&#x8FDB;&#x4E00;&#x6B65; Crinivirus&#xFF0C;&#x5C5E;&#x5185;&#x7684;&#x53D8;&#x5F02;&#x6027;&#xFF0C;&#x6240;&#x663E;&#x793A;&#x7684;&#x5B8C;&#x6574;&#x7684;&#x846B;&#x82A6;&#x4E1D;&#x9EC4;&#x77EE;&#x5316;&#x5931;&#x8C03;&#x75C5;&#x6BD2; RNA &#x57FA;&#x56E0;&#x7EC4;&#x5E8F;&#x5217;&#x7684;&#x6D4B;&#x5B9A;&#x3002;

The complete nucleotide (nt) sequences of genomic RNAs 1 and 2 of Cucurbit yellow stunting disorder virus (CYSDV) were determined for the Spanish isolate CYSDV-AlLM. RNA1 is 9123 nt long and contains at least five open reading frames (ORFs). Computer-assisted analyses identified papain-like protease, methyltransferase, RNA helicase and RNA-dependent RNA polymerase domains in the first two ORFs of RNA1. This is the first study on the sequences of RNA1 from CYSDV. RNA2 is 7976 nt long and contains the hallmark gene array of the family Closteroviridae, characterized by ORFs encoding a heat shock protein 70 homologue, a 59 kDa protein, the major coat protein and a divergent copy of the coat protein. This genome organization resembles that of Sweet potato chlorotic stunt virus (SPCSV), Cucumber yellows virus (CuYV) and Lettuce infectious yellows virus (LIYV), the other three criniviruses sequenced completely to date. However, several differences were observed. The most striking novel features of CYSDV compared to SPCSV, CuYV and LIYV are a unique gene arrangement in the 3'-terminal region of RNA1, the identification in this region of an ORF potentially encoding a protein which has no homologues in any databases, and the prediction of an unusually long 5' non-coding region in RNA2. Additionally, the CYSDV genome resembles that of SPCSV in having very similar 3' regions in RNAs 1 and 2, although for CYSDV similarity in primary structures did not result in predictions of equivalent secondary structures. Overall, these data reinforce the view that the genus Crinivirus contains considerable genetic variation. Additionally, several subgenomic RNAs (sgRNAs) were detected in CYSDV-infected plants, suggesting that generation of sgRNAs is a strategy used by CYSDV for the expression of internal ORFs.

Caract&eacute;ristiques nouvelles les plus frappantes de CYSDV contre SPCSV, CuYV et LIYV sont un arrangement de g&egrave;ne unique en 3&#39;-r&eacute;gion terminale du RNA1, l&#39;identification de cette r&eacute;gion un ORF potentiellement codant pour une prot&eacute;ine qui n&#39;a aucuns homologues dans des bases de donn&eacute;es et la pr&eacute;diction d&#39;une longue inhabituelle 5&#39; non codante r&eacute;gion RNA2. En outre, le g&eacute;nome CYSDV qui SPCSV ressemble &agrave; ayant des r&eacute;gions tr&egrave;s similaire de 3&#39; ARN 1 et 2, m&ecirc;me si pour CYSDV similitude de structure primaire n&#39;entra&icirc;ne pas de pr&eacute;diction de structures secondaires &eacute;quivalentes. Dans l&#39;ensemble, ces donn&eacute;es renforcent l&#39;opinion que le genre Crinivirus comporte une variation g&eacute;n&eacute;tique consid&eacute;rable. ABSTRACT The pattern of accumulation of Cucurbit yellow stunting disorder virus (CYSDV; genus Crinivirus, family Closteroviridae) RNA has been analyzed in several cucurbit accessions. In susceptible accessions of melon (Cucumis melo), cucumber (Cucumis sativus), marrow (Cucurbita maxima), and squash (Cucurbita pepo), CYSDV RNA accumulation peaked during the first to second week postinoculation in the first to third leaf above the inoculated one; younger leaves showed very low or undetectable levels of CYSDV. Three melon accessions previously shown to remain asymptomatic after CYSDV inoculation under natural conditions were also assayed for their susceptibility to CYSDV. Hybridization and reverse transcription-polymerase chain reaction (RT-PCR) analysis of noninoculated leaves showed that only one of these, C-105, remained virus-free for up to 6 weeks after whitefly inoculation. In this accession, very low CYSDV levels were detected by RT-PCR in whitefly-inoculated leaves, and therefore, multiplication or spread of CYSDV in C-105 plants appeared to remain restricted to the inoculated leaves. When C-105 plants were graft inoculated, CYSDV RNA could be detected in phloem tissues, but the systemic colonization of C-105 by CYSDV upon graft inoculation seemed to be seriously impeded. Additionally, in situ hybridization experiments showed that, after C-105 graft inoculation, only a portion of the vascular bundles in petioles and stems were colonized by CYSDV and virus could not be found in leaf veins. RT-PCR experiments using primers to specifically detect negative-sense CYSDV RNA were carried out and showed that CYSDV replication took place in graft-inoculated C-105 scions. Therefore, the resistance mechanism may involve a restriction of the virus movement in the vascular system of the plants and/or prevention of high levels of virus accumulation.

Accessions sensibles du melon (Cucumis melo), concombre (Cucumis sativus), moelle osseuse (Cucurbita maxima) et courges (Cucurbita pepo), accumulation de CYSDV RNA a atteint un sommet au cours de l&#39;apr&egrave;s l&#39;inoculation premi&egrave;re &agrave; la deuxi&egrave;me semaine dans la feuille de premier &agrave; troisi&egrave;me au-dessus de l&#39;inoculation ; jeunes feuilles pr&eacute;sentent des niveaux tr&egrave;s faibles ou ind&eacute;tectables de CYSDV. ABSTRAKTE Muster der Akkumulation von Cucurbit gelb stunting Disorder Virus (CYSDV; Gattung Crinivirus, Familie Closteroviridae) RNA in mehreren Cucurbit Beitritte analysiert wurde. Schaffte in anf&auml;lligen Beitritte von Melone (Cucumis Melo), Gurke (Cucumis Sativus), Knochenmark (Cucurbita Maxima) und Squash (Cucurbita Pepo) CYSDV RNA Anh&auml;ufung w&auml;hrend der erste bis zweite Woche Postinoculation im ersten bis dritten Blatt &uuml;ber dem beimpften; j&uuml;ngere Bl&auml;tter zeigten sehr geringe oder nicht nachweisbar Ebenen der CYSDV. Drei Melonen Beitritte nach CYSDV Inokulation unter nat&uuml;rlichen Bedingungen asymptomatisch bleiben zuvor gezeigt wurden auch f&uuml;r ihre Anf&auml;lligkeit f&uuml;r CYSDV normalisiert. Hybridisierung und reverse Transkription-Polymerase-Kettenreaktion (RT-PCR) Analyse der noninoculated Bl&auml;tter zeigte, dass nur eine von diesen C-105, f&uuml;r bis zu 6 Wochen nach der Inokulation Mottenschildl&auml;use blieb frei von Viren. In diesem Beitritt in Mottenschildl&auml;use verimpft Bl&auml;tter wurden sehr geringe CYSDV durch RT-PCR erkannt und daher Multiplikation oder Verbreitung von CYSDV in C-105 Pflanzen auf die beimpften Bl&auml;tter beschr&auml;nkt bleiben. Als C-105 Pflanzen Transplantat geimpft waren, CYSDV RNA in Phloem Geweben nachgewiesen werden konnte, aber die systemische Kolonisation des C-105 von CYSDV auf Transplantat Inokulation schien ernsthaft behindert werden. Dar&uuml;ber hinaus zeigten in situ Hybridisierung Experimente, dass nach der C-105 Transplantat Inokulation, nur ein Teil der vaskul&auml;re B&uuml;ndel in den Blattstielen und St&auml;ngeln von CYSDV besiedelt wurden und Virus nicht in Blatt-Adern gefunden. RT-PCR-Experimente mit Z&uuml;ndkapseln um negativ-Sinn CYSDV RNA spezifisch zu erreichen wurden durchgef&uuml;hrt und zeigten, dass CYSDV Replikation in Transplantat-verimpft C-105 Pfropfreiser stattfand. Der Widerstand-Mechanismus kann daher eine Beschr&auml;nkung des Kapitalverkehrs Virus im Gef&auml;&szlig;system der Pflanzen bzw. Vermeidung hoher Gehalte an Virus Anh&auml;ufung beinhalten.

Resumen el patr&oacute;n de acumulaci&oacute;n de cucurbit&aacute;ceas amarillo retraso en el crecimiento trastorno virus (CYSDV; g&eacute;nero Crinivirus, familia Closteroviridae) RNA ha sido analizado en varias adhesiones de cucurbit&aacute;ceas. En adhesiones susceptibles de mel&oacute;n (Cucumis melo), pepino (Cucumis sativus), m&eacute;dula (Cucurbita maxima) y calabaza (Cucurbita pepo), acumulaci&oacute;n de RNA CYSDV alcanz&oacute; el puesto durante el posinoculaci&oacute;n de primera a segunda semana en la hoja de primera a tercera sobre el inoculado; hojas j&oacute;venes mostraron niveles muy bajos o indetectables de CYSDV. Tambi&eacute;n se ensayaron tres adhesiones de mel&oacute;n demostrados previamente que siguen siendo asintom&aacute;ticos despu&eacute;s de la inoculaci&oacute;n de CYSDV bajo condiciones naturales para su susceptibilidad a CYSDV. Hibridaci&oacute;n y an&aacute;lisis de la reacci&oacute;n en cadena (RT-PCR) de reversa de la transcripci&oacute;n-polimerasa de hojas inoculados mostraron que s&oacute;lo uno de ellos, C-105, permaneci&oacute; libre de virus por hasta 6 semanas despu&eacute;s de la inoculaci&oacute;n de la mosca blanca. En este proceso de adhesi&oacute;n, se detectaron niveles muy bajos de CYSDV por RT-PCR en hojas inoculadas mosca blanca, y por lo tanto, multiplicaci&oacute;n o propagaci&oacute;n de CYSDV en C-105 plantas parec&iacute;a seguir siendo restringido a las hojas inoculadas. Cuando C-105 plantas fueron inoculada de injerto, CYSDV ARN podr&iacute;a detectarse en los tejidos del floema, pero la colonizaci&oacute;n sist&eacute;mica de C-105 por CYSDV sobre inoculaci&oacute;n de injerto parec&iacute;a verse obstaculizado seriamente. Adem&aacute;s, hibridaci&oacute;n in situ los experimentos demostraron que, despu&eacute;s de la inoculaci&oacute;n de injerto C-105, solamente una porci&oacute;n de los haces vasculares en pec&iacute;olos y tallos fueron colonizados por CYSDV y virus no se encuentra en las venas de la hoja. Experimentos de RT-PCR usando las cartillas para detectar espec&iacute;ficamente CYSDV ARN negativo fueron llevadas a cabo y demostr&oacute; que la replicaci&oacute;n CYSDV ocurri&oacute; en injerto inocular C-105 scions. Por lo tanto, el mecanismo de resistencia puede implicar una restricci&oacute;n de la circulaci&oacute;n del virus en el sistema vascular de las plantas y/o prevenci&oacute;n de altos niveles de acumulaci&oacute;n de virus.
