The success of helper-dependent adenoviral (HD-Ad) vector mediated lung gene therapy is hampered by the host immune response, which limits pulmonary transgene expression following multiple rounds of vector readminstration. Here we show that HD-Ad mediated pulmonary gene expression is sustained even upon three rounds of readministrations to immunodeficient mice, highlighting the need to suppress adaptive immune response for sustained gene expression following vector readministration. Therefore, we devised a dendritic cell (DC) based strategy for induction of immunological tolerance towards HD-Ad vectors. DCs derived in presence of IL-10 are refractory to HD-Ad induced maturation and instead facilitate generation of IL-10 producing Tr1 regulatory T cells which suppress HD-Ad-induced T cell proliferation. Delivery of HD-Ad pulsed, IL-10 modified DCs to mice induces long lasting immunological tolerance towards HD-Ad vectors, whereby pulmonary DC maturation, T cell response along with antibody response to HD-Ad vectors is suppressed even after three rounds of pulmonary HD-Ad readministrations. Moreover, sustained transgene expression is also observed in the lungs of mice immunized with HD-Ad pulsed IL-10 modified modified DCs even after three rounds of pulmonary HD-Ad delivery. Taken together these studies identify the use of DCs generated in presence of IL-10 as a novel strategy to induce long lasting immune tolerance towards HD-Ad vectors.

Le succ&egrave;s de la th&eacute;rapie g&eacute;nique de d&eacute;pendant d&#39;assistance ad&eacute;noviraux (HD-Ad) vecteur m&eacute;di&eacute;e par poumon est entrav&eacute; par la r&eacute;ponse immunitaire h&ocirc;te, qui limite l&#39;expression TRANSGENIQUE pulmonaire apr&egrave;s plusieurs tours de vecteur readminstration. Nous d&eacute;montrons que HD-Ad, l&#39;expression des g&egrave;nes pulmonaires m&eacute;diatis&eacute;e est maintenue m&ecirc;me apr&egrave;s trois rondes de readministrations &agrave; des souris immunod&eacute;ficientes, soulignant qu&#39;il fallait supprimer la r&eacute;ponse immunitaire adaptative pour subies apr&egrave;s la r&eacute;administration du vecteur l&#39;expression g&eacute;nique. Livraison de HD-publicit&eacute; puls&eacute;s, IL-10 DCs modifi&eacute;s chez la souris induit la tol&eacute;rance immunologique longue dur&eacute;e vers les vecteurs HD-Ad, auquel cas la maturation pulmonaire de DC, la r&eacute;ponse des lymphocytes T ainsi que la r&eacute;ponse anticorps &agrave; des vecteurs de HD-Ad est supprim&eacute;e m&ecirc;me apr&egrave;s trois rondes de pulmonaires readministrations HD-Ad. Der Erfolg der Helfer-abh&auml;ngige adenoviral (HD-Ad) Vektor-vermittelte Lunge Gentherapie wird durch die Host Immunantwort behindert welche Grenzen der pulmonalen Transgenexpression nach mehreren Runden Readminstration Vektor. Hier zeigen wir, dass HD-Fernsehwerbespot vermittelten pulmonale Genexpression sogar nach drei Runden von Readministrations auf immungeschw&auml;chte M&auml;use, aufrecht erhalten wird die Notwendigkeit f&uuml;r adaptive Immunantwort unterdr&uuml;cken Genexpression nach Vektor Readministration aufrecht erhalten. Daher entwickelten wir eine Strategie dendritischen Zellen (DC) auf der Grundlage f&uuml;r die Induktion der immunologischen Toleranz gegen&uuml;ber HD-Ad-Vektoren. DCs abgeleitet in Gegenwart von IL-10 sind feuerfeste HD-Ad-induzierte Reifung und stattdessen erleichtern Generation von IL-10 produzieren Tr1 regulatorischen T-Zellen, die HD-Ad-induzierte T-Zellproliferation zu unterdr&uuml;cken. Lieferung von HD-Ad gepulste, IL-10 modifizierte DCs auf M&auml;use induziert lang anhaltenden immunologische Toleranz gegen&uuml;ber HD-Ad-Vektoren, wobei pulmonale DC Reifung, T-Zell-Antwort zusammen mit Antik&ouml;rper-Reaktion zu HD-Ad Vektoren auch nach drei Runden der pulmonalen HD-Ad Readministrations unterdr&uuml;ckt wird. Dar&uuml;ber hinaus wird auch die anhaltende Transgenexpression beobachtet in den Lungen von M&auml;usen mit HD-Ad geimpft gepulste IL-10 ge&auml;ndert ge&auml;ndert DCs auch nach drei Runden der pulmonalen HD-Ad-Lieferung. Zusammengenommen bestimmen diese Studien die Verwendung von DCs erstellt in Anwesenheit von IL-10 als eine neuartige Strategie zu lang anhaltenden immun-Toleranz gegen&uuml;ber HD-Ad-Vektoren zu induzieren.

Il successo di helper-dependent adenovirale (HD-Ad) vettore mediata del polmone terapia genica &egrave; ostacolato dalla risposta immunitaria, quali limiti di espressione del transgene polmonare seguendo molteplici colpi di vector readminstration. Qui vi mostriamo che HD-Ad espressione genica mediata polmonare &egrave; sostenuta anche su tre turni di readministrations a topi immunodeficienti, evidenziando la necessit&agrave; di sopprimere la risposta immunitaria adattativa per sostenuta espressione genica seguito vettore readministration. Pertanto, abbiamo messo a punto una strategia di cellule dendritiche (DC) basata per l&#39;induzione di tolleranza immunologica nei confronti di vettori HD-Ad. DCs derivato in presenza di IL-10 sono refrattari a maturazione HD-annuncio indotta e invece facilitare la generazione di cellule T regolatorie IL-10 producendo Tr1 che sopprimere la proliferazione delle cellule T HD-annuncio-indotta. Consegna di HD-annuncio pulsato, IL-10 DCs modificato per topi induce duratura tolleranza immunologica verso vettori HD-Ad, per cui maturazione polmonare di DC, risposta delle cellule T con risposta anticorpale a vettori HD-Ad &egrave; soppressa anche dopo tre turni di readministrations HD-annuncio polmonare. Inoltre, l&#39;espressione del transgene sostenuta inoltre &egrave; osservato nei polmoni di topi immunizzati con HD-Ad impulsi IL-10 modificato modificato DCs anche dopo tre turni di consegna HD-annuncio polmonare. Presi insieme questi studi identificano l&#39;uso di DCs generato in presenza di IL-10 come una romanzo strategia per indurre tolleranza immune duraturo verso vettori HD-Ad.

El &eacute;xito de la ayudante dependiente (HD-Ad) adenoviral vector mediada por pulm&oacute;n terapia g&eacute;nica se ve entorpecido por la inmunorespuesta del anfitri&oacute;n, que l&iacute;mites de expresi&oacute;n del transg&eacute;n pulmonar tras m&uacute;ltiples rondas de vector readminstration. Aqu&iacute; mostramos que HD-Ad expresi&oacute;n g&eacute;nica pulmonar mediado se sustenta sobre tres rondas de readministrations a ratones inmunodeficientes, destacando la necesidad de suprimir la respuesta inmune adaptativa para mantenerse siguiendo el readministration del vector de expresi&oacute;n de genes. Por lo tanto, dise&ntilde;&oacute; una estrategia de c&eacute;lulas dendr&iacute;ticas (DC) base para la inducci&oacute;n de tolerancia inmunol&oacute;gica hacia vectores HD-Ad. DCs derivadas en presencia de IL-10 son refractarios a HD-Ad inducida por maduraci&oacute;n y en su lugar facilitan la generaci&oacute;n de producci&oacute;n de IL-10 Tr1 las c&eacute;lulas T reguladoras que inhiben la proliferaci&oacute;n celular inducida por HD-anuncio de T. Entrega de HD-Ad DCs modificados pulsadas, IL-10 ratones induce tolerancia inmunol&oacute;gica duradera hacia vectores HD-Ad, por el que se suprime la maduraci&oacute;n pulmonar de DC, respuesta de los linfocitos T junto con la respuesta de anticuerpos a vectores HD-Ad incluso despu&eacute;s de tres rondas de readministrations pulmonares de HD-Ad. Por otra parte, tambi&eacute;n se observa la expresi&oacute;n del transg&eacute;n sostenida en los pulmones de ratones inmunizados con HD-Ad pulsada IL-10 modificado modificado DCs incluso despu&eacute;s de tres rondas de entrega pulmonar de HD-Ad. En conjunto estos estudios identifican el uso de DCs generadas en presencia de IL-10 como una nueva estrategia para inducir tolerancia inmunol&oacute;gica duradera hacia vectores HD-Ad.
