1. 动物护理
注意:LLB 的动物模型仅受其可用性和减震管容量的限制，以适应其大小。此处描述的休克管是专门为小鼠设计的。
<ol><li>根据实验需要，使用 3-4 个月大的雄性或雌性 C57BL/6J 小鼠或其他经批准的小鼠品系/品系。将小鼠维持在特定的无病原体设施中 12 小时的暗光循环中，可 随意 获取食物和水。小鼠通常被关在一个笼子里，有 4 或 5 只。将设施温度保持在 20-22 °C。</li>
	<li>将装有 Mlast 和 Sam 老鼠的笼子带到附近的饲养区。带上单独的空笼子，以便将单只小鼠转移到鼓炎室或从喷砂室转移。</li>
</ol>2. 减震管准备
<ol><li>（安全检查） 确认已完成特定系统的必要安全检查。确保气源（氦气）和主电源已关闭/断开。</li>
	<li>根据需要准备膜，以进行特定数量的低强度喷射（图 1.1）。根据本协议中使用的特定减震管的要求切割膜尺寸:<ol><li>将一张塑料保鲜膜切成 5.5 英寸 x 5.5 英寸的正方形以密封线轴，使其加压。</li>
		<li>将一张标准 8.5 英寸 x 11 英寸复印纸（75 克/米2 重量）切割成 5.5 英寸 x 11 英寸;将生成的纸张对折，形成一个 5.5 英寸 x 5.5 英寸的正方形。</li>
		<li>获得一张 500 G 聚酯薄膜（125 μm 厚度）。 注意:这些片材不会因标准低强度喷砂而破裂或明显变形，并且可以在一天的手术期间重复使用。</li>
	</ol></li>
	<li>取一块正方形的保鲜膜和一块折叠的纸，将它们放在平坦的表面上（图 1.2）。将折叠好的纸放在保鲜膜的顶部，并尽可能将两者对齐（图 1.3）。为了加快重复的爆炸速度，现在就安排好所有的膜堆栈。</li>
	<li>将聚酯薄膜卷成一根小管（大约与食指一样大;图 1.4,1.5）。将其完全插入机构中，然后松开以使其在将驱动器部分与线轴分开的橡胶密封件上展开。将线轴推向驱动器以将聚酯薄膜片固定到位;这将从减震管的驱动部分解封线轴。</li>
	<li>将手指放在保鲜膜的上半部分下方，小心地将保鲜膜和纸张朝您的方向滚动，确保它们一起卷起而不会错位（图 1.6）。</li>
	<li>将膜组插入线轴和减震管的驱动部分之间（图 1.7）。</li>
	<li>让膜叠展开，使塑料密封件朝向线轴，而纸张朝向管的驱动部分（图 1.8）。 注意:此方向将形成气密密封，以便系统可以加压。</li>
	<li>关闭线轴组件（图 1.9、1.10）。根据需要，用手或液压拧紧螺栓，固定驾驶员滑阀芯减震管组件，以便系统可以加压。（安全检查;图 1.10） 注意:对于液压系统，请确保达到瓶盖组件的目标压力，以防止失火，这可能需要更换膜并减慢 LLB 暴露过程。我们使用液压系统在 500 psi 的压力下关闭我们的组件。</li>
</ol>3. 动物准备
<ol><li>打开麻醉室下方的循环水加热垫，温度设置为 37 °C（图 1.11）。在加热垫顶部放置一个吸水医用垫。</li>
	<li>在拘留室中，从其家笼中取出一只鼠标，并将其放入空的转移笼中。将笼中老鼠带入喷砂室。</li>
	<li>将氧气流速调至 1.0 L/min （lpm） 并打开真空扫气系统（图 1.12）。</li>
	<li>将异氟醚调至 5%（以诱导快速失去知觉）并将流量引导至啮齿动物麻醉室（图 1.13）。</li>
	<li>将鼠标放入腔室中以诱导麻醉（图 1.14）。</li>
	<li>一旦小鼠完全麻醉并表现出稳定的呼吸 30 秒，伸手进入腔室并敲击鼠标，以便在研究的剩余时间内明确地长期识别小鼠。现在执行此步骤是必要的，以避免干扰爆炸后的恢复时间。然后，将无菌眼用润滑剂涂抹在双眼上，以防止角膜干燥。</li>
	<li>从腔室中取出鼠标，将其鼻子放入鼻锥中（图 1.15）。将麻醉液（例如异氟醚）从诱导室切换到鼻锥。</li>
	<li>使用小块实验室胶带轻轻地将小鼠的四肢束缚在轮床上（图 1.16）。</li>
	<li>束缚鼠标后，将金属丝扭结带绕在每个肢体上并拧紧，将鼠标固定在手腕和脚踝的轮床上（图 1.17）。在胸部周围放一条较大的扭结带，系得非常松散，这样老鼠的呼吸就不会受到限制。这将作为辅助约束机制，以防任何肢体约束装置松动。</li>
	<li>抬起鼠标的尾巴并将其放在左脚下方，以确保在将轮床插入减震管时它不会被夹住（图 1.18）。</li>
</ol>4. 法学学士程序
<ol><li>打开激波管的动物暴露部分并调整鼠标的方向，使其面向迎面而来的冲击波（图 1.19）。</li>
	<li>将轮床固定/悬挂在动物暴露部分（图 1.20）。</li>
	<li>关紧动物暴露部分的门，确保麻醉液流管不被门夹住（图 1.21）。</li>
	<li>将麻醉剂减少至 2.5-3% 异氟醚，在剩余的疗程中为 1 升/分钟。</li>
	<li>根据需要为系统供电（图 1.22）。</li>
	<li>找到并连接压缩氦气的供应管线（图 1.23、1.24）。</li>
	<li>离开喷砂室，进入相邻房间的喷砂管控制台，并确保喷砂室内没有人员或动物。 注意:机构或运营条件可能需要听力保护。此类情况可能包括控制台与减震管位于同一开放空间的减震管布置。</li>
	<li>在控制台上，打开采集软件以记录爆炸事件（参见图 1.25 中的绿色框）。 注意:对于这些程序，我们以 20 千克赫兹 （kHz） 的采样率收集传感器数据，然后使用 LabView 软件进行处理。我们建议以 ≥10 kHz 的频率采集传感器采样，以获得高质量的时间与压力曲线。</li>
	<li>松开任何安全锁（例如，电源控制键，如图 1.26 中的绿色箭头所示）。</li>
	<li>关闭两个排气口并被动地对阀芯加压（图 1.27）。请勿使用驾驶员侧。继续填充，直到膜在目标峰 psi 处自行破裂，该峰由使用的膜片数量决定。</li>
	<li>记录动物位置的峰值压力、正相位持续时间和脉冲。（图 1.28）。关闭填充机制。</li>
	<li>返回减震管，断开氦气进料管，并关闭爆炸控制电路的电源（图 1.29）。</li>
	<li>要对同一只动物进行重复的 LLB 暴露，请打开线轴，取出线轴膜堆栈，然后滚动并插入另一个线轴膜堆栈（图 1.30、1.31、1.32）。展平膜堆栈并重新合上组件。 注意:为了模拟经验定义的 SOF 训练期间低水平爆炸暴露的临床体验，我们每天将小鼠暴露于 5-6 个 LLB，将每日暴露限制在保守的 ~20 累积总 psi45。强调机制和剂量-反应关系的研究可以选择使用一致数量的 LLB 暴露，每次会话具有定义的超压参数。</li>
	<li>在当前动物的最终 LLB 之后，将其从休克管中取出，保持麻醉开启（图 1.33）。</li>
	<li>在动物处于麻醉状态时解开动物的绳子。将其从麻醉鼻锥中取出，将其仰卧在加热的水垫上（图 1.34）。</li>
	<li>一旦动物被放在水垫上，启动一个计时器并记录鼠标自行翻转到其腹侧（即胃）的时间（图 1.35）。将此时间记录为扶正时间。鼠标恢复后，将其放回笼子并根据需要继续监控。</li>
</ol>5. 多日程序
<ol><li>为了模拟 SOF 近距离战斗训练期间使用的违规指控的常规 LLB 暴露，每周 5 天（周一至周五）每天对小鼠进行重复暴露，总共 15 天，共 3 个标准工作周。</li>
</ol>6. 改变 LLB 峰值压力
<ol><li>通过使用更坚固的膜材料或简单地堆叠额外的膜来提高峰值压力。例如，使用 Mylar Roll Clear 0.005 （500 G） 膜产生 ~20 psi 峰值压力（当用作驱动膜和阀芯膜时）或 Mylar Roll Clear 0.002 （200 G） 膜产生 ~10 psi 峰值压力。</li>
	<li>调整 blast 的正相位持续时间和脉冲参数，以满足实验需求。要调整正相位持续时间和脉冲，请通过替换压缩气源47,49 或尽可能更改驱动器长度来凭经验确定目标条件。上述协议使用氦气产生类似于理想化的弗里德兰德曲线的尖锐峰值压力和波形。</li>
</ol>7. 组织采集
注:组织采集实践可根据实验需要进行调整。
<ol><li>通过腹膜内注射 210 mg/kg 戊巴比妥麻醉小鼠。</li>
	<li>将鼠标放入带有杆或预制网格的小鼠或鼠笼中;将笼式鼠标放入通风橱中。</li>
	<li>一旦鼠标没有反应，将其仰卧在笼子顶部的杆上，并在其中一根杆周围闭上嘴巴，以帮助它在灌注期间保持原位。</li>
	<li>抓住胃的皮肤，向上拉，用一把大剪刀在腹腔上剪一个洞，小心不要割伤任何器官。继续沿着肋骨底部向下切得更远，以便更自由地铰接胸腔。</li>
	<li>使用止血钳，从侧面接近鼠标，直接抓住胸腔顶部的组织，将止血钳向后滚动，以保持胸腔底部的角度处于易于接近的位置。使用一把镊子或类似工具将止血钳固定到位。</li>
	<li>使用一把小手术剪刀，小心地剪断隔膜，以便进入心脏。使用一对镊子轻轻地调整心脏的角度，使底部直接面向胸腔的开放底部。快速工作，使心脏在灌注期间仍会跳动。</li>
	<li>如果采集血液，用一对镊子夹住心脏，并使用 3 mL 注射器小心地刺穿右心室，注射器尖端有 0.5 英寸 25 G 针头。从心室底部插入并纵向插入，注意不要刺穿心室的另一侧。轻轻拉动注射器，直到收集到 0.5-1.0 mL 血液或血流停止，然后取出注射器。</li>

	<li>灌注动物。<ol><li>将装有 50 mL 磷酸盐缓冲盐水 （PBS） 的注射器连接到蝶形针头上，并以大约 10 mL/min 的速率灌注。寻找肝脏漂白作为适当灌注的标志。注射器排空后，将其与蝶形针头断开。</li>
		<li>为了制备用于显微镜的组织，用含有 50 mL 10% 中性缓冲福尔马林 （NBF） 或 4% 甲醛溶液的注射器替换空的 PBS 注射器。重复上述步骤以灌注福尔马林。 注意:应观察灌注小鼠在灌注过程中抽搐;这应该会导致手术完成后全身僵硬或僵硬。</li>
	</ol></li>
	<li>从心脏中取出蝶针，从笼杆中取出鼠标以进行组织收集。</li>
	<li>根据需要对靶器官进行切除和细分;收集新鲜、未固定的材料时，请小心在冰上执行程序。</li>
	<li>快速冷冻在液氮中收集的任何未固定组织，并将其储存在 -80 °C 下，直至用于检测蛋白质或 RNA 靶标的方案。</li>
	<li>对于固定组织，取出到装有福尔马林的标记 50 mL 锥形管中（每个器官 1 管）。</li>
</ol>

Shocktube 方法模拟小鼠高度重复的低水平爆炸

作者没有经济利益需要披露。

我们无法充分治疗我们不充分理解的东西，我们还不了解与高度重复的 LLB 暴露相关的损伤机制。许多 SOF 人员报告说，在运营服务的 5 到 10 年内，被认为与高度重复的 LLB 暴露有关的健康相关损害的发展50,51。一些 SOF 人员在接触 LLB 后立即出现急性创伤性脑损伤 （TBI） 样神经认知影响39。此外，临床医生报告说，原始细胞暴露引起的症状通常对传统治疗无效，这可能促使 SOF 和临床医生寻求替代疗法52,53。尽管 SOF 经常暴露于 LLB 和超压机制45，由此产生的症状的严重程度和治疗耐药性，以及记录在案的与爆炸相关的星形胶质细胞瘢痕形成模式51，但长期健康结果仍然相对未知。临床医生和军队领导依靠建模研究来揭示损伤机制和病理生理学。这些模型对于制定早期识别、中断、预防和治疗病理过程的政策和策略至关重要。
至关重要的是，常见军用 LLB 暴露的小鼠建模有望为健康预测模型提供信息。临床实践将受益于 LLB 预测模型，这些模型可以确定谁可能最有可能患上与爆炸相关的病理，哪些爆炸特性会引发最严重的后果，以及疾病过程如何根据爆炸暴露的慢性性、剂量或特异性而演变。因此，对重复的 LLB 暴露进行建模对于开发暴露将如何影响 SOF 和其他服务成员的健康结果的假设和预测至关重要。预测和伤害机制模型将为诊断和治疗以及基于症状和暴露的重返工作岗位决策提供信息。
近年来，小鼠爆炸诱导的 TBI （bTBI） 的研究取得了重大进展，特别是随着预测人类慢性重复轻度 bTBI 后结果的模型的开发54,55。虽然使用电击管对中高水平爆炸暴露的研究已经发展成熟，有数百篇 PubMed 索引文章 46,56,57,58，但在常规军事训练超压（<6 psi 峰值压力40）附近的爆炸研究中使用电击管的使用不太发达，在最近的 PubMed 搜索中确定的文章不到10 篇 19,20， 22,23,26,27,28。为了促进这个研究不足的领域的发展，所提出的模型侧重于小鼠持续 LLB 超压、爆炸后恢复和监测的关键考虑因素，同时指出该模型相对于使用露天炸药的几个明显优势。事实上，我们认为所描述的实验室 LLB 模型可能能够开发慢性重复 LLB 后临床结果的预测模型。
与开场爆炸模型相比，LLB 模型具有关键优势，尤其是在动物福利方面。旷场模型可能导致 3-8% 的死亡率25,36，而这种基于实验室的 LLB 模型显示没有损失。这种区别至关重要，尤其是在模拟军事训练典型的高累积暴露时，几乎没有受训者会因 LLB 暴露而经历致命后果。明显不存在呼吸暂停或其他死亡原因，例如致命的肺外伤，确保了该模型的可靠性和一致性，使其成为重复 LLB 临床相关影响研究的首选。
该协议专用于具有三部分设计的"开放式"减震管，由驱动器、阀芯和从动部分组成。高度重复的 LLB 可以通过其他减震管设计来实现，只需对方案进行适当修改即可。开放式减震管设计经常用于爆炸引起的神经创伤的研究 46,47,48。开口激波管具有开放的出口端，允许产生的冲击波在离开管的另一端之前，沿着与目标（例如，动物受试者）相遇的管子长度自由传播。这种设计有助于再现和研究相对纯净的初级爆炸超压，近似于爆炸爆炸的特性，因为它们发生在开阔场地48。因此，将经验测量的爆炸超压波的保真度与理想化的弗里德兰德波进行了比较;这允许评估管材性能以产生特定的超压事件。为了模拟 LLB 暴露，我们使用了前面描述的48 个定制的开炸管，最初设计用于再现超过 200+ 磅三硝基甲苯 （TNT） 在 ~25 英尺的对峙距离下 HLB 爆炸的效果。为了实现高峰值超压，将气体加压到驱动器中，驱动器通过膜与阀芯隔开，从而密封驱动器中的气体。反过来，线轴也被另一个膜与开口部分隔开。第二个膜允许对阀芯单独加压。双腔系统允许驱动器中的气体加压超过膜的正常破裂点。发生这种情况是因为加压阀芯充当缓冲器，在驱动器和阀芯的界面处支撑膜，从而防止其破裂。当减震管操作员希望在目标压力下产生冲击波时，电子阀从阀芯中排出气体，迅速降低阀芯中的压力，并允许驱动部分的超压气体破裂驱动器和阀芯膜，并迅速向下扩展管的长度，在目标区域中遇到动物。能够研究这种设计的高性能管材中 LLB 的关键修改是，我们挡住了驱动器，并且仅将线轴与低阈值膜结合使用。
为了确保 LLB 实验的可靠性和可重复性，在设置过程中必须采取某些措施。将手臂和腿紧紧固定在手腕和脚踝处至关重要。这最大限度地减少了身体运动和爆炸暴露的可变性，并防止了可能混淆结果的意外伤害。此外，向内旋转手腕和脚踝有助于将附属物向动物中线移动，从而降低可能影响后续运动性能评估的远端损伤风险。头部的伸直和脊柱弯曲是确保受试者之间均匀爆炸暴露的另一个重要因素，因为它有助于减少运动范围的潜在差异。对于持续数天或数周的方案，建议增加用于麻醉的异氟醚的百分比。这种调整有助于在整个延长的实验期间保持一致的麻醉深度。根据我们的经验，增加 0.5% 的异氟醚就足以维持足够的麻醉。
然而，并非所有喷丸管设计都无法通过鼻锥进行麻醉输送，尤其是那些具有不允许将管插入驱动部分的完全封闭设计。在这种情况下，注射麻醉剂可能更可取。我们建议确定进行重复连续爆炸所需的时间，然后给予足够的麻醉剂以在整个手术过程中保持昏迷。在开发这种修改后的方法期间，可能需要进行额外的动物福利检查，以确保适当的麻醉维护。此外，使用注射剂可能会导致急性反应后监测（例如收集扶正时间测量值）变得不可能。
在动物研究中，伦理考虑至关重要，这种基于实验室的 LLB 模型结合了全面的爆炸后恢复和监测方案。密切观察爆炸暴露后的人道终点，包括呼吸困难、无法自行扶正、2 小时观察后不能走动状态、癫痫样运动、笨拙运动、视力障碍以及内出血或四肢骨折的证据。值得注意的是，LLB 原始小鼠在我们的实验中没有表现出任何这些情况。然而，在 HLB 期间可能会发生肢体骨折，通常是由于操作员错误。减轻这种风险包括在轮床固定期间将手和脚向动物中线旋转。这种技术可以防止爆炸风向后扫动附属物并折断相关的骨骼。
这种重复的法学学士模式的优势不仅限于道德考虑，还延伸到实践和方法方面。其基于实验室的设计消除了处理爆炸物的需要，从而提高了安全性和可访问性。该模型具有高度可重复性和可定制性，允许研究人员通过使用不同的气体类型、设备设置和膜强度来影响暴露参数。这里选择氦是因为它能够再现旷场爆炸动力学49，可能提供可靠的基线 47,59,60。通过修改保留膜厚度或强度，凭经验调整峰值压力，从而能够根据特定的实验要求进行微调。最后，LLB 模型消除了季节性或天气变化对数据、动物暴露和其他实验因素的影响。这种一致性确保了稳健可靠的结果，使这种重复的 LLB 模型成为纵向和高度重复爆炸研究的宝贵工具。
了解与爆炸相关的神经创伤需要阐明损伤机制、爆炸强度指标和阈值。然而，爆炸情景中人脑损伤机制存在不确定性。以前提出的爆炸暴露后人体损伤的标准依赖于动物研究，但由于物种61 的标量标准不完整，将这些研究直接应用于人类具有挑战性。鉴于存在公认的标准62,63，基于动物体重的肺损伤量表是一个例外。然而，基于身体64,65 或大脑质量66 的拟议脑效应缩放定律忽略了已知和未知的解剖学差异，尤其是关于大脑内部和周围的保护结构。质量缩放预测体型较小的物种受伤风险更高，这与鸟类 67,68,69 和人类 70 的研究相矛盾。因此，制定准确的缩放定律需要对外部爆炸事件强度与跨物种内部大脑效应之间的关系进行实证理解。就 LLB 而言，对动物模型或人类的单次或慢性暴露知之甚少。因此，我们的方法可以催化在 LLB 强度范围内为未来缩放定律的发展提供信息所需的实证研究。
总之，这种基于实验室的冲击管模型代表了小鼠 LLB 暴露慢性影响研究的重大进展。通过结合对持续超压进行建模的程序，优先考虑爆炸后的恢复和监测，并突出相对于替代模型的独特优势，这种基于实验室的 LLB 模型可能为促进我们对与慢性 LLB 暴露相关的伤害的理解提供可靠且合乎道德的选择。

当个人或建筑物受到相对较低强度的爆炸力时，就会发生低水平爆炸 （LLB） 暴露，这通常是由小型工业事故、受控拆除或某些军事训练活动引起的。相比之下，高级别爆炸 （HLB） 暴露需要暴露于强烈且具有潜在破坏性的爆炸力，这在军事战斗、恐怖袭击或大规模意外爆炸中很常见。因此，LLB 和 HLB 之间的主要区别在于爆炸事件的强度，以及引申开来，暴露人员在遭受身体或功能伤害之前忍受反复暴露的能力。在这方面，HLB 暴露的影响往往比 LLB 暴露的影响更明显。正因为如此，大量接触 LLB 的人可能更容易发生缓慢发生的伤害或缺陷，这些损伤或缺陷在它们的累积影响变得明显之前不会被发现。
正在进行的研究旨在增强我们对爆炸暴露特性（例如强度或重复性）如何导致伤害的理解，以便我们更好地指导预防和医疗管理。在军事医学中，了解爆炸暴露的临床意义至关重要，因此，需要能够告知这些结果的动物模型。尽管动物模型有助于阐明 HLB 的影响，但 LLB 暴露的影响在很大程度上仍未得到充分研究。许多建模研究检查了接近或高于 10 磅/平方英寸 （psi） 峰值压力 1,2,3,4,5,6,7,8,9,10,11,12,13,14,15， 16,17,18，但很少有报告关注 1 到 7 psi 的压力水平 19,20,21,22,23,24,25,26,27,28,29,30,31， 32,33,34,35,36，这在军事训练环境中更为常见 37,38,39,40，并且接近 4 psi 的历史阈值，以实现安全环境暴露。因此，更广泛地传播 LLB 常用峰值压力研究方法可能有助于促进快速临床见解，以应用于军事医学和力量优化。
从军事 LLB 的流行病学调查中可以看出 LLB 的职业风险与各种临床诊断之间的显着关联 41,42,43,44。这些研究支持定义不明确的剂量依赖性关系，重复的 LLB 暴露表明风险增加41。这表明，增加累积爆炸暴露在塑造军事环境中的临床结果方面起着至关重要的作用。
以前对 10 psi 以下的 LLB 进行的动物建模研究主要使用炸药或减震管系统来研究暴露的影响。尽管这些模型通常检查一到三次暴露的影响，但它们仍然有助于加深对机制 19,20,30,31、神经病理学 29,31,33 和行为后果 19,20,23,25,32,34 的理解，与军事训练环境中典型的低强度爆炸暴露有关。
检查由开场炸药产生的单个 LLB 的研究报告了细微的脑部病变和行为变化的证据，这些变化通常与创伤后压力有关。Woods 及其同事24 无法在 2.5-5.5 psi 下检测到微观脑损伤，但他们确实通过质谱法检测到了脑组织鞘糖脂的定量变化。使用相同的峰值压力和实验设计，Rubovitch 及其同事25 观察到了爆炸后的行为变化，当通过光学显微镜测量时，这些爆炸发生在类似的缺乏脑部病变的情况下。然而，在随后的病理学研究中，通过电子显微镜 29,30,31,32,33 在 6.7 psi LLB 暴露的小鼠中确定了对脑髓鞘、线粒体、神经元和神经血管系统的明确超微结构损伤。有趣的是，几项使用压力为 ~10 psi 及以下的开场炸药的 LLB 研究报告称，单次暴露后死亡率约为 3-8%25,36。

A Mouse Model of Fatigue Induced by Peripheral Irradiation

Cancer-related fatigue (CRF) is a distressing and costly condition that often affects patients receiving cancer treatments, including radiation therapy. Here we describe a method using targeted peripheral irradiation to induce fatigue-like behavior in mice. With appropriate shielding, the irradiation targets the lower abdominal/pelvic region of the mouse, sparing the brain, in an effort to model radiation treatment received by individuals with pelvic cancers. We deliver an irradiation dose that is sufficient to induce fatigue-like behavior in mice, measured by voluntary wheel-running activity (VWRA), without causing obvious morbidity. Since wheel running is a normal, voluntary behavior in mice, its use should have little confounding effect on other behavioral tests or biological measures. Hence, wheel running can be used as a feasible outcome measure in understanding the behavioral and biological correlates of fatigue. CRF is a complex condition with frequent comorbidities, and likely has causes related both to cancer and its various treatments. The methods described in this paper are useful for investigating radiation-induced changes that contribute to the development of CRF and, more generally, to explore the biological networks that can explain the development and persistence of a peripherally-triggered but centrally-driven behavior like fatigue.

Video-oculography in Mice

Eye movements are very important in order to track an object or to stabilize an image on the retina during movement. Animals without a fovea, such as the mouse, have a limited capacity to lock their eyes onto a target. In contrast to these target directed eye movements, compensatory ocular eye movements are easily elicited in afoveate animals1,2,3,4. Compensatory ocular movements are generated by processing vestibular and optokinetic information into a command signal that will drive the eye muscles. The processing of the vestibular and optokinetic information can be investigated separately and together, allowing the specification of a deficit in the oculomotor system. The oculomotor system can be tested by evoking an optokinetic reflex (OKR), vestibulo-ocular reflex (VOR) or a visually-enhanced vestibulo-ocular reflex (VVOR). The OKR is a reflex movement that compensates for "full-field" image movements on the retina, whereas the VOR is a reflex eye movement that compensates head movements. The VVOR is a reflex eye movement that uses both vestibular as well as optokinetic information to make the appropriate compensation. The cerebellum monitors and is able to adjust these compensatory eye movements. Therefore, oculography is a very powerful tool to investigate brain-behavior relationship under normal as well as under pathological conditions (f.e. of vestibular, ocular and/or cerebellar origin).
Testing the oculomotor system, as a behavioral paradigm, is interesting for several reasons. First, the oculomotor system is a well understood neural system5. Second, the oculomotor system is relative simple6; the amount of possible eye movement is limited by its ball-in-socket architecture ("single joint") and the three pairs of extra-ocular muscles7. Third, the behavioral output and sensory input can easily be measured, which makes this a highly accessible system for quantitative analysis8. Many behavioral tests lack this high level of quantitative power. And finally, both performance as well as plasticity of the oculomotor system can be tested, allowing research on learning and memory processes9.
Genetically modified mice are nowadays widely available and they form an important source for the exploration of brain functions at various levels10. In addition, they can be used as models to mimic human diseases. Applying oculography on normal, pharmacologically-treated or genetically modified mice is a powerful research tool to explore the underlying physiology of motor behaviors under normal and pathological conditions. Here, we describe how to measure video-oculography in mice8.

Video-oculography is a very quantitative method to investigate ocular motor performance as well as motor learning. Here, we describe how to measure video-oculography in mice. Establishment of a Robust and Reproducible Model of Radiation-Induced Skin and Muscle Fibrosis

Radiation-induced skin fibrosis (RISF) can result from a plethora of scenarios including cancer therapy, accidental exposure, or acts of terrorism. Radioactive beams can penetrate through the skin and affect the structures in their path including skin, muscles, and internal organs. Skin is the first structure to get exposed to radiation and is susceptible to develop chronic fibrosis, which is challenging to treat. Currently, limited treatment options show moderate efficacy in mitigating radiation-related skin fibrosis. A key factor hindering the development of effective countermeasures is the absence of a convenient and robust model that could allow for translation of the experimental findings to humans. Here, a robust and reproducible murine hind limb skin fibrosis model has been established for prophylactic and therapeutic evaluation of possible agents for functional and molecular recovery.
The right hind limb was irradiated using a single dose of 40 (Gray) Gy to induce skin fibrosis. Subjects developed edema and dermatitis in the early stages proceeded by visible skin constriction. Irradiated limbs showed a significantly reduced limb range of motion in the following weeks. In late stages, acute side effects subsided, yet chronic fibrosis persisted. A gait index was performed as an additional functional assay, which demonstrated the development of functional impairment. These non-invasive methods demonstrated reliable measurements for tracing fibrosis progression, which is supported by histological analyses. The radiation dose, application, and post-irradiation analyses employed in this model offer a vigorous and reproducible method for studying radiation-induced skin fibrosis and testing the efficacy of therapeutical agents.

Modeling Astrocytoma Pathogenesis In Vitro and In Vivo Using Cortical Astrocytes or Neural Stem Cells from Conditional, Genetically Engineered Mice

Current astrocytoma models are limited in their ability to define the roles of oncogenic mutations in specific brain cell types during disease pathogenesis and their utility for preclinical drug development. In order to design a better model system for these applications, phenotypically wild-type cortical astrocytes and neural stem cells (NSC) from conditional, genetically engineered mice (GEM) that harbor various combinations of floxed oncogenic alleles were harvested and grown in culture. Genetic recombination was induced in vitro using adenoviral Cre-mediated recombination, resulting in expression of mutated oncogenes and deletion of tumor suppressor genes. The phenotypic consequences of these mutations were defined by measuring proliferation, transformation, and drug response in vitro. Orthotopic allograft models, whereby transformed cells are stereotactically injected into the brains of immune-competent, syngeneic littermates, were developed to define the role of oncogenic mutations and cell type on tumorigenesis in vivo. Unlike most established human glioblastoma cell line xenografts, injection of transformed GEM-derived cortical astrocytes into the brains of immune-competent littermates produced astrocytomas, including the most aggressive subtype, glioblastoma, that recapitulated the histopathological hallmarks of human astrocytomas, including diffuse invasion of normal brain parenchyma. Bioluminescence imaging of orthotopic allografts from transformed astrocytes engineered to express luciferase was utilized to monitor in vivo tumor growth over time. Thus, astrocytoma models using astrocytes and NSC harvested from GEM with conditional oncogenic alleles provide an integrated system to study the genetics and cell biology of astrocytoma pathogenesis in vitro and in vivo and may be useful in preclinical drug development for these devastating diseases.

A Simple and Inexpensive Method for Determining Cold Sensitivity and Adaptation in Mice

Cold hypersensitivity is a serious clinical problem, affecting a broad subset of patients and causing significant decreases in quality of life. The cold plantar assay allows the objective and inexpensive assessment of cold sensitivity in mice, and can quantify both analgesia and hypersensitivity. Mice are acclimated on a glass plate, and a compressed dry ice pellet is held against the glass surface underneath the hindpaw. The latency to withdrawal from the cooling glass is used as a measure of cold sensitivity.
Cold sensation is also important for survival in regions with seasonal temperature shifts, and in order to maintain sensitivity animals must be able to adjust their thermal response thresholds to match the ambient temperature. The Cold Plantar Assay (CPA) also allows the study of adaptation to changes in ambient temperature by testing the cold sensitivity of mice at temperatures ranging from 30 &#176;C to 5 &#176;C. Mice are acclimated as described above, but the glass plate is cooled to the desired starting temperature using aluminum boxes (or aluminum foil packets) filled with hot water, wet ice, or dry ice. The temperature of the plate is measured at the center using a filament T-type thermocouple probe. Once the plate has reached the desired starting temperature, the animals are tested as described above.
This assay allows testing of mice at temperatures ranging from innocuous to noxious. The CPA yields unambiguous and consistent behavioral responses in uninjured mice and can be used to quantify both hypersensitivity and analgesia. This protocol describes how to use the CPA to measure cold hypersensitivity, analgesia, and adaptation in mice.

Acute skin toxicities from ionizing radiation (IR) are a common side effect from therapeutic courses of external beam radiation therapy (RT) and negatively impact patient quality of life and long term survival. Advances in the understanding of the biological pathways associated with normal tissue toxicities have allowed for the development of interventional drugs, however, current response studies are limited by a lack of quantitative metrics for assessing the severity of skin reactions. Here we present a diffuse optical spectroscopic (DOS) approach that provides quantitative optical biomarkers of skin response to radiation. We describe the instrumentation design of the DOS system as well as the inversion algorithm for extracting the optical parameters. Finally, to demonstrate clinical utility, we present representative data from a pre-clinical mouse model of radiation induced erythema and compare the results with a commonly employed visual scoring. The described DOS method offers an objective, high through-put evaluation of skin toxicity via functional response that is translatable to the clinical setting.

We present a diffuse optical spectroscopic (DOS) approach that provides quantitative optical biomarkers of skin response to radiation. We describe DOS instrumentation design, optical parameters extraction algorithms and the animal handling procedures required to yield representative data from a pre-clinical mouse model of radiation induced erythema.

Generation of a Chronic Obstructive Pulmonary Disease Model in Mice by Repeated Ozone Exposure

Chronic obstructive pulmonary disease (COPD) is characterized by persistent airflow limitation and lung parenchymal destruction. It has a very high incidence in aging populations. The current conventional therapies for COPD focus mainly on symptom-modifying drugs; thus, the development of new therapies is urgently needed. Qualified animal models of COPD could help to characterize the underlying mechanisms and can be used for new drug screening. Current COPD models, such as lipopolysaccharide (LPS) or the porcine pancreatic elastase (PPE)-induced emphysema model, generate COPD-like lesions in the lungs and airways but do not otherwise resemble the pathogenesis of human COPD. A cigarette smoke (CS)-induced model remains one of the most popular because it not only simulates COPD-like lesions in the respiratory system, but it is also based on one of the main hazardous materials that causes COPD in humans. However, the time-consuming and labor-intensive aspects of the CS-induced model dramatically limit its application in new drug screening. In this study, we successfully generated a new COPD model by exposing mice to high levels of ozone. This model demonstrated the following: 1) decreased forced expiratory volume 25, 50, and 75/forced vital capacity (FEV25/FVC, FEV50/FVC, and FEV75/FVC), indicating the deterioration of lung function; 2) enlarged lung alveoli, with lung parenchymal destruction; 3) reduced fatigue time and distance; and 4) increased inflammation. Taken together, these data demonstrate that the ozone exposure (OE) model is a reliable animal model that is similar to humans because ozone overexposure is one of the etiological factors of COPD. Additionally, it only took 6 - 8 weeks, based on our previous work, to create an OE model, whereas it requires 3 - 12 months to induce the cigarette smoke model, indicating that the OE model might be a good choice for COPD research.

Low-intensity Blast Wave Model for Preclinical Assessment of Closed-head Mild Traumatic Brain Injury in Rodents

Traumatic brain injury (TBI) is a large-scale public health problem. Mild TBI is the most prevalent form of neurotrauma and accounts for a large number of medical visits in the United States. There are currently no FDA-approved treatments available for TBI. The increased incidence of military-related, blast-induced TBI further accentuates the urgent need for effective TBI treatments. Therefore, new preclinical TBI animal models that recapitulate aspects of human blast-related TBI will greatly advance the research efforts into the neurobiological and pathophysiological processes underlying mild to moderate TBI as well as the development of novel therapeutic strategies for TBI.
Here we present a reliable, reproducible model for the investigation of the molecular, cellular, and behavioral effects of mild to moderate blast-induced TBI. We describe a step-by-step protocol for closed-head, blast-induced mild TBI in rodents using a bench-top setup consisting of a gas-driven shock tube equipped with piezoelectric pressure sensors to ensure consistent test conditions. The benefits of the setup that we have established are its relative low-cost, ease of installation, ease of use and high-throughput capacity. Further advantages of this non-invasive TBI model include the scalability of the blast peak overpressure and the generation of controlled reproducible outcomes. The reproducibility and relevance of this TBI model has been evaluated in a number of downstream applications, including neurobiological, neuropathological, neurophysiological and behavioral analyses, supporting the use of this model for the characterization of processes underlying the etiology of mild to moderate TBI.

Measuring DNA Damage and Repair in Mouse Splenocytes After Chronic In Vivo Exposure to Very Low Doses of Beta- and Gamma-Radiation

Low dose radiation exposure may produce a variety of biological effects that are different in quantity and quality from the effects produced by high radiation doses. Addressing questions related to environmental, occupational and public health safety in a proper and scientifically justified manner heavily relies on the ability to accurately measure the biological effects of low dose pollutants, such as ionizing radiation and chemical substances. DNA damage and repair are the most important early indicators of health risks due to their potential long term consequences, such as cancer. Here we describe a protocol to study the effect of chronic in vivo exposure to low doses of &#947;- and &#946;-radiation on DNA damage and repair in mouse spleen cells. Using a commonly accepted marker of DNA double-strand breaks, phosphorylated histone H2AX called &#947;H2AX, we demonstrate how it can be used to evaluate not only the levels of DNA damage, but also changes in the DNA repair capacity potentially produced by low dose in vivo exposures. Flow cytometry allows fast, accurate and reliable measurement of immunofluorescently labeled &#947;H2AX in a large number of samples. DNA double-strand break repair can be evaluated by exposing extracted splenocytes to a challenging dose of 2 Gy to produce a sufficient number of DNA breaks to trigger repair and by measuring the induced (1 hr post-irradiation) and residual DNA damage (24 hrs post-irradiation). Residual DNA damage would be indicative of incomplete repair and the risk of long-term genomic instability and cancer. Combined with other assays and end-points that can easily be measured in such in vivo studies (e.g., chromosomal aberrations, micronuclei frequencies in bone marrow reticulocytes, gene expression, etc.), this approach allows an accurate and contextual evaluation of the biological effects of low level stressors.
