Many bacteria spread over surfaces by "swarming" in groups. A problem for scientists who study swarming is the acquisition of statistically significant data that distinguish two observations or detail the temporal patterns and two-dimensional heterogeneities that occur. It is currently difficult to quantify differences between observed swarm phenotypes. Here, we present a method for acquisition of temporal surface motility data using time-lapse fluorescence and bioluminescence imaging. We specifically demonstrate three applications of our technique with the bacterium Pseudomonas aeruginosa. First, we quantify the temporal distribution of P. aeruginosa cells tagged with green fluorescent protein (GFP) and the surfactant rhamnolipid stained with the lipid dye Nile red. Second, we distinguish swarming of P. aeruginosa and Salmonella enterica serovar Typhimurium in a coswarming experiment. Lastly, we quantify differences in swarming and rhamnolipid production of several P. aeruginosa strains. While the best swarming strains produced the most rhamnolipid on surfaces, planktonic culture rhamnolipid production did not correlate with surface growth rhamnolipid production.

Muchas bacterias distribuidas en las superficies por el &#x26;quot;enjambre&#x26;quot; en grupos. Un problema para los cient&#xED;ficos que estudian el enjambre es la adquisici&#xF3;n de datos estad&#xED;sticamente significativos que distinguen a dos observaciones o detalles de los patrones temporales y de dos dimensiones heterogeneidades que se producen. Actualmente es dif&#xED;cil de cuantificar las diferencias entre los fenotipos observados enjambre. A continuaci&#xF3;n, presentamos un m&#xE9;todo para la adquisici&#xF3;n de los datos temporales de la superficie con la motilidad de lapso de tiempo de fluorescencia y las im&#xE1;genes de bioluminiscencia. En especial, demuestran tres aplicaciones de nuestra t&#xE9;cnica con la bacteria Pseudomonas aeruginosa. En primer lugar, cuantificar la distribuci&#xF3;n temporal de las c&#xE9;lulas de P. aeruginosa etiquetados con la prote&#xED;na verde fluorescente (GFP) y el manchado de ramnol&#xED;pido tensioactivo con el tinte de los l&#xED;pidos del Nilo rojo. En segundo lugar, podemos distinguir un enjambre de P. aeruginosa y Salmonella enterica serovar Typhimurium en un experimento coswarming. Por &#xFA;ltimo, cuantificar las diferencias en enjambre y de la producci&#xF3;n ramnol&#xED;pido de varias cepas de P. aeruginosa. Mientras que las mejores cepas produjo el enjambre ramnol&#xED;pido la mayor&#xED;a de las superficies, la cultura de producci&#xF3;n planct&#xF3;nica ramnol&#xED;pido no se correlacion&#xF3; con la producci&#xF3;n de ramnol&#xED;pido superficie de crecimiento.

Over the past 20 years, multimodal imaging strategies have motivated the fusion of Positron Emission Tomography (PET) or Single Photon Emission Computed Tomography (SPECT) scans with an X-ray computed tomography (CT) image to provide anatomical information, as well as a framework with which molecular and functional images may be co-registered. Recently, pre-clinical nuclear imaging technology has evolved to capture multiple SPECT or multiple PET tracers to further enhance the information content gathered within an imaging experiment. However, the use of SPECT and PET probes together, in the same animal, has remained a challenge. Here we describe a straightforward method using an integrated trimodal imaging system and a sequential dosing/acquisition protocol to achieve dual tracer imaging with (99m)Tc and (18)F isotopes, along with anatomical CT, on an individual specimen. Dosing and imaging is completed so that minimal animal manipulations are required, full trimodal fusion is conserved, and tracer crosstalk including down-scatter of the PET tracer in SPECT mode is avoided. This technique will enhance the ability of preclinical researchers to detect multiple disease targets and perform functional, molecular, and anatomical imaging on individual specimens to increase the information content gathered within longitudinal in vivo studies.

En los &uacute;ltimos 20 a&ntilde;os, estrategias de imagen multimodales han motivado la fusi&oacute;n de tomograf&iacute;a de emisi&oacute;n de positrones (PET) o Single Photon emisi&oacute;n computarizada tomograf&iacute;a (SPECT) explora con una imagen de la tomograf&iacute;a computada (CT) de rayos x para proporcionar informaci&oacute;n anat&oacute;mica, as&iacute; como un marco con el cual im&aacute;genes moleculares y funcionales pueden ser colocadas. Recientemente, precl&iacute;nica tecnolog&iacute;a nuclear ha evolucionado para capturar m&uacute;ltiples SPECT o varios trazadores de PET para mejorar el contenido de la informaci&oacute;n reunieron en un experimento de la proyecci&oacute;n de imagen. Sin embargo, el uso de sondas de SPECT y PET juntos, en el mismo animal, sigue siendo un desaf&iacute;o. Aqu&iacute; describimos un m&eacute;todo sencillo usando un sistema de proyecci&oacute;n de imagen de trimodal integrado y un protocolo de dosificaci&oacute;n/adquisici&oacute;n secuencial para lograr tracer doble imagen con (99m) Tc e is&oacute;topos de F (18), junto con CT anat&oacute;mica, en una muestra individual. Dosificaci&oacute;n y la proyecci&oacute;n de imagen completa para que m&iacute;nimo manipulaciones animales deben trimodal completa fusi&oacute;n se conserva y se evita la interferencia de tracer incluyendo abajo-dispersi&oacute;n del trazador de PET en modo SPECT. Esta t&eacute;cnica mejorar&aacute; la capacidad de los investigadores precl&iacute;nicos para detectar objetivos m&uacute;ltiples de la enfermedad y realizar la proyecci&oacute;n de imagen anat&oacute;mica, funcional y molecular en muestras individuales para aumentar el contenido de la informaci&oacute;n reunido en los estudios in vivo longitudinales.
