Die experimentelle autoimmune Enzephalomyelitis kann verhindert werden und durch eine Infektion mit Trypanosoma cruzi geheilt werden.

Trypanosoma cruzi est un parasite intracellulaire qui induit une forte r&#xE9;ponse de type Th1 et l&#x26;#39;immunosuppression au cours de la phase aigu&#xEB; de l&#x26;#39;infection. Pour &#xE9;tudier la fa&#xE7;on dont l&#x26;#39;infection par T. cruzi serait de moduler le d&#xE9;veloppement d&#x26;#39;une maladie auto-immune, nous avons immunis&#xE9; des souris C57BL / 6 et IL-10 ou iNOS souris knock-out de l&#x26;#39;arri&#xE8;re-plan avec le m&#xEA;me enc&#xE9;phalitog&#xE8;ne MOG 35-55 peptide et les infect&#xE9;s par le T. cruzi. Nos r&#xE9;sultats d&#xE9;montrent que l&#x26;#39;infection par T. cruzi emp&#xEA;che compl&#xE8;tement le d&#xE9;veloppement EAE et induit en outre une r&#xE9;mission compl&#xE8;te et durable chez les souris qui ont &#xE9;t&#xE9; infect&#xE9;s par ce parasite, apr&#xE8;s qu&#x26;#39;ils avaient d&#xE9;velopp&#xE9; EAE clinique. L&#x26;#39;oxyde nitrique et de l&#x26;#39;IL-10 de participer dans le d&#xE9;clenchement des m&#xE9;canismes associ&#xE9;s &#xE0; la suppression EAE par l&#x26;#39;infection. Diminution des fr&#xE9;quences lymphoprolif&#xE9;ration et l&#x26;#39;augmentation de l&#x26;#39;annexine cellules positives et des cellules T portant CD95, CD95L ou CTLA-4 ont &#xE9;t&#xE9; observ&#xE9;es dans la rate de souris immunis&#xE9;es / infect&#xE9;s, ainsi que la r&#xE9;duction d&#x26;#39;IL-2 et a augment&#xE9; la production de TGF-b&#xEA;ta en comparaison avec seulement immunis&#xE9; des souris. Nos r&#xE9;sultats indiquent que plusieurs m&#xE9;canismes effecteurs et r&#xE9;gulateurs de la r&#xE9;ponse immunitaire qui surviennent pendant la phase aigu&#xEB; de l&#x26;#39;infection par T. cruzi durablement affecter les fonctions d&#x26;#39;extension et / ou de cellules effectrices enc&#xE9;phalitog&#xE8;nes, pr&#xE9;venir l&#x26;#39;apparition ou d&#x26;#39;induire une r&#xE9;mission compl&#xE8;te de l&#x26;#39;EAE.

Trypanosoma cruzi is an intracellular parasite that induces a strong Th1-type response and immunosuppression during the acute phase of infection. To study how the infection with T. cruzi would modulate the development of an autoimmune disease, we immunized C57BL/6 mice and IL-10 or iNOS knock-out mice of the same background with the encephalitogenic MOG 35-55 peptide and infected them with T. cruzi. Our results demonstrate that infection with T. cruzi completely prevents EAE development and furthermore induces complete and lasting remission in mice that were infected with this parasite after they had developed clinical EAE. Nitric oxide and IL-10 participate in triggering the mechanisms associated with EAE suppression by the infection. Decreased lymphoproliferation and increased frequencies of Annexin-positive cells and of T cells bearing CD95, CD95L or CTLA-4 were observed in the spleen from immunized/infected mice, as well as lower IL-2 and increased TGF-beta production in comparison with only immunized mice. Our results indicate that several effector and regulatory mechanisms of the immune response that arise during the acute phase of T. cruzi infection lastingly affect the expansion and/or effector functions of encephalitogenic cells, preventing the onset or inducing complete remission of EAE.

Experimental autoimmune encephalomyelitis can be prevented and cured by infection with Trypanosoma cruzi.

Trypanosoma cruzi &egrave; un parassita intracellulare che induce una forte risposta di tipo Th1 e immunosoppressione durante la fase acuta dell&#39;infezione. Per studiare come l&#39;infezione da T. cruzi sarebbe modulano lo sviluppo di una malattia autoimmune, siamo immunizzati topi C57BL/6 e IL-10 o iNOS topi knock-out dello stesso sfondo con il peptide MOG 35-55 encephalitogenic e li infettati con T. cruzi. I nostri risultati dimostrano che l&#39;infezione da T. cruzi completamente impedisce lo sviluppo della EAE e inoltre induce la remissione completa e duratura nei topi che sono stati infettati con questo parassita dopo che avevano sviluppato EAE clinico. Ossido nitrico e IL-10 partecipare nell&#39;innescare i meccanismi associati con soppressione EAE dall&#39;infezione. Linfoproliferazione diminuito ed aumentate le frequenze delle cellule Annexin-positive e di cellule T cuscinetto CD95, CD95L o CTLA-4 sono state osservate nella milza da topi immunizzati/infetti, come pure IL-2 inferiore e aumentato la produzione di TGF-beta rispetto ai topi immunizzati soli. I nostri risultati indicano che diversi effettori e meccanismi regolatori della risposta immunitaria che si presentano durante la fase acuta dell&#39;infezione da T. cruzi lastingly influenzano le funzioni di espansione e/o effettrici delle cellule encephalitogenic, prevenire l&#39;insorgenza o indurre una remissione completa della EAE.

Encefalomielite autoimmune sperimentale pu&ograve; essere evitata e curata da infezione da Trypanosoma cruzi.

Trypanosoma cruzi es un par&#xE1;sito intracelular que induce una fuerte respuesta de tipo Th1 y la inmunosupresi&#xF3;n durante la fase aguda de la infecci&#xF3;n. Para estudiar c&#xF3;mo la infecci&#xF3;n con T. cruzi podr&#xED;a modular el desarrollo de una enfermedad autoinmune, se inmunizaron ratones C57BL / 6 e IL-10 o la iNOS ratones knock-out de los mismos antecedentes con la encefalitog&#xE9;nico MOG 35-55 p&#xE9;ptido e infectados con T. cruzi. Nuestros resultados demuestran que la infecci&#xF3;n con T. cruzi impide totalmente el desarrollo de la EAE y, adem&#xE1;s, induce una remisi&#xF3;n completa y duradera en los ratones que fueron infectados con el par&#xE1;sito despu&#xE9;s de que se hab&#xED;a desarrollado EAE cl&#xED;nica. El &#xF3;xido n&#xED;trico y la IL 10-participar en la activaci&#xF3;n de los mecanismos asociados con la supresi&#xF3;n de la EAE por la infecci&#xF3;n. La disminuci&#xF3;n de frecuencias y el aumento de linfoproliferaci&#xF3;n Anexina de c&#xE9;lulas positivas y de las c&#xE9;lulas T con CD95, CD95L o CTLA 4-fueron observados en el bazo de ratones inmunizados / infectados, as&#xED; como bajos de IL-2 y el aumento de la producci&#xF3;n de TGF-beta en comparaci&#xF3;n con s&#xF3;lo ratones inmunizados. IL-12 is one of the main cytokines driving the immune response to a resistant phenotype in leishmaniasis and in several other diseases involving intracellular microbes. In this study, we investigated IL-12 production by mononuclear phagocytes at several developmental stages when stimulated with Leishmania major, L. amazonensis or L. chagasi. Bone marrow cells were cultured for 4-6 days in vitro in the presence of M-CSF, GM-CSF or IL-3. After density separation, only cells banding at the 40-50% Percoll interface, but not those at 20-40% or 50-80% interfaces, produced large amounts of IL-12 p40 when stimulated with LPS or live Leishmania promastigotes. However, only low levels of IL-12 p70 were produced under these conditions. The high IL-12 p40-producing cells could be similarly derived from mouse strains with different susceptibility to Leishmania. Quantitative analysis of monocyte/macrophage lineage marker expression, in combination with positive and negative selection, led to the conclusion that the high IL-12 p40-producing cells are macrophages at an intermediate stage of maturation between immature and fully differentiated cells, expressing ER-HR3 but only low levels of the mature markers, scavenger receptor and CD11b/Mac-1. They do not express any of the precursor markers CD31/ER-MP12, Ly-6C/ER-MP20 or ER-MP58. Because recruitment of monocytes to an infection site and its draining lymph node is a general phenomenon, the notion that, developing from these monocytes, a population of mononuclear phagocytes at an intermediate maturation stage has the capacity to synthesize large amounts of IL-12 p40 has significant bearing on our understanding of immune regulation in leishmaniasis and also in infections by other pathogens.

Dans cette &#xE9;tude, nous avons &#xE9;tudi&#xE9; la production d&#x26;#39;IL-12 par les phagocytes mononucl&#xE9;aires &#xE0; plusieurs stades de d&#xE9;veloppement quand ils sont stimul&#xE9;s par Leishmania major, L. amazonensis ou L. chagasi. Cellules de moelle osseuse ont &#xE9;t&#xE9; cultiv&#xE9;es pendant 4-6 jours in vitro en pr&#xE9;sence de M-CSF, GM-CSF ou IL-3. Apr&#xE8;s s&#xE9;paration par densit&#xE9;, seules les cellules de baguage &#xE0; l&#x26;#39;interface 40-50% de Percoll, mais pas ceux &#xE0; 20-40% ou 50-80%, les interfaces produit de grandes quantit&#xE9;s d&#x26;#39;IL-12 p40 lorsqu&#x26;#39;il est stimul&#xE9; avec promastigotes de Leishmania LPS ou en direct. Toutefois, seuls les faibles niveaux d&#x26;#39;IL-12 p70 ont &#xE9;t&#xE9; produites dans ces conditions. L&#x26;#39;IL-12 de haute cellules p40-producteurs pourrait &#xEA;tre similaire provenant de souches de souris pr&#xE9;sentant une sensibilit&#xE9; diff&#xE9;rente &#xE0; Leishmania. IL-12 &egrave; uno dei principali citochine guida la risposta immunitaria ad un fenotipo resistente a leishmaniosi e in diverse altre malattie che coinvolgono i microbi intracellulari. In questo studio, abbiamo studiato la produzione di IL-12 da fagociti mononucleati a diversi stadi di sviluppo quando stimolato con Leishmania major, L. amazonensis o L. chagasi. Le cellule del midollo osseo sono state coltivate per 4-6 giorni in vitro in presenza di M-CSF, GM-CSF o IL-3. Dopo la densit&agrave; di separazione, solo cellule banding presso l&#39;interfaccia di Percoll 40-50%, ma non quelli a 50-80% o del 20-40% le interfacce prodotte grandi quantit&agrave; di IL-12 p40 quando stimolate con LPS o live Leishmania promastigotes. Perch&eacute; il reclutamento dei monociti a un sito di infezione e il linfonodo drenante &egrave; un fenomeno generale, la nozione che, sviluppando da questi monociti, una popolazione dei fagociti mononucleati in una fase di maturazione intermedia ha la capacit&agrave; di sintetizzare grandi quantit&agrave; di IL-12 p40 ha cuscinetto significativo sulla nostra comprensione della regolazione immunitaria nella leishmaniosi e anche nelle infezioni da altri patogeni.

IL-12 es una de las citoquinas principales de conducci&#xF3;n de la respuesta inmune a un fenotipo resistente en la leishmaniasis y en varias otras enfermedades que implican microorganismos intracelulares. En este estudio, se investig&#xF3; la producci&#xF3;n de IL-12 por los fagocitos mononucleares en varias etapas de desarrollo cuando se estimula con Leishmania major, L. amazonensis y L. chagasi. C&#xE9;lulas de m&#xE9;dula &#xF3;sea se cultivaron durante 4-6 d&#xED;as in vitro en presencia de M-CSF, GM-CSF o IL-3. Despu&#xE9;s de separaci&#xF3;n por densidad, s&#xF3;lo las c&#xE9;lulas de bandas en el 40-50% de Percoll interfaz, pero no los que est&#xE1;n en un 20-40% o 50-80% de interfaces, producido grandes cantidades de IL-12 p40 cuando son estimuladas con LPS o en vivo promastigotes de Leishmania. Sin embargo, s&#xF3;lo bajos niveles de IL-12 p70 fueron producidos bajo estas condiciones. La IL-12 p40 alta c&#xE9;lulas que producen podr&#xED;an ser similarmente originadas a partir de cepas de rat&#xF3;n con diferente susceptibilidad a Leishmania. El an&#xE1;lisis cuantitativo de expresi&#xF3;n linaje marcador de monocitos / macr&#xF3;fagos, en combinaci&#xF3;n con selecci&#xF3;n positiva y negativa, llev&#xF3; a la conclusi&#xF3;n de que la IL-12 p40 alta c&#xE9;lulas productoras son los macr&#xF3;fagos en una fase intermedia de maduraci&#xF3;n entre las c&#xE9;lulas inmaduras y diferenciado completamente, expresando ER- HR3, pero s&#xF3;lo los bajos niveles de los marcadores maduros, receptor scavenger y CD11b/Mac-1. Ellos no expresan ninguno de los marcadores de precursores CD31/ER-MP12, Ly-6C/ER-MP20 o ER-MP58. Debido a que el reclutamiento de monocitos a un sitio de la infecci&#xF3;n y su nodo de drenaje de la linfa es un fen&#xF3;meno general, la idea de que, el desarrollo de estos monocitos, una poblaci&#xF3;n de fagocitos mononucleares en una etapa de maduraci&#xF3;n intermedia tiene la capacidad de sintetizar grandes cantidades de IL-12 p40 tiene incidencia importante en nuestra comprensi&#xF3;n de la regulaci&#xF3;n inmune en la leishmaniasis, as&#xED; como en las infecciones causadas por otros pat&#xF3;genos.

Hemos observado que mientras Tregs completamente impedido la divisi&#xF3;n celular de c&#xE9;lulas T que muestran una baja afinidad por ligandos auto, ten&#xED;an una menos marcada, aunque efecto significativo, a la entrada del ciclo celular de las c&#xE9;lulas T que muestran una mayor afinidad. Homeostatic proliferation of T cells leads to the generation of effector/memory cells, which have the potential to cause harm to the host. The role of Tregs in the control of homeostatic proliferation is unclear. In this study we utilized mice that either harbor or lack Tregs as recipients of monoclonal or polyclonal T cells. We observed that while Tregs completely prevented cell division of T cells displaying low affinity for self ligands, they had a less marked, albeit significant, effect on cell cycle entry of T cells displaying higher affinity. The presence of Tregs resulted in a lower accumulation of T cells, enhanced apoptosis, and impaired differentiation to a cytokine-producing state. We conclude that Tregs play a major role in the control of homeostatic proliferation.

Dans cette &#xE9;tude nous avons utilis&#xE9; des souris qui soit le port ou l&#x26;#39;absence Tregs en tant que b&#xE9;n&#xE9;ficiaires de cellules T monoclonaux ou polyclonaux. Nous avons observ&#xE9; que, bien que Tregs a compl&#xE8;tement emp&#xEA;ch&#xE9; la division cellulaire des cellules T pr&#xE9;sentant une faible affinit&#xE9; pour les ligands de soi, ils avaient une moins marqu&#xE9;e, quoique significative, l&#x26;#39;effet sur l&#x26;#39;entr&#xE9;e du cycle cellulaire des cellules T pr&#xE9;sentant une affinit&#xE9; plus &#xE9;lev&#xE9;e. La pr&#xE9;sence de Tregs a entra&#xEE;n&#xE9; une moindre accumulation de cellules T, l&#x26;#39;apoptose accrue, et la diff&#xE9;renciation avec facult&#xE9;s affaiblies &#xE0; un &#xE9;tat de cytokine production. Using intravital two-photon laser scanning microscopy we determined that, in the absence of T reg cells, the locomotion of autoantigen-specific T cells inside lymph nodes is decreased, and the contacts between T cells and antigen-loaded dendritic cells (DCs) are of longer duration. Thus, T reg cells can exert an early effect on immune responses by attenuating the establishment of stable contacts during priming of naive T cells by DCs.

IL-17-producing T lymphocytes have been recently shown to comprise a distinct lineage of proinflammatory T helper cells, termed Th17 cells, that are major contributors to autoimmune disease. We show here that the orphan nuclear receptor RORgammat is the key transcription factor that orchestrates the differentiation of this effector cell lineage. RORgammat induces transcription of the genes encoding IL-17 and the related cytokine IL-17F in naÃ¯ve CD4(+) T helper cells and is required for their expression in response to IL-6 and TGF-beta, the cytokines known to induce IL-17. Th17 cells are constitutively present throughout the intestinal lamina propria, express RORgammat, and are absent in mice deficient for RORgammat or IL-6. Mice with RORgammat-deficient T cells have attenuated autoimmune disease and lack tissue-infiltrating Th17 cells. Together, these studies suggest that RORgammat is a key regulator of immune homeostasis and highlight its potential as a therapeutic target in inflammatory diseases.

Many studies have positioned Notch signaling at various critical junctions during T-cell development. There is, however, debate regarding the role of Notch in the CD4 versus CD8 lineage commitment. Because there are 4 Notch receptors and RBP-Jkappa-independent Notch signaling has been reported, we decided to eliminate gamma-secretase activity once its activity is required for all forms of Notch signaling. T-cell-specific elimination of gamma-secretase was carried out by crossing presenilin-1 (PS1) floxed mice with CD4-Cre mice and PS2 KO mice, generating PS KO mice. Thymic CD4+CD8+ double-positive (DP) cells from these mice were strikingly resistant to apoptosis by anti-CD3 treatment in vivo and expressed more Bcl-X(L) than control thymocytes, and deletion of only one allele of Bcl-X(L) gene restored wild-type levels of sensitivity to apoptosis. In addition, these PS KO animals displayed a significant decrease in the number of CD8+ T cells in the periphery, and these cells had higher level of phosphorylated p38 than cells from control littermates. Our results show that ablation of presenilins results in deficiency of CD8 cells in the periphery and a dramatic change in the physiology of thymocytes, bringing to our attention the potential side effects of presenilin inhibitors in ongoing clinical trials.

The mouse spinal cord is an important site for autoimmune and injury models. Skull thinning surgery provides a minimally invasive window for microscopy of the mouse cerebral cortex, but there are no parallel methods for the spinal cord. We introduce a novel, facile and inexpensive method for two-photon laser scanning microscopy of the intact spinal cord in the mouse by taking advantage of the naturally accessible intervertebral space. These are powerful methods when combined with gene-targeted mice in which endogenous immune cells are labeled with green fluorescent protein (GFP). We first demonstrate that generation of the intervertebral window does not elicit a reaction of GFP(+) microglial cells in CX3CR1(gfp/+) mice. We next demonstrate a distinct rostrocaudal migration of GFP(+) immune cells in the spinal cord of CXCR6(gfp/+) mice during active experimental autoimmune encephalomyelitis (EAE). Interestingly, infiltration of the cerebral cortex by GFP(+) cells in these mice required three conditions: EAE induction, cortical injury and expression of CXCR6 on immune cells.

Das Maus-R&uuml;ckenmark ist ein wichtiger Standort f&uuml;r autoimmune und Verletzungen-Modelle. Sch&auml;del Ausd&uuml;nnung Chirurgie bietet eine minimal-invasive Fenster f&uuml;r die Mikroskopie der Gro&szlig;hirnrinde Maus, aber es gibt keine parallelen Verfahren f&uuml;r das R&uuml;ckenmark. Wir f&uuml;hren einen Roman, einfache und kosteng&uuml;nstige Methode f&uuml;r die zwei-Photonen-Laser-Rastermikroskopie des R&uuml;ckenmarks intakt in die Maus unter Ausnutzung der nat&uuml;rlich zug&auml;nglich Foramina Raum. Hierbei handelt es sich um leistungsf&auml;hige Methoden in Kombination mit Gen-bezogene M&auml;use in der endogene Immunzellen mit gr&uuml;n fluoreszierendes Protein (GFP) beschriftet sind. Zuerst zeigen wir, dass die Generation der Foramina Fenster eine Reaktion der GFP(+) Microglial Zellen bei CX3CR1(gfp/+) M&auml;usen nicht entlocken wird. Als N&auml;chstes zeigen wir eine unterschiedliche Rostrocaudal Migration von GFP(+) Immunzellen im R&uuml;ckenmark von M&auml;usen CXCR6(gfp/+) w&auml;hrend der aktiven experimentellen autoimmunen Enzephalomyelitis (EAE). Interessanterweise Infiltration der Gro&szlig;hirnrinde von GFP(+) Zellen in diesen M&auml;usen drei Voraussetzungen erforderlich: EAE Induktion, kortikale Verletzungen und Berechnung der CXCR6 auf Immunzellen.
