Continuous 12-lead electrocardiographic (ECG) monitoring can identify transient myocardial ischemia, even when asymptomatic, among patients with suspected acute coronary syndrome (ACS). In this article we describe our method for initiating patient monitoring using a Holter device, downloading the ECG data for off-line analysis, and how to utilize the ECG software to identify transient ischemia.

Rearing Ixodes scapularis, the Black-legged Tick: Feeding Immature Stages on Mice
Andrew B. Nuss 1, Manoj G. Mathew 2, Monika Gulia-Nuss 2
1Department of Agriculture, Nutrition, and Veterinary Sciences, University of Nevada, Reno, 2Department of Biochemistry and Molecular Biology, University of Nevada, Reno

The production of healthy laboratory-reared ticks is essential to studies on tick biology, and tick-pathogen interactions. Here we demonstrate a simple protocol for immature tick feeding that is cost-effective and less stressful to mice.

Methods for Presenting Real-world Objects Under Controlled Laboratory Conditions
Carissa A. Romero 1, Jacqueline C. Snow 1
1Department of Psychology, University of Nevada, Reno

We describe methods for presenting real-world objects and matched images of the same objects under tightly-controlled experimental conditions. The methods are described in the context of a decision-making task, but the same real-world approach can be extended to other cognitive domains such as perception, attention, and memory.

Bacterial Expression and Purification of Human Matrix Metalloproteinase-3 using Affinity Chromatography
Alexander J. Bolt *1, Linh D. Do *1, Maryam Raeeszadeh-Sarmazdeh 1
1Department of Chemical & Materials Engineering, University of Nevada, Reno

His-tag purification, dialysis, and activation are employed to increase yields of soluble, active matrix metalloproteinase-3 catalytic domain protein expression in bacteria. Protein fractions are analyzed via SDS-PAGE gels.

Embryo Injection Technique for Gene Editing in the Black-Legged Tick, Ixodes scapularis
Arvind Sharma 1, Michael Pham 1, Robert A. Harrell II 2, Andrew B. Nuss 3, Monika Gulia-Nuss 1
1Department of Biochemistry and Molecular Biology, University of Nevada, Reno, 2Insect Transformation Facility, University of Maryland Institute for Bioscience and Biotechnology Research, 3Department of Agriculture, Veterinary, and Rangeland Sciences, University of Nevada, Reno

The present protocol describes a method for injecting tick embryos. Embryo injection is the preferred technique for genetic manipulation to generate transgenic lines.

Measurement of Poly A Tail Length from Drosophila Larva Brain and Cell Line
Monika Singh 1, Jung Hwan Kim 1
1Department of Biology, University of Nevada, Reno

The protocol describes an efficient and reliable method for quantifying the poly(A) length of the gene of interest from the Drosophila nervous system, which can be easily adapted to tissues or cell types from other species.
