In most organisms, low ethanol doses induce increased activity, while high doses are sedating. To investigate the underlying mechanisms, we isolated Drosophila mutants with altered ethanol responsiveness. Mutations in white rabbit (whir), disrupting RhoGAP18B, are strongly resistant to the sedating effects of ethanol. This resistance can be suppressed by reducing the levels of Rho1 or Rac, implicating these GTPases in the behavioral response to ethanol. Indeed, expression of constitutively active forms of Rho1 or Rac1 in adult flies results in ethanol resistance similar to that observed in whir mutants. Drosophila melanogaster is commonly found near rotting or fermenting fruit, reflected in its name pomace, or vinegar fly. In such environments, flies often encounter significant levels of ethanol. Three observations have made Drosophila a very promising model organism to understand the genetic contributions to the behavioral responses to alcohol. First, similar to higher vertebrates, flies show hyperactivation upon exposure to a low to medium dose of alcohol, while high doses can lead to sedation. In addition, when given a choice, flies will actually prefer alcohol-containing food over regular food. Second, the genes and biochemical pathways implicated in controlling these behavioral responses in flies are also participating in determining alcohol responses, and drinking behavior in mammals. Third, the fact that flies have been studied genetically for over one hundred years means that an exceptional repertoire of genetic tools are at our disposal. Here, we will review some of these tools and experimental approaches, survey the methods for, and measures after Drosophila ethanol exposure, and discuss the different molecular components and functional pathways involved in these behavioral responses to alcohol.

Recently, the fruit fly Drosophila melanogaster has been introduced as a model system to study the molecular bases of a variety of ethanol-induced behaviors. It became immediately apparent that the behavioral changes elicited by acute ethanol exposure are remarkably similar in flies and mammals. Flies show signs of acute intoxication, which range from locomotor stimulation at low doses to complete sedation at higher doses and they develop tolerance upon intermittent ethanol exposure. Genetic screens for mutants with altered responsiveness to ethanol have been carried out and a few of the disrupted genes have been identified. This analysis, while still in its early stages, has already revealed some surprising molecular parallels with mammals. The availability of powerful tools for genetic manipulation in Drosophila, together with the high degree of conservation at the genomic level, make Drosophila a promising model organism to study the mechanism by which ethanol regulates behavior and the mechanisms underlying the organism's adaptation to long-term ethanol exposure.

R&#xE9;cemment, la mouche Drosophila melanogaster a &#xE9;t&#xE9; pr&#xE9;sent&#xE9; comme un syst&#xE8;me mod&#xE8;le pour &#xE9;tudier les bases mol&#xE9;culaires d&#x26;#39;une vari&#xE9;t&#xE9; de comportements induits par l&#x26;#39;&#xE9;thanol. Il est devenu imm&#xE9;diatement &#xE9;vident que les changements de comportement suscit&#xE9;s par l&#x26;#39;exposition aigu&#xEB; d&#x26;#39;&#xE9;thanol sont remarquablement similaires chez les mouches et les mammif&#xE8;res. Vor kurzem wurde der Fruchtfliege Drosophila Melanogaster eingef&uuml;hrt, als Modellsystem, die molekularen Grundlagen von einer Vielzahl von &Auml;thanol-induzierte Verhalten zu studieren. Es wurde sofort klar, dass die Verhaltens&auml;nderungen, hervorgerufen durch akute &Auml;thanol Exposition fliegen und S&auml;ugetieren bemerkenswert &auml;hnlich sind. Fliegen zeigen Anzeichen f&uuml;r eine akute Intoxikation, die im Bereich von lokomotorischen Anregung in niedriger Dosierung Sedierung bei h&ouml;heren Dosen abgeschlossen und entwickeln Toleranz bei intermittierender &Auml;thanol-Exposition. Genetische Schirme f&uuml;r Mutanten mit ver&auml;nderter Reaktionsf&auml;higkeit zu Ethanol durchgef&uuml;hrt worden und ein paar der gest&ouml;rte Gene identifiziert wurden. Diese Analyse ergab noch in einem fr&uuml;hen Stadium bereits einige &uuml;berraschenden molekularen &Auml;hnlichkeiten mit S&auml;ugetieren. Die Verf&uuml;gbarkeit von leistungsstarken Werkzeugen f&uuml;r Genmanipulation in Drosophila, zusammen mit der hohen Erhaltung auf genomischer Ebene stellen Drosophila vielversprechende Modellorganismus den Mechanismus durch welche &Auml;thanol zu studieren Verhalten und die Mechanismen des Organismus Anpassung an &Auml;thanol Langzeitbelastung regelt.

Rab GTPases are considered master regulators of membrane trafficking in all cells, and only very few Rabs have known neuron-specific functions. Here, we present the first systematic characterization of neuronal expression, subcellular localization, and function of Rab GTPases in an organism with a brain.
