Liposome capillary electrophoresis (LCE) was used as a model for the in vitro evaluation of the degree of membrane penetration of organic compounds. The hydrophobic parameters of 9 compounds were determined by LCE. In comparison of the correlation coefficient of log P(lw) (the logarithm of the partition coefficient of liposome/water system, calculated from retention factor) vs. log P(m) (the logarithm of osmotic coefficient) with that of log P(ow) (the logarithm of the partition coefficient of octanol/water system) vs. log P(m), the former was 0.94, much greater than the latter 0.78. It means that as the biomembrane model, liposome is more similar than the octanol/water system. The LCE method has the potential to be an in vitro screening model.

Fezf2 (also known as Fezl, ZNF312, or Zfp312) is an evolutionarily conserved forebrain-specific zinc finger transcription factor that is expressed during development and is implicated in patterning as well as neurogenesis in both zebrafish and mice. Despite these findings, the expression of fezf2 in the adult brain has not been well characterized, and fezf2 function in the adult brain remains unknown. The zebrafish has recently emerged as a new model system to study adult neurogenesis, given its similarity to mammalian systems and enhanced capability of undergoing adult neurogenesis. Through RNA in situ hybridization and using a fezf2 promoter-driven GFP transgenic line, we present data showing that fezf2 is expressed in radial glial progenitor cells of the telencephalic ventricular zone in the adult zebrafish brain, which co-express markers of neural stem cells and proliferation. Additionally, we identify the preoptic region and the hypothalamus as fezf2-expressing neurogenic regions in the adult zebrafish brain, where fezf2 labels progenitor cells as well as postmitotic neurons. Our findings establish Fezf2 as a novel marker for adult telencephalic ventricular progenitor cells that express markers of neural stem cells in zebrafish and lay a critical foundation for future investigation of Fezf2 function in the maintenance and differentiation of neural stem cells in the adult vertebrate brain.

Fezf2 (tambi&#xE9;n conocido como Fezl, ZNF312, o Zfp312) es un evolutivamente conservado prosenc&#xE9;falo espec&#xED;fica dedos de zinc factor de transcripci&#xF3;n que se expresa durante el desarrollo y est&#xE1; implicada en la morfog&#xE9;nesis, as&#xED; como la neurog&#xE9;nesis en tanto pez cebra y ratones. D&#xE9;couvrir la logique de cis-r&#xE9;gulation des exhausteurs de d&#xE9;veloppement est essentielle pour comprendre le r&#xF4;le de l&#x26;#39;ADN non codant dans le d&#xE9;veloppement. Cependant, il est lourd &#xE0; identifier des motifs fonctionnels au sein des activateurs et exhausteurs de vert&#xE9;br&#xE9;s donc peu ont leurs motifs fonctionnels de base a r&#xE9;v&#xE9;l&#xE9;. Nous rapportons ici une approche exp&#xE9;rimentale et de calcul pour d&#xE9;couvrir des motifs r&#xE9;glementaires en exhausteurs de d&#xE9;veloppement. Faisant usage de la base de donn&#xE9;es l&#x26;#39;expression des g&#xE8;nes du poisson z&#xE8;bre, nous avons identifi&#xE9; informatiquement conserv&#xE9; non des &#xE9;l&#xE9;ments de codage (CNE) susceptibles d&#x26;#39;avoir un choix de sp&#xE9;cificit&#xE9; tissulaire bas&#xE9;e sur l&#x26;#39;expression des g&#xE8;nes voisins. Gr&#xE2;ce &#xE0; un d&#xE9;bit &#xE9;lev&#xE9; et robuste dosage enhancer, nous avons test&#xE9; l&#x26;#39;activit&#xE9; d&#x26;#39;environ 100 CNE et efficacement ces d&#xE9;couvertes exhausteurs de d&#xE9;veloppement avec des profils d&#x26;#39;expression souhait&#xE9;s spatiales et temporelles dans le cerveau du poisson z&#xE8;bre. Application des algorithmes de pr&#xE9;diction de novo motif sur un groupe d&#x26;#39;exhausteurs de prosenc&#xE9;phale identifi&#xE9; cinq mieux class&#xE9;s, les motifs qui ont toutes &#xE9;t&#xE9; valid&#xE9;es exp&#xE9;rimentalement comme critique pour prosenc&#xE9;phale rehausseur d&#x26;#39;activit&#xE9;. Uncovering the cis-regulatory logic of developmental enhancers is critical to understanding the role of non-coding DNA in development. However, it is cumbersome to identify functional motifs within enhancers, and thus few vertebrate enhancers have their core functional motifs revealed. Here we report a combined experimental and computational approach for discovering regulatory motifs in developmental enhancers. Making use of the zebrafish gene expression database, we computationally identified conserved non-coding elements (CNEs) likely to have a desired tissue-specificity based on the expression of nearby genes. Through a high throughput and robust enhancer assay, we tested the activity of approximately 100 such CNEs and efficiently uncovered developmental enhancers with desired spatial and temporal expression patterns in the zebrafish brain. Application of de novo motif prediction algorithms on a group of forebrain enhancers identified five top-ranked motifs, all of which were experimentally validated as critical for forebrain enhancer activity. These results demonstrate a systematic approach to discover important regulatory motifs in vertebrate developmental enhancers. Moreover, this dataset provides a useful resource for further dissection of vertebrate brain development and function.

Descubrir la l&#xF3;gica de cis-regulador de potenciadores del desarrollo es fundamental para comprender el papel de ADN no codificante en el desarrollo. Sin embargo, es engorroso para identificar motivos funcionales dentro de potenciadores, y potenciadores de vertebrados por lo tanto pocos tienen sus motivos funcionales b&#xE1;sicos revelado. Aqu&#xED; se presenta un enfoque combinado experimental y computacional para descubrir los motivos de reglamentaci&#xF3;n en potenciadores del desarrollo. Haciendo uso de la base de datos de expresi&#xF3;n gen&#xE9;tica del pez cebra, que computacionalmente identificados conserva los elementos no codificantes (CNES) que puedan tener un deseado especificidad tisular basada en la expresi&#xF3;n de genes cercanos. A trav&#xE9;s de un alto rendimiento y robusta ensayo de potenciador, hemos probado la actividad de aproximadamente 100 CNE tales y eficiente descubierto potenciadores de desarrollo deseados, con los patrones de expresi&#xF3;n espacial y temporal en el cerebro del pez cebra. Uno studio retrospettivo di indagare le caratteristiche cliniche e epidemiologiche di vertigine &egrave; stato effettuato su 187 pazienti con vertigini. Una storia clinica per ogni paziente &egrave; stata registrata proprio sull&#39;attacco, la frequenza e lo sviluppo di vertigo, la durata, intensit&agrave; e accompagnati sintomi tra cui i fattori di rischio per malattie cerebrovascolari, ecc. Tutti i pazienti sono stati sottoposti ad esame fisico con particolare attenzione ai sistemi neurologici e manovra di Dix-Hallpike, calcolate tomografia/computato tomografia-angiografia (CT/CTA) e scansione MRI sono state eseguite quando necessario. Maggioranza dei pazienti in questo studio ha sofferto con circolazione posteriore ischemia (59.89%) e la vertigine parossistica posizionale benigna (16,04%). Altre malattie che questi pazienti inclusi emicrania, malattia di Meniere (1,6%), improvvisa perdita dell&#39;udito (1,07%) neuritis vestibolare, sclerosi multipla, encefalite virale acuta, meningioma, nevrosi, vertigini post-traumatica, infarto miocardico acuto (lo 0,53%) e nevrosi (14.97%). Sembrava che in confronto a pazienti pi&ugrave; giovani della popolazione anziana rischia di essere pi&ugrave; suscettibili alla vertigine. Vertigo attacca i pazienti con varie malattie, che pre-dispose i pazienti affetti da questa malattia. Presentazione di vertigine pu&ograve; essere clinicamente diagnosticata nella maggior parte dei casi di pazienti affetti da ischemia circolazione posteriore.

A retrospective study to investigate the clinical epidemiological characteristics of vertigo was carried out on 187 patients with vertigo. A clinical history for each patient was recorded precisely about the attack, frequency, and development of vertigo, its duration, intensity, and the accompanied symptoms including the risk factors for cerebrovascular disease, etc. All the patients were subjected to physical examination with special attention to neurologic systems and Dix-Hallpike maneuver, computed tomography/computed tomography-angiography (CT/CTA) and MRI scan were performed when necessary. Majority of the patients in this study suffered with posterior circulation ischemia (59.89%) and benign paroxysmal positional vertigo (16.04%). Other ailments that affected these patients included migraine, Meniere's disease (1.6%), sudden hearing loss (1.07%), vestibular neuronitis, multiple sclerosis, acute viral encephalitis, meningioma, neurosis, posttraumatic vertigo, acute myocardial infarction (0.53%), and neurosis (14.97%). It appeared that in comparison to younger patients the elderly population is likely to be more susceptible to vertigo. Vertigo attacks patients with various diseases, which pre-dispose the patients to this disease. Presentation of vertigo can be clinically diagnosed in most cases of patients suffering from posterior circulation ischemia.

Une histoire clinique de chaque patient a &eacute;t&eacute; enregistr&eacute;e avec pr&eacute;cision sur l&#39;attaque, la fr&eacute;quence et d&eacute;veloppement du vertige, sa dur&eacute;e, intensit&eacute; et les sympt&ocirc;mes accompagn&eacute;s, y compris les facteurs de risque pour les maladies vasculaires c&eacute;r&eacute;brales, etc.. Inoltre, si conferma attraverso esperimenti di BrdU-etichettatura che la zona posteriore (Dp) &egrave; formata da migrazione radiale e omologhe della corteccia del piriform mammiferi. Basato sui nostri risultati, vi proponiamo un modello nuovo di organizzativo e di sviluppo di zebrafish pallio-uno che &egrave; il risultato di un complesso piegare verso l&#39;esterno.

El pez cebra como modelo experimental neurogen&#xE9;tica depende de la comprensi&#xF3;n correcta de su neuroanat&#xF3;mica la organizaci&#xF3;n del cerebro. En este sentido, abordar la cuesti&#xF3;n sin resolver en relaci&#xF3;n con un hom&#xF3;logo pez cebra posible de la divisi&#xF3;n dorsal paleal, la regi&#xF3;n que en los mam&#xED;feros da lugar a la isocorteza. Analizando la distribuci&#xF3;n de la nicotina adenina dinucle&#xF3;tido fosfato diphorase (NADPHd) parvalb&#xFA;mina actividad y en el telenc&#xE9;falo del pez cebra anterior, se demuestra que contra los supuestos anteriores, la central (DC) la zona posee su propia regi&#xF3;n germinativa en la zona proliferativa dorsal. Se define la central (DC) de zona como topol&#xF3;gicamente correspondiente a la divisi&#xF3;n dorsal paleal de otros vertebrados (mam&#xED;feros isocorteza). Adem&#xE1;s, confirmamos a trav&#xE9;s de experimentos BrdU-etiquetado que la parte posterior (DP) la zona est&#xE1; formada por la migraci&#xF3;n radial y hom&#xF3;loga a la de mam&#xED;feros corteza piriforme. En base a nuestros resultados, proponemos un nuevo modelo de desarrollo y de organizaci&#xF3;n del pez cebra, un palio que es el resultado de un complejo hacia el exterior hacia el interior de plegado.

Zebrafish as a neurogenetic model system depends on the correct neuroanatomical understanding of its brain organization. Here, we address the unresolved question regarding a possible zebrafish homologue of the dorsal pallial division, the region that in mammals gives rise to the isocortex. Analyzing the distributions of nicotine adenine dinucleotide phosphate diphorase (NADPHd) activity and parvalbumin in the anterior zebrafish telencephalon, we show that against previous assumptions the central (Dc) zone possesses its own germinative region in the dorsal proliferative zone. We define the central (Dc) zone as topologically corresponding to the dorsal pallial division of other vertebrates (mammalian isocortex). In addition, we confirm through BrdU-labeling experiments that the posterior (Dp) zone is formed by radial migration and homologous to the mammalian piriform cortex. Based on our results, we propose a new developmental and organizational model of the zebrafish pallium-one which is the result of a complex outward-inward folding.

Zebrafisch als neurogenetische Modellsystem h&auml;ngt von der richtigen neuroanatomischen Verst&auml;ndnis seiner Gehirn-Organisation. Hier wenden wir uns die ungel&ouml;ste Frage hinsichtlich einer m&ouml;glichen Zebrafisch-Homolog der dorsalen Pallialdr&uuml;sensekret Division, der Region, die bei S&auml;ugetieren die Isocortex begr&uuml;ndet. Analysieren die Verteilungen von Nikotin adenin-dinucleotid-phosphat Diphorase (NADPHd) Aktivit&auml;t und Parvalbumin in den vorderen Zebrafisch-Telencephalon, zeigen wir, dass gegen die bisherigen Annahmen die zentrale (Dc)-Zone eine eigene Keimschicht Region der dorsalen proliferative Zone besitzt. Wir definieren die zentrale Zone (Dc) als topologisch, der dorsalen Pallialdr&uuml;sensekret Division Produkte gegen sonstige Wirbeltiere (S&auml;ugetier-Isocortex) entspricht. Au&szlig;erdem best&auml;tigen wir durch BrdU-beschriften-Experimente, dass die hintere (Dp)-Zone durch radiale Migration geformt und Homologe, S&auml;ugetier-Piriform Rinde ist. Auf der Grundlage unserer Ergebnisse, schlagen wir ein neues Entwicklungs- und organisatorische Modell die Zebrafisch Pallium-eins, das Ergebnis einer komplexen nach au&szlig;en-nach innen zu Falten ist.

In this two-years surveillance of 2009 pandemic influenza A (H1N1) (pH1N1) in Guangzhou, China, we reported here that the scale and duration of pH1N1 outbreaks, severe disease and fatality rates of pH1N1 patients were significantly lower or shorter in the second epidemic year (May 2010-April 2011) than those in the first epidemic year (May 2009-April 2010) (P<0.05), but similar to those of seasonal influenza (P>0.05). Similar to seasonal influenza, pre-existing chronic pulmonary diseases was a risk factor associated with fatal cases of pH1N1 influenza. Different from seasonal influenza, which occurred in spring/summer seasons annually, pH1N1 influenza mainly occurred in autumn/winter seasons in the first epidemic year, but prolonged to winter/spring season in the second epidemic year. The information suggests a tendency that the epidemics of pH1N1 influenza may probably further shift to spring/summer seasons and become a predominant subtype of seasonal influenza in coming years in Guangzhou, China.

En China el efecto protector de la vacuna contra la influenza estacional s&oacute;lo ha sido evaluado en ensayos cl&iacute;nicos controlados y demostrado para ser altamente eficaz. Sin embargo, la efectividad de la forma de vacuna contra la influenza no se ha examinado. En nuestro estudio todos los casos de la gripe de los 19 sitios de vigilancia en Guangzhou fueron confirmado durante 2009 y 2010. Controles se seleccionaron al azar de los ni&ntilde;os de 6 a 59 meses en ampliado programado inmunizaci&oacute;n computarizado sistema infantil administrativo. Finalmente se inscribieron 2529 casos y 4539 controles. Despu&eacute;s de ajustar por sexo, edad y zona de residencia, la efectividad de la vacuna de vacunaci&oacute;n completa fue 51.79% y 57.78% en la temporada de gripe 2009 y 2010, respectivamente. Vacunaci&oacute;n parcial proporciona protecci&oacute;n 39.38% y 35.98% a ni&ntilde;os de 24 a 59 meses en 2009 y 2010, respectivamente, y ning&uacute;n efecto protector se observ&oacute; entre los ni&ntilde;os m&aacute;s peque&ntilde;os. In China the protective effect of seasonal influenza vaccine has only been assessed in controlled clinical trials and proven to be highly effective. However, the post-licensure effectiveness of influenza vaccine has not been examined. In our study all influenza cases from the 19 surveillance sites in Guangzhou were laboratory confirmed during 2009 and 2010. Controls were randomly selected from children aged 6 to 59 months in the Children's Expanded Programmed Immunization Administrative Computerized System. 2529 cases and 4539 controls were finally enrolled. After adjusting for gender, age and area of residence, the vaccine effectiveness of full vaccination was 51.79% and 57.78% in the 2009 and 2010 influenza season, respectively. Partial vaccination provided 39.38% and 35.98% protection to children aged 24 to 59 months in 2009 and 2010, respectively, and no protective effect was observed among younger children. Full vaccination is highly protective and partial vaccination is protective for older children. Influenza vaccination in general should be encouraged, and full vaccination should be particularly encouraged because its protective effect is much stronger than that of partial vaccination.

Dans notre &eacute;tude, tous les cas de grippe dans les 19 sites de surveillance &agrave; Guangzhou ont &eacute;t&eacute; confirm&eacute;s en 2009 et 2010 en laboratoire. Les contr&ocirc;les ont &eacute;t&eacute; choisis au hasard aupr&egrave;s d&#39;enfants &acirc;g&eacute;s de 6 &agrave; 59 mois dans &eacute;largi programm&eacute; administratif informatis&eacute; syst&egrave;me enfants d&#39;immunisation. 2529 cas et 4539 contr&ocirc;les &eacute;taient finalement inscrits. Apr&egrave;s ajustement pour le sexe, &acirc;ge et r&eacute;gion de r&eacute;sidence, l&#39;efficacit&eacute; du vaccin de la vaccination compl&egrave;te &eacute;tait de 51,79 % et 57,78 % durant la saison de grippe de 2009 et 2010, respectivement. Jedoch ist die post-licensure Wirksamkeit der Influenza-Impfstoff nicht &uuml;berpr&uuml;ft worden. In unserer Studie waren alle Grippe-F&auml;lle aus den 19 &Uuml;berwachung-Standorte in Guangzhou Labor w&auml;hrend 2009 und 2010 best&auml;tigt. Vollst&auml;ndige Impfung ist sehr sch&uuml;tzende und teilweise Impfung ist Schutzma&szlig;nahmen f&uuml;r &auml;ltere Kinder. Grippeimpfung im allgemeinen gef&ouml;rdert werden, und volle Impfung sollten besonders gef&ouml;rdert werden, weil seine sch&uuml;tzende Wirkung viel h&ouml;her als das der partiellen Impfung ist.
