A functional assessment of the neuromuscular junction (NMJ) can provide essential information on the communication between muscle and nerve. Here we describe a protocol to comprehensively evaluate both the NMJ and muscle functionality using two different muscle-nerve preparations, i.e. soleus-sciatic and diaphragm-phrenic.

A Novel In Vitro Wound Healing Assay to Evaluate Cell Migration
Floriana Cappiello 1, Bruno Casciaro 1, Maria Luisa Mangoni 1
1Department of Biochemical Sciences, Sapienza University of Rome

Here, we present a protocol to evaluate the effect of peptides on the migration of bronchial epithelial cells. This method allows for the rapid and highly reproducible obtainment of quantitative data on the speed of cell migration and wound closure.

Blastocyst biopsy and vitrification are required to efficiently conduct preimplantation genetic testing. An approach entailing the sequential opening of the zona pellucida and retrieval of 7-8 trophectoderm cells in day 5-7 post-insemination limits both the number of manipulations required and the exposure of the embryo to sub-optimal environmental conditions.

Phase Change Dimethyldioctadecylammonium-Shelled Microdroplets as a Promising Drug Delivery System: Results on 3D Spheroids of Mammalian Tumor Cells
Elisabetta Tortorella *1,2, Damiano Palmieri *1, Martina Piermarini 1, Daniele Gigante 1, Letizia Oddo 1, Yosra Toumia 1, Gaio Paradossi 1, Fabio Domenici 1
1Department of Chemical Sciences and Technologies, University of Rome Tor Vergata, 2Department of Molecular Medicine, Sapienza University of Rome

Decafluoropentane microdroplets developed with a shell of dimethyldioctadecylammonium bromide exhibited an exceptional colloidal stability and an actractive biointerface. DDAB-MDs proved to be efficient drug reservoirs characterized by a high affinity to plasma membranes together with enhanced uptake and antitumor activity of Doxorubicin against human triple-negative breast cancer (MDA-MB-231) 3D model.
