In 1970's some Scaphium lychnophorum seedlings were introduced from Kampuchea to Xishuangbanna, Yunnan Province. These seedlings have grown into the large trees and bore fruits to propagate successfully in the nature over many years. Some problems of the plant adaptability and phenological reaction to the local environment, as well as the main cultivable measures were summarized in this paper.

C-reactive protein (CRP) is considered as one of the most sensitive markers of inflammation. The aim of the present study is to investigate the effects of CRP on the production of adiponectin in 3T3-L1 adipocytes. Northern and western blot analysis revealed that CRP treatment inhibited adiponectin mRNA expression and secretion in a dose- and time-dependent manner. Co-incubation of adipocytes with rosiglitazone and CRP decreased induction of adiponectin gene expression by rosiglitazone. However, luciferase reporter assays did not show that CRP affected the activity of approximately 2.1 kb adiponectin gene promoter, which was increased by rosiglitazone alone. Pharmacological inhibition of phosphatidylinositol (PI)-3 kinase by LY294002 partially reversed inhibition of adiponectin gene expression by CRP. These results collectively suggest that CRP suppresses adiponectin gene expression partially through the PI-3 kinase pathway, and that decreased production of adiponectin might represent a mechanism by which CRP regulates insulin sensitivity.

La pr&eacute;sente &eacute;tude vise &agrave; &eacute;tudier les effets du PRC sur la production d&#39;adiponectine dans 3 t 3-L1 adipocytes. Buvardage septentrionale et occidentale a r&eacute;v&eacute;l&eacute; que le traitement de CRP inhib&eacute;e adiponectine expression de l&#39;ARNm et la s&eacute;cr&eacute;tion d&#39;une mani&egrave;re dose-d&eacute;pendante et temps. Incubation des adipocytes avec la rosiglitazone et de la CRP a diminu&eacute; l&#39;induction de l&#39;expression g&eacute;nique de l&#39;adiponectine rosiglitazone. Sin embargo, reportero de luciferasa ensayos no mostraron que CRP afect&oacute; a la actividad de aproximadamente 2,1 kb adiponectina promotor del gen, que fue aumentado por rosiglitazona solo. Inhibici&oacute;n farmacol&oacute;gica de fosfatidilinositol (PI) -3 quinasa por LY294002 revoc&oacute; parcialmente la inhibici&oacute;n de la expresi&oacute;n g&eacute;nica de adiponectina por CRP. Very-low-density lipoprotein receptor (VLDLR) is a multi ligand apolipoprotein E (apoE) receptor and is involved in brain development through Reelin signaling. Different forms of VLDLR can be generated by alternative splicing. VLDLR-I contains all exons. VLDLR-II lacks an O-linked sugar domain encoded by exon 16, while VLDLR-III lacks the third complement-type repeat in the ligand binding domain encoded by exon 4. We quantitatively compared lipoprotein binding to human VLDLR variants and analyzed their mRNA expression in both human cerebellum and mouse brain. VLDLR-III exhibited the highest capacity in binding to apoE enriched beta-VLDL in vitro and was more effective in removing apoE containing lipoproteins from the circulation than other variants in vivo. In human cerebellum, the major species was VLDLR-II, while the second most abundant species was a newly identified VLDLR-IV which lacks both exon 4 and 16. VLDLR-I was present at low levels. In adult mice, exon 4 skipping varied between 30 and 47% in different brain regions, while exon 16 skipping ranged by 51-76%. Significantly higher levels of VLDLR proteins were found in mouse cerebellum and cerebral cortex than other regions. The deletions of exon 4 and exon 16 frequently occurred in primary neurons, indicating that newly identified variant VLDLR-IV is abundant in neurons. In contrast, VLDLR mRNA lacking exon 4 was not detectable in primary astrocytes. Such cell type-specific splicing patterns were found in both mouse cerebellum and cerebral cortex. These results suggest that a VLDLR variant lacking the third complement-type repeat is generated by neuron-specific alternative splicing. Such differential splicing may result in different lipid uptake in neurons and astrocytes.

Sehr-Low-Density-Lipoprotein-Rezeptor (VLDLR) ist ein Multi-Ligand Apolipoprotein E (ApoE) Rezeptor und engagiert sich in der Entwicklung des Gehirns durch Reelin signalisieren. Verschiedene Formen der VLDLR k&ouml;nnen durch alternatives Splei&szlig;en generiert werden. VLDLR-I enth&auml;lt alle Exons. VLDLR-II fehlt eine O-chromosomale Zucker-Dom&auml;ne durch Exon 16 kodiert, w&auml;hrend VLDLR-III fehlt die dritte Erg&auml;nzung-Art in der Liganden-bindende Dom&auml;ne codiert von Exon 4 wiederholen. Wir quantitativ Lipoprotein Bindung an menschliche VLDLR Varianten verglichen und analysiert ihre mRNA Ausdruck im menschlichen Kleinhirn und Gehirn der Maus. VLDLR-III stellte die h&ouml;chste Kapazit&auml;t in Bindung an ApoE bereichert Beta-VLDL in-vitro- und war effektiver bei der Beseitigung von ApoE mit Lipoproteinen aus dem Blutkreislauf als andere Varianten, die in-vivo. Im menschlichen Kleinhirn die gro&szlig;e Gattung sei VLDLR-II, w&auml;hrend der Zweitens am h&auml;ufigsten vorkommenden Arten wurde eine neu identifizierte VLDLR-IV die beiden Exon 4 und 16 fehlt. VLDLR-ich war auf einem niedrigen Niveau. Erwachsene M&auml;use variiert in Exon 4 &uuml;berspringen zwischen 30 und 47 % in verschiedenen Hirnregionen, w&auml;hrend Exon 16 &uuml;berspringen von 51-76 % reichten. Diferentes formas de VLDLR pueden ser generadas por splicing alternativo. VLDLR-contiene todos los exones. VLDLR-II carece de un dominio de az&uacute;car vinculados O codificado por el ex&oacute;n 16, mientras que carece de VLDLR-III el tercer tipo de complemento se repita en el dominio de uni&oacute;n a ligando codificado por ex&oacute;n 4. Aims Interleukin-7 (IL-7) is a master regulator of T-cell development and homoeostasis. Increased IL-7 levels are associated with inflammatory diseases. The aims of this study were to determine whether IL-7 is a biomarker for inflammatory conditions or an active participant in atherogenesis. Methods and results Advanced atherosclerotic lesions in Apoe(-/-) mice were regressed by long-term cholesterol lowering through treatment with a helper-dependent adenovirus expressing apolipoprotein E (n= 6-10). Using this model, gene expression patterns in the aorta were analysed at an early phase of regression by microarray. After stringent statistical analysis, we found that IL-7 expression is significantly reduced in response to lowering of cholesterol (n= 6). To understand the importance of IL-7 down-regulation for atherosclerotic regression, we studied the effects and mechanisms of action of IL-7 on endothelial cells (ECs) in vitro as well as in vivo. Our major findings are: (i) IL-7 up-regulates cell adhesion molecules and monocyte chemoattractant protein-1 in ECs and promotes monocyte adhesion to ECs; (ii) this regulation is mediated by phosphatidylinositol 3-kinase (PI3K)/AKT-dependent and -independent activation of NF-ÎºB; (iii) elevation of plasma IL-7 induces recruitment of monocytes/macrophages to endothelium without affecting plasma cholesterol (n= 5, 6); and (4) lack of IL-7 in bone marrow-derived cells reduces migration of monocytes/macrophages to the lesions (n= 5, 6). Conclusion These results suggest that IL-7 inflames endothelium via PI3K/AKT-dependent and -independent activation of NF-ÎºB and recruits monocytes/macrophages to the endothelium, thus playing an active role in atherogenesis.

Objectifs interleukine-7 (IL-7) est un ma&icirc;tre r&eacute;gulateur du d&eacute;veloppement des cellules T et l&#39;hom&eacute;ostasie. Une augmentation des niveaux de IL-7 sont associ&eacute;s &agrave; des maladies inflammatoires. Les objectifs de cette &eacute;tude &eacute;taient de d&eacute;terminer si l&#39;IL-7 est un biomarqueur pour affections inflammatoires ou un participant actif dans l&#39;ath&eacute;rogen&egrave;se. M&eacute;thodes et r&eacute;sultats des l&eacute;sions ath&eacute;roscl&eacute;reuses avanc&eacute;e chez les souris Apoe(-/-) ont r&eacute;gress&eacute; par le cholest&eacute;rol &agrave; long terme gr&acirc;ce &agrave; un traitement avec un ad&eacute;novirus helper-d&eacute;pendante exprimant l&#39;apolipoprot&eacute;ine E (n = 6-10). En utilisant ce mod&egrave;le, des profils d&#39;expression g&eacute;nique dans l&#39;aorte ont &eacute;t&eacute; analys&eacute;s &agrave; une phase pr&eacute;coce de la r&eacute;gression par microarray. Apr&egrave;s une analyse statistique rigoureuse, nous avons constat&eacute; que l&#39;expression d&#39;IL-7 est consid&eacute;rablement r&eacute;duite en r&eacute;ponse &agrave; une baisse des taux de cholest&eacute;rol (n = 6). Pour comprendre l&#39;importance de la r&eacute;gulation &agrave; la baisse IL-7 pour la r&eacute;gression ath&eacute;roscl&eacute;rose, nous avons &eacute;tudi&eacute; les effets et les m&eacute;canismes d&#39;action de l&#39;IL-7 sur les cellules endoth&eacute;liales (CE) in vitro comme in vivo. Nos principales conclusions sont: (i) IL-7 haut-r&eacute;gule les mol&eacute;cules d&#39;adh&eacute;rence cellulaire et monocyte chemoattractant protein-1 en ECs et favorise l&#39;adh&eacute;rence des monocytes &agrave; ECs ; (ii) le pr&eacute;sent r&egrave;glement est m&eacute;di&eacute;e par la phosphatidylinositol 3-kinase (PI3K) / AKT d&eacute;pendants et ind&eacute;pendante activation de NF-&kappa;B ; (iii) l&#39;&eacute;l&eacute;vation du plasma IL-7 induit le recrutement des monocytes/macrophages &agrave; l&#39;endoth&eacute;lium sans affecter le taux de cholest&eacute;rol plasmatique (n = 5, 6) ; et (4) manque d&#39;IL-7 dans les cellules de moelle osseuse r&eacute;duit la migration des monocytes/macrophages aux l&eacute;sions (n = 5, 6). Nach strengen statistischen Auswertung, wir fanden, dass IL-7 Ausdruck als Reaktion auf die Senkung des Cholesterins deutlich reduziert ist (n = 6). Obiettivi interleuchina-7 (IL-7) &egrave; un regolatore master di sviluppo delle cellule T e dell&#39;omeostasi. Aumentati livelli di IL-7 sono associati a malattie infiammatorie. Gli obiettivi di questo studio sono stati per determinare se IL-7 &egrave; un biomarcatore per condizioni infiammatorie o un partecipante attivo nella aterogenesi. Metodi e risultati lesioni aterosclerotiche avanzate nei topi Apoe(-/-) erano regredite da colesterolo, a lungo termine mediante trattamento con un adenovirus helper-dependent esprimendo apolipoproteina E (n = 6-10). Utilizzando questo modello, pattern di espressione genica nell&#39;aorta sono stati analizzati in una prima fase di regressione da microarray. Dopo rigorose analisi statistica, abbiamo trovato che l&#39;espressione di IL-7 &egrave; significativamente ridotta in risposta all&#39;abbassamento del colesterolo (n = 6). Per capire l&#39;importanza di IL-7 down-regulation per regressione aterosclerotica, abbiamo studiato gli effetti e meccanismi d&#39;azione di IL-7 sulle cellule endoteliali (ECs) oltre in vitro che in vivo. I nostri risultati principali sono: (i) IL-7 up-regola molecole di adesione cellulare e monocyte chemoattractant protein-1 in ECs e promuove l&#39;adesione di monociti ECS; (ii) il presente regolamento &egrave; mediato dal fosfatidilinositolo 3-chinasi (PI3K) / AKT-dipendente e - indipendenti attivazione di NF-&kappa;B; (iii) elevazione del plasma IL-7 induce reclutamento dei monociti/macrofagi all&#39;endotelio senza influire sul colesterolo del plasma (n = 5, 6); e (4) mancanza di IL-7 in cellule derivate dal midollo osseo riduce la migrazione dei monociti/macrofagi per le lesioni (n = 5, 6). The circadian clock has been shown to regulate metabolic homeostasis. Mice with a deletion of Bmal1, a key component of the core molecular clock, develop hyperglycemia and hypoinsulinemia, suggesting Î²-cell dysfunction. However, the underlying mechanisms are not fully known. In this study, we investigated the mechanisms underlying the regulation of Î²-cell function by Bmal1. We studied Î²-cell function in global Bmal1-/- mice, in vivo and in isolated islets ex vivo, as well as in rat insulinoma cell lines with shRNA-mediated Bmal1 knockdown. Global Bmal1-/- mice develop diabetes secondary to a significant impairment in glucose-stimulated insulin secretion (GSIS). There is a blunting of GSIS in both isolated Bmal1-/- islets and in Bmal1 knockdown cells, as compared to controls, suggesting that this is secondary to a loss of cell-autonomous effect of Bmal1. In contrast to previous studies, in these Bmal1-/- mice on a C57Bl/6 background, the loss of stimulated insulin secretion, interestingly, is with glucose but not to other depolarizing secretagogues, suggesting that events downstream of membrane depolarization are largely normal in Bmal1-/- islets. This defect in GSIS occurs as a result increased mitochondrial uncoupling with consequent impairment of glucose-induced mitochondrial potential generation and ATP synthesis, due to an upregulation of Ucp2. Inhibition of Ucp2, in isolated islets, leads to a rescue of the glucose-induced ATP production and insulin secretion in Bmal1-/- islets. Thus, Bmal1 regulates mitochondrial energy metabolism to maintain normal GSIS and its disruption leads to diabetes due to a loss of GSIS.

Loss of Bmal1 leads to uncoupling and impaired glucose-stimulated insulin secretion in Î²-cells.

Hemmung der Ucp2, isolierten Inseln, f&uuml;hrt zu einer Rettungsaktion der Glukose-induzierte ATP Produktion und Insulin-Sekretion in Bmal1/Inselchen. So Bmal1 regelt die mitochondriale Energiestoffwechsel um normale GSIS beizubehalten und seine St&ouml;rung f&uuml;hrt zu Diabetes mit einem Verlust von GSIS.

Bmal1 &#xC190;&#xC2E4; uncoupling &#xBC0F; &#xC7A5;&#xC560;&#xC778; &#xD3EC;&#xB3C4; &#xB2F9; &#xC790;&#xADF9; &#xC778;&#xC290;&#xB9B0; &#xBD84; &#xBE44; &beta; &#xC138;&#xD3EC;&#xC5D0; &#xC5F0;&#xACB0; &#xB429;&#xB2C8;&#xB2E4;.

El reloj circadiano se ha demostrado que regulan la homeostasis metab&oacute;lica. Ratones con una canceladura de Bmal1, un componente clave del reloj molecular base, desarrollan hiperglucemia e hipoinsulinemia, sugiriendo la disfunci&oacute;n de las c&eacute;lulas &beta;. Sin embargo, los mecanismos subyacentes no se conocen completamente. En este estudio, investigamos los mecanismos subyacentes a la regulaci&oacute;n de la funci&oacute;n de las c&eacute;lulas &beta; por Bmal1. Se estudi&oacute; la funci&oacute;n de las c&eacute;lulas &beta; en global Bmal1-/-ratones, en vivo y en islotes aislados ex vivo, as&iacute; como en l&iacute;neas de celulares de insulinoma rata con ca&iacute;da de Bmal1 shRNA-mediada. Global Bmal1-/-ratones desarrollan diabetes secundaria a un deterioro significativo en la secreci&oacute;n de insulina estimulada por glucosa (GSIS). Hay un embotamiento de GSIS en ambos Bmal1/islotes aislados y en c&eacute;lulas de precipitaci&oacute;n de Bmal1, en comparaci&oacute;n con controles, sugiriendo que esto es secundario a una p&eacute;rdida de c&eacute;lula aut&oacute;noma efecto de Bmal1. A diferencia de estudios anteriores, en estos Bmal1-/-ratones sobre un fondo de C57Bl/6, la p&eacute;rdida de la secreci&oacute;n de insulina estimulada, curiosamente, es con glucosa, pero no a otros secretagogos despolarizaci&oacute;n, sugiriendo que los eventos aguas abajo de la despolarizaci&oacute;n de la membrana son en gran parte normales en Bmal1-/-islotes. Este defecto en GSIS ocurre como un resultado hab&iacute;a aumentado desacoplamiento mitocondrial con el consiguiente deterioro del valor de generaci&oacute;n potencial mitocondrial inducida por glucosa y s&iacute;ntesis de ATP, debido a una regulaci&oacute;n al alza de la Ucp2. Inhibici&oacute;n de la Ucp2, en islotes aislados, conduce a un rescate de la producci&oacute;n de ATP inducida por glucosa y la secreci&oacute;n de insulina en Bmal1-/-islotes. As&iacute;, Bmal1 regula el metabolismo energ&eacute;tico mitocondrial para mantener GSIS normal y su interrupci&oacute;n conduce a la diabetes debido a la p&eacute;rdida de GSIS.
