The human natural killer gene complex (NKC) encodes for numerous C-type lectin-like receptors (CTLR), which are expressed on various immune cells including natural killer (NK) cells and myeloid cells. Certain activation-induced, NKC-encoded CTLR are grouped into the C-type lectin domain family 2 (CLEC2 family) which, in humans, comprises AICL (CLEC2B), CD69 (CLEC2C), and LLT1 (CLEC2D). In this paper, we characterize a novel member of the CLEC2 family, the human orphan gene CLEC2A. The C-type lectin-like domain (CTLD) of CLEC2A is most similar to the CTLD of LLT1 ( approximately 60% similarity). Like mouse CLEC2 family members Clr-b and Clr-g, CLEC2A lacks two highly conserved cysteines (Cys4 and Cys5), which form an intramolecular bond in the CTLD of most CTLR. Alternative splicing of exon 2 and of two distinct terminal exons (exon 5A/B), respectively, gives rise to four CLEC2A variants differing in the usage of the transmembrane domain and/or in the carboxyterminal portion of the CTLD. CLEC2A transcripts were detected primarily in myeloid cell lines, but not in epithelial cell lines. In tissues, CLEC2A is selectively expressed in the skin and, at lower abundance, in hematopoietic and gonadal tissues. Finally, we show that the CLEC2A1 variant is readily expressed at the cell surface, where it may serve as a ligand for NKC-encoded NK receptors.

CLEC2A: a novel, alternatively spliced and skin-associated member of the NKC-encoded AICL-CD69-LLT1 family.

Natural killer g&egrave;ne humain complex (NKC) encode pour nombreux r&eacute;cepteurs de type lectine type C (CTLR), qui se sont exprim&eacute;s sur les cellules immunitaires diverses, y compris les cellules tueuses naturelles (NK) et cellules my&eacute;lo&iuml;des. Bestimmte Aktivierung induziert, NKC-codierte CTLR gliedern sich in die C-Typ-Lektin-Dom&auml;ne-Familie 2 (CLEC2-Familie), die beim Menschen AICL (CLEC2B), CD69 umfasst (CLEC2C), und LLT1 (CLEC2D). In diesem Papier pr&auml;gen wir neuartige Mitglied der CLEC2 Familie, das menschliche verwaiste gen CLEC2A. Im Gewebe wird CLEC2A in der Haut und am unteren F&uuml;lle, in h&auml;matopoetischen und Gonadale Gewebe selektiv ausgedr&uuml;ckt. Schlie&szlig;lich zeigen wir, dass die CLEC2A1-Variante ohne weiteres an der Zelloberfl&auml;che ausgedr&uuml;ckt wird wo es als ein Ligand f&uuml;r NK NKC-codierte Rezeptoren dienen.

Natural killer gene humano complejo (NKC) codifica para numerosos C-tipo lectina-como receptores (CTLR), que se expresan en diferentes c&eacute;lulas inmunes, incluyendo las c&eacute;lulas asesinas naturales (NK) y c&eacute;lulas mieloides. Cierto CTLR inducida por activaci&oacute;n, NKC-codificado se agrupan en la familia de dominio del lectinas tipo C 2 (familia CLEC2) que, en los seres humanos, se compone de AICL (CLEC2B), CD69 (CLEC2C) y LLT1 (CLEC2D). En este trabajo se caracteriza a un nuevo miembro de la familia de CLEC2, el gen humano hu&eacute;rfano CLEC2A. El dominio de tipo lectina tipo C (CTLD) de CLEC2A es m&aacute;s similar a la CTLD de LLT1 (aproximadamente 60% de similitud). Como miembros de la familia de rat&oacute;n CLEC2 Clr-b y Clr-g, CLEC2A carece de dos ciste&iacute;nas altamente conservados (Cys4 y Cys5), que forman un enlace intramolecular en el CTLD de CTLR mayor&iacute;a. Splicing alternativo del ex&oacute;n 2 y de dos distintos exones terminales (ex&oacute;n 5A/B), respectivamente, da lugar a cuatro variantes de CLEC2A diferentes en el uso del dominio transmembrana o en la porci&oacute;n carboxitrminal de la CTLD. CLEC2A transcripciones fueron detectadas principalmente en las l&iacute;neas celulares mieloide, pero no en l&iacute;neas de c&eacute;lulas epiteliales. En los tejidos, el CLEC2A se expresa selectivamente en la piel y, en menor abundancia, en tejidos hematopoy&eacute;ticos y gonadales. Finalmente, demostramos que la variante CLEC2A1 se expresa f&aacute;cilmente en la superficie celular, donde puede servir como un ligando para receptores NK NKC-codificado.
