&#x6C2F;&#x4E59;&#x9178;&#x916F;&#x5BF9;&#x8C37;&#x80F1;&#x7518;&#x80BD;&#x4EE3;&#x8C22;&#x53CA;&#x5F71;&#x54CD;&#x661F;&#x5F62;&#x80F6;&#x8D28;&#x7EC6;&#x80DE;&#x7684;&#x7CD6;&#x9175;&#x89E3;&#x3002;

The chlorinated acetates monochloroacetate (MCA), dichloroacetate (DCA), and trichloroacetate (TCA) are generated in water disinfection processes and are formed during metabolic detoxification of industrial solvents such as trichloroethylene. In order to test for consequences of an exposure of brain cells to the different chlorinated acetates, glutathione levels and lactate production of primary astrocyte cultures were investigated as indicators for the potential of chlorinated acetates to disturb cellular detoxification processes and glucose metabolism, respectively. Application of MCA to cultured astrocytes caused a time and concentration dependent deprivation of cellular glutathione, inactivation of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) activity, and loss in cell viability with halfmaximal effects observed for MCA concentrations between 0.3 and 3Â mM. In contrast, the presence of acetate, DCA, or TCA in a concentration of 10Â mM did not compromise cell viability nor affect cellular glutathione content or GAPDH activity. However, the presence of DCA and TCA significantly lowered the rate of cellular lactate production in viable astrocytes. These data demonstrate that the extent of chlorination strongly determines the potential of chlorinated acetates to disturb glutathione and/or glucose metabolism of astrocytes.

Effects of chlorinated acetates on the glutathione metabolism and on glycolysis of cultured astrocytes.

Les ac&eacute;tates chlor&eacute;s monochloroac&eacute;tate (MCA), le dichloroac&eacute;tate (DCA) et trichloroac&eacute;tate (TCA) sont g&eacute;n&eacute;r&eacute;s dans le processus de d&eacute;sinfection de l&#39;eau et sont form&eacute;s au cours de la d&eacute;toxication m&eacute;tabolique des solvants industriels comme le trichlor&eacute;thyl&egrave;ne. Afin de tester des cons&eacute;quences d&#39;une exposition des cellules du cerveau pour les ac&eacute;tates chlor&eacute;s diff&eacute;rents, les niveaux de glutathion et de la production des cultures primaires d&#39;astrocytes de lactate a &eacute;t&eacute; &eacute;tudi&eacute;es comme indicateurs du potentiel d&#39;ac&eacute;tates chlor&eacute;s de perturber les processus de d&eacute;toxification cellulaire et le m&eacute;tabolisme du glucose, respectivement. Application de MCA d&#39;astrocytes cultiv&eacute;s pos&eacute;s un temps et la privation d&eacute;pendant de la concentration du glutathion cellulaire, l&#39;inactivation de la glyc&eacute;rald&eacute;hyde-3-phosphate d&eacute;shydrog&eacute;nase (GAPDH) et la perte dans la viabilit&eacute; des cellules halfmaximal des effets observ&eacute;s pour des concentrations de MCA entre 0,3 et 3 mM. En revanche, la pr&eacute;sence d&#39;ac&eacute;tate, DCA ou TCA dans une concentration de 10 mM n&#39;a pas compromettre la viabilit&eacute; des cellules ni affectent le contenu en glutathion cellulaire ou activit&eacute; GAPDH. Toutefois, la pr&eacute;sence de DCA et de TCA a significativement abaiss&eacute; le taux de production de lactate cellulaire dans les astrocytes viables. Ces r&eacute;sultats d&eacute;montrent que l&#39;&eacute;tendue de la chloration d&eacute;termine fortement le potentiel des ac&eacute;tates chlor&eacute;s &agrave; perturber le m&eacute;tabolisme de glutathion et/ou de glucose des astrocytes.

Die chlorierten Acetate Monochloroacetate (MCA), Dichloroacetate (DCA) und Trichloroacetate (TCA) werden in Wasser Desinfektionsprozesse generiert und w&auml;hrend metabolische Entgiftung des industriellen L&ouml;sungsmittel wie Trichlorethylen gebildet. Um die Auswirkungen der Exposition von Gehirnzellen, die verschiedene chlorierte Acetate testen, wurden Glutathion und Laktat-Produktion der prim&auml;ren Gliazelle Kulturen als Indikatoren f&uuml;r das Potenzial der chlorierten Acetate, zellul&auml;re Entgiftungsverfahren und Glukosestoffwechsel, bzw. st&ouml;ren untersucht. Anwendung der MCA auf kultivierte Astrozyten verursachte eine Zeit und Konzentration abh&auml;ngige Entzug der zellul&auml;ren Glutathion, Inaktivierung von Glycerinaldehyd-3-phosphat-Dehydrogenase (GAPDH) Aktivit&auml;t und Verlust Zellviabilit&auml;t mit Halfmaximal Effekten f&uuml;r die MCA-Konzentrationen zwischen 0,3 und 3 mM beobachtet. Im Gegensatz dazu das Vorhandensein von Acetat, DCA oder TCA in einer Konzentration von 10 mM nicht gef&auml;hrden Zellviabilit&auml;t noch zellul&auml;re Glutathion Inhalt oder GAPDH Aktivit&auml;t beeinflussen. Allerdings senkte das Vorhandensein von DCA und TCA deutlich die zellul&auml;re Laktat-Produktion in lebensf&auml;higen Astrozyten. Diese Daten zeigen, dass das Ausma&szlig; der Chlorung stark, das Potenzial von chlorierten Acetate bestimmt, Glutathion und/oder Glukose-Stoffwechsel der Astrozyten zu st&ouml;ren.

Il monocloroacetato acetati clorurati (MCA), dicloroacetato (DCA) e trichloroacetate (TCA) sono generate nei processi di disinfezione dell&#39;acqua e si formano durante la disintossicazione metabolica dei solventi industriali come tricloruro di etilene. Al fine di verificare le conseguenze di un&#39;esposizione delle cellule di cervello per i diversi acetati clorurati, i livelli di glutatione e la produzione di lattato di colture primarie di astrociti sono state studiate come indicatori per il potenziale di acetati clorurati a disturbare i processi di disintossicazione cellulare e il metabolismo del glucosio, rispettivamente. Applicazione di MCA di astrociti colti ha causato un tempo e privazione dipendente dalla concentrazione di glutatione cellulare, inattivazione della gliceraldeide-3-fosfato deidrogenasi (GAPDH) attivit&agrave; e perdita di attuabilit&agrave; delle cellule con effetti halfmaximal osservati per concentrazioni di MCA tra 0.3 e 3 mM. In contrasto, la presenza di acetato, DCA o TCA in una concentrazione di 10 mM non compromettere la vitalit&agrave; cellulare n&eacute; influenzare cellulare glutatione contenuto o attivit&agrave; GAPDH. Tuttavia, la presenza di DCA e TCA abbassato notevolmente il tasso di produzione di lattato cellulare negli astrociti vitali. Questi dati dimostrano che il grado di clorurazione determina fortemente il potenziale di acetati clorurati disturbare glutatione e/o glucosio metabolismo degli astrociti.

Los acetatos clorados Monocloroacetato (MCA), dicloroacetato (DCA) y tricloro acetato (TCA) se generan en procesos de desinfecci&oacute;n de agua y se forman durante la desintoxicaci&oacute;n metab&oacute;lica de solventes industriales como tricloroetileno. Para probar las consecuencias de una exposici&oacute;n de las c&eacute;lulas del cerebro a los acetatos clorados diferentes, niveles de glutati&oacute;n y la producci&oacute;n de lactato de culturas Astrocito primaria investigaron como indicadores del potencial de acetatos clorados perturbar los procesos de desintoxicaci&oacute;n celular y el metabolismo de la glucosa, respectivamente. Gustatory stimuli allow an organism not only to orient in its environment toward energy-rich food sources to maintain nutrition but also to avoid unpleasant or even poisonous substrates. For both mammals and insects, sugars-perceived as "sweet"-potentially predict nutritional benefit. Interestingly, even Drosophila adult flies are attracted to most high-potency sweeteners preferred by humans. However, the gustatory information of a sugar may be misleading as some sugars, although perceived as "sweet," cannot be metabolized. Accordingly, in adult Drosophila, a postingestive system that additionally evaluates the nutritional benefit of an ingested sugar has been shown to exist. By using a set of seven different sugars, which either offer (fructose, sucrose, glucose, maltodextrin, and sorbitol) or lack (xylose and arabinose) nutritional benefit, we show that Drosophila, at the larval stage, can perceive and evaluate sugars based on both nutrition-dependent and -independent qualities. In detail, we find that larval survival and feeding mainly depend on the nutritional value of a particular sugar. In contrast, larval choice behavior and learning are regulated in a more complex way by nutrition value-dependent and nutrition value-independent information. The simplicity of the larval neuronal circuits and their accessibility to genetic manipulation may ultimately allow one to identify the neuronal and molecular basis of the larval sugar perception systems described here behaviorally.

The Drosophila larva has turned into a particularly simple model system for studying the neuronal basis of innate behaviors and higher brain functions. Neuronal networks involved in olfaction, gustation, vision and learning and memory have been described during the last decade, often up to the single-cell level. Thus, most of these sensory networks are substantially defined, from the sensory level up to third-order neurons. This is especially true for the olfactory system of the larva. Given the wealth of genetic tools in Drosophila it is now possible to address the question how modulatory systems interfere with sensory systems and affect learning and memory. Here we focus on the serotonergic system that was shown to be involved in mammalian and insect sensory perception as well as learning and memory. Larval studies suggested that the serotonergic system is involved in the modulation of olfaction, feeding, vision and heart rate regulation. In a dual anatomical and behavioral approach we describe the basic anatomy of the larval serotonergic system, down to the single-cell level. In parallel, by expressing apoptosis-inducing genes during embryonic and larval development, we ablate most of the serotonergic neurons within the larval central nervous system. When testing these animals for naÃ¯ve odor, sugar, salt and light perception, no profound phenotype was detectable; even appetitive and aversive learning was normal. Our results provide the first comprehensive description of the neuronal network of the larval serotonergic system. Moreover, they suggest that serotonin per se is not necessary for any of the behaviors tested. However, our data do not exclude that this system may modulate or fine-tune a wide set of behaviors, similar to its reported function in other insect species or in mammals. Based on our observations and the availability of a wide variety of genetic tools, this issue can now be addressed.

La larva de Drosophila se ha convertido en un sistema modelo simple para estudiar la base neuronal de comportamientos naturales y funciones superiores del cerebro. Redes neuronales implicados en el olfato, percepci&oacute;n, visi&oacute;n y aprendizaje y la memoria se han descrito durante la &uacute;ltima d&eacute;cada, a menudo hasta el nivel unicelular. As&iacute;, la mayor&iacute;a de estas redes sensoriales se define sustancialmente, desde el nivel sensorial hasta las neuronas de tercer orden. Esto es especialmente cierto para el sistema olfativo de la larva. Dada la riqueza de herramientas gen&eacute;ticas en Drosophila ahora es posible para que aborden el asunto sistemas moduladores interfieran con sistemas sensoriales y afectan el aprendizaje y la memoria. Aqu&iacute; nos centramos en el sistema serotonin&eacute;rgico que mostr&oacute; ser involucrados en la memoria, as&iacute; como el aprendizaje y la percepci&oacute;n sensorial de mam&iacute;fera e insecto. Larvarios estudios sugirieron que el sistema serotonin&eacute;rgico est&aacute; implicado en la modulaci&oacute;n de olfato, alimentaci&oacute;n, regulaci&oacute;n de la visi&oacute;n y la frecuencia card&iacute;aca. En un doble enfoque anat&oacute;mico y de comportamiento se describe la anatom&iacute;a b&aacute;sica del sistema serotonin&eacute;rgico larval, hasta el nivel unicelular. En paralelo, expresando los genes que inducen apoptosis durante el desarrollo embrionario y larval, nos ablar de la mayor&iacute;a de las neuronas serotonin&eacute;rgicas dentro de la larva del sistema nervioso central. Al probar estos animales para olor de ingenuos, az&uacute;car, sal y luz de la percepci&oacute;n, no profunda fenotipo era perceptible; incluso apetitivo y aprendizaje aversivo era normal. Nuestros resultados proporcionan la primera descripci&oacute;n completa de la red neuronal del sistema serotonin&eacute;rgico larval. Adem&aacute;s, sugieren que la serotonina per no se es necesaria para cualquiera de las conductas probadas. Sin embargo, nuestros datos no excluyen que este sistema puede modular o ajustar un amplio conjunto de comportamientos, similares a su funci&oacute;n reportado en otras especies de insectos o mam&iacute;feros. Basado en nuestras observaciones y la disponibilidad de una amplia variedad de herramientas gen&eacute;ticas, este problema puede ahora ser abordado.
