Measurements of oxygen partial pressure (pO(2)) with high temporal and spatial resolution in three dimensions is crucial for understanding oxygen delivery and consumption in normal and diseased brain. Among existing pO(2) measurement methods, phosphorescence quenching is optimally suited for the task. However, previous attempts to couple phosphorescence with two-photon laser scanning microscopy have faced substantial difficulties because of extremely low two-photon absorption cross-sections of conventional phosphorescent probes. Here we report to our knowledge the first practical in vivo two-photon high-resolution pO(2) measurements in small rodents' cortical microvasculature and tissue, made possible by combining an optimized imaging system with a two-photon-enhanced phosphorescent nanoprobe. The method features a measurement depth of up to 250 microm, sub-second temporal resolution and requires low probe concentration. The properties of the probe allowed for direct high-resolution measurement of cortical extravascular (tissue) pO(2), opening many possibilities for functional metabolic brain studies.

Les mesures de la pression partielle d&#x26;#39;oxyg&#xE8;ne (pO (2)) avec une r&#xE9;solution temporelle et spatiale en trois dimensions est cruciale pour la compr&#xE9;hension de l&#x26;#39;apport d&#x26;#39;oxyg&#xE8;ne et la consommation dans le cerveau normal et pathologique. Parmi les m&#xE9;thodes existantes de mesure pO (2), trempe la phosphorescence est parfaitement adapt&#xE9; &#xE0; la t&#xE2;che. Proc&#xE9;d&#xE9; comporte une profondeur de mesure jusqu&#x26;#39;&#xE0; 250 microm&#xE8;tre, inf&#xE9;rieur &#xE0; la seconde r&#xE9;solution temporelle et n&#xE9;cessite concentration de la sonde basse. Les propri&#xE9;t&#xE9;s de la sonde a permis de directe &#xE0; haute r&#xE9;solution de mesure de extravasculaire corticale (tissus) pO (2), l&#x26;#39;ouverture de nombreuses possibilit&#xE9;s pour des &#xE9;tudes fonctionnelles du cerveau m&#xE9;taboliques.

Messung der Sauerstoff-Partialdruck (pO(2)) mit hoher zeitlicher und r&auml;umlicher Aufl&ouml;sung dreidimensional ist entscheidend f&uuml;r das Verst&auml;ndnis der Sauerstoffzufuhr und Verbrauch im normalen und kranken Gehirn. Unter den vorhandenen pO(2)-Messverfahren ist die Phosphoreszenz abschrecken optimal auf die Aufgabe. Fr&uuml;here Versuche paar Phosphoreszenz mit zwei-Photonen-Laser-scanning-Mikroskopie haben jedoch erhebliche Schwierigkeiten wegen extrem geringe zwei-Photonen-Absorption Querschnitte des konventionellen phosphoreszierende Sonden konfrontiert. Hier berichten wir unseres Wissens die ersten praktischen in-vivo pO(2) der zwei-Photonen-hochaufl&ouml;sende Messungen in kleine Nagetiere kortikale Microvasculature und Gewebe, erm&ouml;glicht durch ein optimiertes-imaging-System mit einer zwei-Photonen-verbesserte phosphoreszierendes Nanosonde kombinieren. Die Methode verf&uuml;gt &uuml;ber eine Messung Tiefe von bis zu 250 Microm, Schadensmeldungen zeitliche Aufl&ouml;sung und niedrigen Sonde Konzentration erfordert. Die Eigenschaften der Sonde f&uuml;r hochaufl&ouml;sende Direktmessung der kortikalen extravaskul&auml;ren (Gewebe) pO(2), viele M&ouml;glichkeiten f&uuml;r funktionale metabolische Gehirn Studien zul&auml;ssig.

Le misurazioni della pressione parziale dell&#39;ossigeno (pO(2)) con alta risoluzione spaziale e temporale in tre dimensioni &egrave; fondamentale per comprendere la fornitura di ossigeno e consumo nel cervello normale e malato. Tra i metodi di misurazione esistenti pO(2), fosforescenza tempra &egrave; ottimamente adatto per il compito. Tuttavia, precedenti tentativi di fosforescenza coppia con scansione microscopia del due-fotone laser hanno dovuto affrontare difficolt&agrave; notevoli a causa delle sezioni trasversali estremamente basso assorbimento del due-fotone di sonde convenzionali fosforescente. Qui riportiamo a nostra conoscenza le prime misure pratiche in vivo due-fotone ad alta risoluzione pO(2) in piccoli roditori microvasculature corticale e tessuto, reso possibile dalla combinazione di un sistema di imaging ottimizzato con un due-fotone-enhanced nanosonda fosforescente. Il metodo presenta una profondit&agrave; di misurazione di fino a 250 microm, risoluzione temporale subentrer&agrave; e richiede sonda bassa concentrazione. Le propriet&agrave; della sonda ha permesso per la misura diretta ad alta risoluzione della pO(2) corticale extravascolare (tessuto), aprire molte possibilit&agrave; per gli Studi metabolici funzionale del cervello.

Las mediciones de la presi&#xF3;n parcial de ox&#xED;geno (PO (2)) con una alta resoluci&#xF3;n temporal y espacial en tres dimensiones es crucial para comprender el aporte de ox&#xED;geno y el consumo en el cerebro normal y enfermo. Entre los actuales m&#xE9;todos de medici&#xF3;n pO (2), apagar la fosforescencia es &#xF3;ptimo para la tarea. Sin embargo, los intentos previos a la fosforescencia pareja con microscop&#xED;a de escaneo l&#xE1;ser de dos fotones se han enfrentado a grandes dificultades debido a la muy baja absorci&#xF3;n de dos fotones secciones transversales de las sondas convencionales fosforescentes. Aqu&#xED; mostramos a nuestro conocimiento de la pr&#xE1;ctica por primera vez en vivo de dos fotones de alta resoluci&#xF3;n PO (2) las medidas en la microvasculatura de los peque&#xF1;os roedores y el tejido cortical, que fue posible mediante la combinaci&#xF3;n de un sistema de imagen optimizada con un Nanoprobe fosforescente de dos fotones mejorada. El m&#xE9;todo cuenta con una medici&#xF3;n de profundidad de hasta 250 micras, menos de un segundo la resoluci&#xF3;n temporal y requiere de concentraci&#xF3;n de la sonda de baja.
