the largest group among functionally categorized transcripts. Cell wall proteins and protein kinases were also frequently found. Several transcription factors were isolated that are reactive to pathogens or wounding in other plants. The library contained a number of jasmonic acid, salicylic acid and ethylene responsive genes as well as genes directly involved in signaling pathways. Besides a mechanistic interconnection among signaling pathways another factor explaining the activation of different pathways could be the hemibiotrophic life style of Phytophthora triggering different signals in both stages.

Defense responses of Fagus sylvatica seedlings elicited by infection with the root pathogen Phytophthora citricola and root or leaf wounding were compared at local and systemic levels in differential display experiments using two-dimensional gel electrophoresis followed by homology-driven mass spectrometric identification of proteins. A total of 68 protein spots were identified representing 51 protein functions related to protein synthesis and processing, energy, primary and secondary metabolism, as well as signal transduction, stress and defense. Changes in the abundance of root and leaf proteins partly overlapped between plant responses to the different stressors. The response to pathogen infection was rather late, weak and unspecific and accompanied by adjustments of the energy and primary metabolism which suggested either a lack of recognition or a suppression of host's defense reaction by the invading pathogen. The response to wounding involved changes in the basal metabolism as well as activation of defense mechanisms. Both types of changes were largely specific to the wounded organ. Similarities between the defense mechanisms activated by root infection and root wounding were also observed.

Respuestas de defensa de Fagus sylvatica pl&aacute;ntulas provocada por infecci&oacute;n con la ra&iacute;z pat&oacute;geno Phytophthora citricola y ra&iacute;z o herir la hoja se compararon a nivel local y sist&eacute;mica en experimentos de despliegue diferencial mediante electroforesis bidimensional seguida por la identificaci&oacute;n de espectrometr&iacute;a de masa basada en la homolog&iacute;a de las prote&iacute;nas. Se identificaron un total de 68 puntos de prote&iacute;na que representa 51 funciones de prote&iacute;nas relacionadas con la s&iacute;ntesis de prote&iacute;nas y procesamiento, energ&iacute;a, metabolismo primario y secundario, as&iacute; como la transducci&oacute;n de la se&ntilde;al, el estr&eacute;s y la defensa. Cambios en la abundancia de prote&iacute;nas de ra&iacute;z y hoja parcialmente solaparon entre las respuestas de la planta a los diferentes factores de estr&eacute;s. La respuesta a la infecci&oacute;n del pat&oacute;geno fue bastante tarde, d&eacute;bil e inespec&iacute;ficas y acompa&ntilde;ada de ajustes de la energ&iacute;a y el metabolismo primario que sugiere una falta de reconocimiento o una supresi&oacute;n de la reacci&oacute;n de defensa del hu&eacute;sped por el pat&oacute;geno invasor. La respuesta a herida cambios implicados en el metabolismo basal, as&iacute; como la activaci&oacute;n de mecanismos de defensa. Ambos tipos de cambios fueron en gran parte espec&iacute;ficas al &oacute;rgano herido. An&aacute;lisis prote&oacute;mico comparativo de las respuestas a la infecci&oacute;n del pat&oacute;geno y herir en Fagus sylvatica.

Phytophthora citricola is a wide spread and highly aggressive pathogen of Fagus sylvatica. The hemibiotrophic oomycete infects the roots and establishes a compatible interaction with F. sylvatica. To investigate the transcriptional changes associated with P. citricola infection, 68 custom oligo-microarray measurements were conducted. Hierarchical as well as non-hierarchical clustering was carried out to analyze the expression profiles. Experimental setup includes a time scale covering the biotrophic and necrotrophic stages of interaction as well as comparative analyses of the local and systemic responses. The local reaction of F. sylvatica is characterized by a striking lack of defense gene induction leading to the conclusion that P. citricola escapes the main recognition systems and/or suppresses the host's response. The analysis of the systemic reaction revealed a massive shift in gene expression patterns during the biotrophic phase that is interpreted as evidence of resource allocation into the roots to support the increased sink caused by pathogen growth. Defense genes known to be responsive to salicylic acid (effective against biotrophs), jasmonic acid, and ethylene (effective against necrotrophs and herbivores) are represented on the arrays. All significant changes in gene expression measured for salicylic acid responsive genes were down-regulations in roots and leaves while some jasmonic acid responsive genes showed a very late up-regulation only in leaves, probably caused by the desiccation shortly before plant death. Together, these expression changes could explain the success of the pathogen.

Down-regulation of defense genes and resource allocation into infected roots as factors for compatibility between Fagus sylvatica and Phytophthora citricola.

In 2006, a controlled infection study was performed in the 'Kranzberger Forst' to address the following questions: (1) Will massive artificial inoculation with Apiognomonia errabunda override the previously observed inhibitory effect of chronic ozone? (2) Can biochemical or molecular markers be detected to account for the action of ozone? To this end six adult beech trees were chosen, three ozone fumigated (2x ozone) and three control trees (ambient = 1x ozone). Spore-sprayed branches of sun and shade crown positions of each of the trees, and uninoculated control branches, were enclosed in 100-L plastic bags for one night to facilitate infection initiation. Samples were taken within a five-week period after inoculation. A. errabunda infestation levels quantified by real-time PCR increased in leaves that were not fumigated with additional ozone. Cell wall components and ACC (ethylene precursor 1-amino cyclopropane-1-carboxylic acid) increased upon ozone fumigation and may in part lead to the repression of fungal infection.

Ozone fumigation (twice ambient) reduces leaf infestation following natural and artificial inoculation by the endophytic fungus Apiognomonia errabunda of adult European beech trees.

The psyllid species Cacopsylla melanoneura (Förster) and Cacopsylla picta (Förster) are vectors of 'Candidatus Phytoplasma mali', the causal agent of apple proliferation, one of the economically most important apple diseases in Europe. Both vectors are present in apple orchards of South Tyrol and Trentino provinces in Northern Italy. As no direct treatment of the disease is possible, monitoring of the psyllids provides information about the vector presence in the orchards and enables targeted control. Thus, fast and reliable identification of the various psyllids occurring in the apple orchards is required. Morphological differentiation is problematic due to extensive resemblance of some psyllid species especially among females and is error-prone for nymphs. Here we present a rapid and cost-effective polymerase chain reaction-restriction fragment length polymorphism method based on the cytochrome c oxidase subunit I region for the molecular identification of the vector species as well as eight further Cacopsylla species present in the orchards. This method was verified through 98.9% consensus with morphologically identified males, through sequencing and subsequent phylogenetic analysis. In case of doubtful morphological identification of females, the method was able to provide a refined species assignment and could also remarkably facilitate the identification of nymphs.

Molecular Identification of Two Vector Species, Cacopsylla melanoneura and Cacopsylla picta (Hemiptera: Psyllidae), of Apple Proliferation Disease and Further Common Psyllids of Northern Italy.

The plant pathogen Candidatus Phytoplasma mali (P. mali) is the causative agent of apple proliferation, a disease of increasing importance in apple growing areas within Europe. Despite its economic importance little is known about the molecular mechanisms of disease manifestation within apple trees. In this study we identified two TCP transcription factors of Malus x domestica as binding partners of the P. mali SAP11-like effector ATP_00189. Phytohormone analyses revealed an effect of P. mali infection on jasmonates, salicylic acid, and abscisic acid levels showing that P. mali affects phytohormonal levels in apple trees, which is in line with the functions of the effector assumed from its binding to TCP transcription factors. To our knowledge, this is the first characterization of the molecular targets of a P. mali effector and thus provides the basis to better understand symptom development and disease progress during apple proliferation. Since SAP11 homologues are found in several phytoplasma species infecting a broad range of different plants, SAP11-like proteins seem to be key players in phytoplasmal infection.
