The etiology of idiopathic pulmonary fibrosis (IPF) is unknown. Because viral pathogenesis of IPF has been suggested, we have established a murine model of progressive pulmonary fibrosis by infecting IFN-gammaR-deficient mice (IFN-gammaR(-/-)) with the murine gamma-herpesvirus 68. Because alveolar macrophages in humans with IPF have been implicated in driving the profibrotic response, we studied their role in our model. Chronic herpesvirus infection of the lung was associated with recruitment of alveolar macrophages to areas with epithelial hyperplasia and fibrosis in infected lungs. Using immunohistochemistry, Western blot, and RT-PCR techniques, we demonstrated that recruited alveolar macrophages showed high levels of expression of the proteins Ym1/2, FIZZ1 (found in inflammatory zone 1), insulin-like growth factor-1, and arginase I, and also active transcription of fibronectin, indicative of activation of macrophages by an alternative pathway. Arginase I expression was also evident in interstitial fibroblasts, and increased arginase activity was found in lungs of infected animals. Lung tissue from patients with IPF showed increased expression of arginase I in epithelial cells, fibroblast foci, and alveolar macrophages compared with normal lung. These results suggest that virus-induced upregulation of arginase I could be a mechanism driving lung fibrogenesis.

Activation of alveolar macrophages via the alternative pathway in herpesvirus-induced lung fibrosis.

L&#x26;#39;&#xE9;tiologie de la fibrose pulmonaire idiopathique (IPF) est inconnu. Parce que la pathogen&#xE8;se virale de l&#x26;#39;IPF a &#xE9;t&#xE9; sugg&#xE9;r&#xE9;, nous avons &#xE9;tabli un mod&#xE8;le murin de fibrose pulmonaire progressive en infectant l&#x26;#39;IFN-Gammar souris d&#xE9;ficientes (IFN-Gammar (- / -)) avec le gamma-herp&#xE8;svirus murin 68. Parce que les macrophages alv&#xE9;olaires chez l&#x26;#39;homme avec l&#x26;#39;IPF ont &#xE9;t&#xE9; impliqu&#xE9;s dans la conduite de la r&#xE9;ponse profibrotique, nous avons &#xE9;tudi&#xE9; leur r&#xF4;le dans notre mod&#xE8;le. L&#x26;#39;infection chronique de l&#x26;#39;herp&#xE8;svirus du poumon a &#xE9;t&#xE9; associ&#xE9;e avec le recrutement des macrophages alv&#xE9;olaires dans les zones avec une hyperplasie &#xE9;pith&#xE9;liale et la fibrose dans les poumons infect&#xE9;s. Utilisation de l&#x26;#39;immunohistochimie, western blot et RT-PCR, nous avons d&#xE9;montr&#xE9; que les macrophages alv&#xE9;olaires recrut&#xE9;s ont montr&#xE9; des niveaux &#xE9;lev&#xE9;s d&#x26;#39;expression des prot&#xE9;ines YM1 / 2, FIZZ1 (trouv&#xE9; dans inflammatoire zone 1), l&#x26;#39;insuline-like growth factor-1, et l&#x26;#39;arginase I , et aussi la transcription active de la fibronectine, indicative de l&#x26;#39;activation des macrophages par une voie alternative. Arginase je expression &#xE9;tait &#xE9;galement &#xE9;vident dans les fibroblastes interstitiels, et augmentation de l&#x26;#39;activit&#xE9; arginase a &#xE9;t&#xE9; trouv&#xE9; dans les poumons des animaux infect&#xE9;s. Le tissu pulmonaire des patients atteints de FPI a montr&#xE9; une expression accrue de l&#x26;#39;arginase I dans les cellules &#xE9;pith&#xE9;liales, les fibroblastes, des foyers et les macrophages alv&#xE9;olaires par rapport &#xE0; un poumon normal. Ces r&#xE9;sultats sugg&#xE8;rent que la r&#xE9;gulation positive induite par le virus de l&#x26;#39;arginase je pourrais &#xEA;tre un m&#xE9;canisme d&#x26;#39;entra&#xEE;nement fibrogen&#xE8;se pulmonaire.

Activation des macrophages alv&#xE9;olaires par la voie alternative de la fibrose pulmonaire induite par l&#x26;#39;herp&#xE8;svirus

Die &Auml;tiologie der idiopathischen Lungenfibrose (IPF) ist unbekannt. Da virale Pathogenese der IPF vorgeschlagen wurde, haben wir einem murinen Modell der progressive Lungenfibrose etabliert, durch infizieren von IFN-GammaR-defizienten M&auml;usen (IFN-gammaR(-/-)) mit murinen Gamma-Herpesvirus 68. Da alveol&auml;re Makrophagen bei Menschen mit IPF beim Fahren der Profibrotic Antwort verwickelt haben, studierte wir ihre Rolle in unserem Modell. Chronische Herpesvirus-Infektion der Lunge war Rekrutierung der alveol&auml;ren Makrophagen in Gebiete mit epitheliale Hyperplasie und Fibrose in infizierten Lunge zugeordnet. Mittels Immunhistochemie, Westernblot und RT-PCR-Techniken, wir bewiesen, dass rekrutierte alveol&auml;re Makrophagen hohe Expression der Proteine Ym1/2, FIZZ1 (zu finden im entz&uuml;ndlichen Zone 1) und Insulin-Like Growth Factor 1 Arginase zeigte ich, und auch aktive Transkription von Fibronektin, indikative der Aktivierung der Makrophagen durch einen alternativen Weg. Arginase ich Ausdruck zeigte sich auch in interstitielle Fibroblasten und erh&ouml;hte Arginase Aktivit&auml;t wurde in den Lungen von infizierten Tieren gefunden. Lungengewebe von Patienten mit IPF zeigte erh&ouml;hte Expression von Arginase, die ich in Epithelzellen, Fibroblasten-Foki und alveol&auml;ren Makrophagen mit normalen Lunge verglichen. Diese Ergebnisse legen nahe, dass Viren induzierte Hochregulation von Arginase k&ouml;nnte ich einen Mechanismus Lunge Fibrogenesis fahren.

L&#39;eziologia della fibrosi polmonare idiopatica (FPI) &egrave; sconosciuta. Perch&eacute; virale patogenesi della IPF sono stato suggerito, abbiamo stabilito un modello murino di fibrosi polmonare progressiva infettando topi IFN-gammaR-carenti (IFN-gammaR(-/-)) con la murino gamma-herpesvirus 68. Perch&eacute; macrofagi alveolari in esseri umani con IPF sono stati implicati nella guida la risposta profibrotic, abbiamo studiato il loro ruolo nel nostro modello. Infezione da herpesvirus cronica del polmone &egrave; stato associato con assunzione di macrofagi alveolari a zone con iperplasia epiteliale e fibrosi nei polmoni infetti. Mediante immunoistochimica, Western blot e le tecniche di RT-PCR, abbiamo dimostrato che ha reclutati macrofagi alveolari hanno mostrato alti livelli di espressione delle proteine Ym1/2, FIZZ1 (che si trova in zona infiammatoria 1), insulin-like growth factor-1 e arginase I e inoltre attiva trascrizione di fibronectina, indicativa di attivazione dei macrofagi da un percorso alternativo. Arginase era evidente nei fibroblasti interstiziali anche espressione e arginase maggiore attivit&agrave; &egrave; stato trovato nei polmoni degli animali infetti. Tessuto polmonare di pazienti con IPF ha mostrato maggiore espressione di arginase in cellule epiteliali, foci dei fibroblasti e macrofagi alveolari rispetto ai normale del polmone. Questi risultati suggeriscono che upregulation virus-indotta di arginase che potrei essere un meccanismo di guida fibrogenesis del polmone.

Debido a que los macr&#xF3;fagos alveolares en los seres humanos con IPF han sido implicados en la conducci&#xF3;n de la respuesta profibr&#xF3;tico, hemos estudiado su papel en nuestro modelo. La infecci&#xF3;n por herpesvirus cr&#xF3;nica del pulm&#xF3;n se asoci&#xF3; con el reclutamiento de macr&#xF3;fagos alveolares a las zonas con hiperplasia epitelial y fibrosis en los pulmones infectados. El uso de inmunohistoqu&#xED;mica, Western blot, y las t&#xE9;cnicas de RT-PCR, hemos demostrado que reclutaron a los macr&#xF3;fagos alveolares mostraron altos niveles de expresi&#xF3;n de las prote&#xED;nas YM1 / 2, FIZZ1 inflamatoria (se encuentra en zona 1), de crecimiento insul&#xED;nico tipo 1 del factor, y la arginasa I , y tambi&#xE9;n la transcripci&#xF3;n activa de la fibronectina, indicativo de la activaci&#xF3;n de los macr&#xF3;fagos por una v&#xED;a alternativa. Arginasa expresi&#xF3;n que tambi&#xE9;n fue evidente en los fibroblastos intersticiales, y la actividad de la arginasa mayor se encontr&#xF3; en los pulmones de los animales infectados. El tejido pulmonar de pacientes con FPI mostraron una mayor expresi&#xF3;n de arginasa I en las c&#xE9;lulas epiteliales, fibroblastos, los focos y los macr&#xF3;fagos alveolares de pulm&#xF3;n en comparaci&#xF3;n con lo normal. Several lines of evidence indicate that depletion of glutathione (GSH), a critical thiol antioxidant, is associated with the pathogenesis of idiopathic pulmonary fibrosis (IPF). However, GSH synthesis depends on the amino acid cysteine (Cys), and relatively little is known about the regulation of Cys in fibrosis. Cys and its disulfide, cystine (CySS), constitute the most abundant low-molecular weight thiol/disulfide redox couple in the plasma, and the Cys/CySS redox state (E(h) Cys/CySS) is oxidized in association with age and smoking, known risk factors for IPF. Furthermore, oxidized E(h) Cys/CySS in the culture media of lung fibroblasts stimulates proliferation and expression of transitional matrix components. The present study was undertaken to determine whether bleomycin-induced lung fibrosis is associated with a decrease in Cys and/or an oxidation of the Cys/CySS redox state and to determine whether these changes were associated with changes in E(h) GSH/glutathione disulfide (GSSG). We observed distinct effects on plasma GSH and Cys redox systems during the progression of bleomycin-induced lung injury. Plasma E(h) GSH/GSSG was selectively oxidized during the proinflammatory phase, whereas oxidation of E(h) Cys/CySS occurred at the fibrotic phase. In the epithelial lining fluid, oxidation of E(h) Cys/CySS was due to decreased food intake. Thus the data show that decreased precursor availability and enhanced oxidation of Cys each contribute to the oxidation of extracellular Cys/CySS redox state in bleomycin-induced lung fibrosis.

Mehrere Zeilen Beweise zeigen, dass die Dezimierung von Glutathion (GSH), eine kritische Thiol-Antioxidans, die Pathogenese der idiopathischen Lungenfibrose (IPF) zugeordnet ist. Jedoch GSH-Synthese h&auml;ngt von der Aminos&auml;ure Cystein (Cys) und ist relativ wenig bekannt &uuml;ber die Regulierung der Cys bei Fibrose. CYS und seine Disulfid Cystin (CySS), bilden das h&auml;ufigste niedermolekularen Gewicht-Thiol/Disulfid-Redox-Paar im Plasma und der Cys/CySS Redox-Zustand (E(h) Cys/CySS) oxidiert in Verbindung mit dem Alter und Rauchen, Risikofaktoren f&uuml;r IPF bekannt ist. Dar&uuml;ber hinaus regt oxidierte E(h) Cys/CySS in den Kultur-Medien der Lunge Fibroblasten Proliferation und Ausdruck des &Uuml;bergangs Matrix-Komponenten. Die vorliegende Studie wurde durchgef&uuml;hrt, um festzustellen, ob die Bleomycin-induzierte Lungenfibrose einen R&uuml;ckgang der Cys und/oder eine Oxidation des Staates Redox Cys/CySS zugeordnet ist, und um festzustellen, ob diese &Auml;nderungen &Auml;nderungen in E(h) GSH/Glutathion-Disulfid (GSSG) zugeordnet wurden. Plasma E(h) GSH/GSSG wurde w&auml;hrend der Phase Proinflammatory selektiv oxidiert, w&auml;hrend Oxydation des E(h) Cys/CySS bei der fibrotische Phase aufgetreten ist. In der Fl&uuml;ssigkeit epitheliale Auskleidung war Oxydation des E(h) Cys/CySS durch verminderte Nahrungsaufnahme. Somit die Daten-Show, die Vorl&auml;ufer-Verf&uuml;gbarkeit und erweiterte Oxidation von Cys jedes sanken um zur Oxidation von extrazellul&auml;ren Cys/CySS Redox-Zustand in Bleomycin-induzierte Lungenfibrose beitragen.

Several studies have implicated viral infection as an important factor in the pathogenesis of IPF and related fibrotic lung disorders. Viruses are thought to cause epithelial cell injury and promote epithelial-mesenchymal transition (EMT), a process whereby differentiated epithelial cells undergo transition to a mesenchymal phenotype, and considered a source of fibroblasts in the setting of lung injury. We have demonstrated an association between the epithelial injury caused by chronic herpes virus infection with the murine gamma-herpes virus, MHV68, and lung fibrosis. We hypothesize that EMT in this model of virus-induced pulmonary fibrosis is driven by the expression of the transcription factor Twist.

Idiopathic pulmonary fibrosis (IPF) is a chronic progressive lung disorder of unknown etiology. Several studies have demonstrated an association between pulmonary infection with a herpesvirus and IPF. Based on those observations, we have developed a mouse model in which interferon (IFN)gammaR(-/-) mice infected intranasally with murine gammaherpesvirus 68 (MHV68) develop lung fibrosis. We hypothesize that viral load was a critical factor for the development of fibrosis. Because nuclear factor (NF)-kappaB signaling is required to efficiently establish gammaherpesvirus, latency we infected IFNgammaR(-/-) mice with a MHV68 virus that expresses a mutant dominant inhibitor of the NF-kappaB signaling pathway, called IkappaBalphaM. Striking differences were observed at the onset of the chronic infection, which correlated with a decreased virus load in mice infected with MHV68-IkappaBalphaM compared with mice infected with control MHV68 (MHV68-MR). IFNgammaR(-/-) mice infected with MHV68-IkappaBalphaM lacked vasculitis and fibrosis 15 to 120 days post infection. Inhibition of NF-kappaB in MHV68-infected cells of the lungs diminished the expression of the fibrocyte recruiting chemokines monocyte chemoattractant protein 1 (MCP-1) and CXCL12, ameliorated macrophage expression of markers of alternative activation, and failed to increase expression of the integrin alphavbeta6, which is implicated in the activation of the profibrotic factor TGF-beta. Thus, the inhibition of NF-kappaB signaling in the infected lung cells of IFNgammaR(-/-) mice reduces virus persistence and ameliorates profibrotic events. Host determinants of latency might therefore represent new therapeutic targets for gammaherpesvirus-associated pulmonary fibrosis.

Inhibition of NF-kappaB signaling reduces virus load and gammaherpesvirus-induced pulmonary fibrosis.

La fibrose pulmonaire idiopathique (IPF) est un trouble pulmonaire chronique progressive d&#x26;#39;&#xE9;tiologie inconnue. Plusieurs &#xE9;tudes ont d&#xE9;montr&#xE9; une association entre l&#x26;#39;infection pulmonaire avec un virus de l&#x26;#39;herp&#xE8;s et l&#x26;#39;IPF. Sur la base de ces observations, nous avons d&#xE9;velopp&#xE9; un mod&#xE8;le de souris dans laquelle l&#x26;#39;interf&#xE9;ron (IFN) Gammar (- / -) des souris infect&#xE9;es par voie intranasale avec gammaherpesvirus murin 68 (MHV68) d&#xE9;veloppent une fibrose du poumon. Nous &#xE9;mettons l&#x26;#39;hypoth&#xE8;se que la charge virale &#xE9;tait un facteur critique pour le d&#xE9;veloppement de la fibrose. Parce que le facteur nucl&#xE9;aire (NF)-kappaB est n&#xE9;cessaire pour &#xE9;tablir efficacement gammaherpesvirus, nous avons infect&#xE9; des temps de latence IFNgammaR (- / -) des souris avec un virus MHV68 qui exprime un inhibiteur mutant dominant de la voie de signalisation NF-kappa B, appel&#xE9; IkappaBalphaM. Hemmung der NF-KappaB in MHV68-infizierten Zellen der Lunge vermindert den Ausdruck des Fibrozyt Rekrutierung Chemokine Monozyte Chemoattractant Proteins 1 (MCP-1) und CXCL12, Makrophagen Ausdruck von Markern der alternative Aktivierung verbessert und Fehler beim Ausdruck der Integrin-alphavbeta6 zu erh&ouml;hen, was bei der Aktivierung des Faktors Profibrotic TGF-Beta verwickelt ist. Also die Hemmung der NF-KappaB signalisieren in den infizierten Lungenzellen IFNgammaR(-/-) M&auml;use reduziert Virus Persistenz und aerztlichen Profibrotic Ereignisse. Host Determinanten der Latenz k&ouml;nnen daher neue therapeutische Ziele f&uuml;r Gammaherpesvirus-assoziierte Lungenfibrose darstellen.

Diversi studi hanno dimostrato un&#39;associazione tra infezione polmonare con un herpesvirus e IPF. Sulla base di queste osservazioni, abbiamo sviluppato un modello murino in cui l&#39;interferone (IFN)gammaR(-/-) topi infettati intranasale con murino gammaherpesvirus 68 (MHV68) sviluppano fibrosi polmonare. La fibrosis pulmonar idiop&#xE1;tica (FPI) es una enfermedad cr&#xF3;nica pulmonar progresiva, de etiolog&#xED;a desconocida. Varios estudios han demostrado una asociaci&#xF3;n entre la infecci&#xF3;n pulmonar con un herpesvirus y el IPF. Con base en estas observaciones, hemos desarrollado un modelo de rat&#xF3;n en el que el interfer&#xF3;n (IFN) Gammar (- / -) ratones infectados por v&#xED;a intranasal con gammaherpesvirus murino 68 (MHV68) desarrollar fibrosis pulmonar. Nuestra hip&#xF3;tesis es que la carga viral es un factor cr&#xED;tico para el desarrollo de la fibrosis. La inhibici&#xF3;n de NF-kappaB en las c&#xE9;lulas infectadas por MHV68 de los pulmones disminuye la expresi&#xF3;n de las quimiocinas fibrocito reclutamiento de monocitos chemoattractant prote&#xED;na 1 (MCP-1) y CXCL12, la expresi&#xF3;n de los macr&#xF3;fagos mejorado de los marcadores de activaci&#xF3;n alternativa, y no para aumentar la expresi&#xF3;n de la integrina alphavbeta6, que est&#xE1; implicado en la activaci&#xF3;n del factor de profibr&#xF3;tico TGF-beta. Por lo tanto, la inhibici&#xF3;n de NF-kappaB en las c&#xE9;lulas de los pulmones infectados de IFNgammaR (- / -) ratones reduce la persistencia del virus y mejora eventos profibr&#xF3;ticos. Bone marrow-derived mesenchymal stem cells (BMDMSC) are emerging as a therapeutic modality in various inflammatory disease states, including acute lung injury (ALI). A hallmark of inflammation, and a consistent observation in patients with ALI, is a perturbation in the systemic redox environment. However, little is known about the effects of BMDMSC on the systemic redox status. The objective of the present study was to determine whether exogenously infused BMDMSC protect against endotoxin-induced oxidation of plasma cysteine (Cys) and glutathione (GSH) redox states. To determine the effect on the redox state if BMDMSC, mice received endotoxin intraperitoneally (1â€‰mg/kg), followed by intravenous infusion of either 5 Ã— 10(5) BMDMSC or an equal volume of saline solution. Control mice received intraperitoneal endotoxin followed by 5 Ã— 10(5) lung fibroblasts given intravenously. Cys, cystine (CySS), GSH, and glutathione disulfide (GSSG) concentrations were determined by HPLC. Results showed sequential preservation of plasma Cys and GSH levels in response to BMDMSC infusion. The data show that BMDMSC infusion leads to a more reducing Cys and GSH redox state. The findings are the first to demonstrate that BMDMSC have antioxidant effects in vivo, and add to our understanding of the systemic effects of BMDMSC in lung injury.

The incidence of Idiopathic pulmonary fibrosis (IPF) increases with age. The mechanisms that underlie the age-dependent risk for IPF are unknown. Based on studies that suggest an association of IPF and gammaherpesvirus infection, we infected young (2-3 mo) and old (â‰¥18 mo) C57BL/6 mice with the murine gammaherpesvirus 68 (MHV68). Acute MHV68 infection in aging mice resulted in severe pneumonitis and fibrosis compared with young animals. Progressive clinical deterioration and lung fibrosis in the late chronic phase of infection was observed exclusively in old mice with diminution of tidal volume. Infected aging mice showed higher expression of TGF-Î² during the acute phase of infection. In addition, aging infected mice showed elevation of pro-inflammatory cytokines and the fibrocyte recruitment chemokine CXCL12 in bronchoalveolar lavage. Analyses of lytic virus infection and virus reactivation indicate that old mice were able to control chronic infection and elicit anti-virus immune responses. However, old infected mice showed a significant increase in apoptotic responses determined by in situ TUNEL assay, levels of caspase-3 and expression of the pro-apoptotitc molecule BIM. Apoptosis of type II lung epithelial cells in aging lungs was accompanied by upregulation of ER stress marker BiP and splicing of XBP1. These results indicate that the aging lung is more susceptible to injury and fibrosis associated with ER stress, apoptosis of type II lung epithelial cells and activation of pro-fibrotic pathways.

Role of Endoplasmic Reticulum Stress in Age-Related Susceptibility to Lung Fibrosis.

L&#x26;#39;incidence de la fibrose pulmonaire idiopathique (IPF) augmente avec l&#x26;#39;&#xE2;ge. Les m&#xE9;canismes qui sous-tendent le risque d&#xE9;pend de l&#x26;#39;&#xE2;ge pour l&#x26;#39;IPF sont inconnus. Bas&#xE9; sur des &#xE9;tudes qui sugg&#xE8;rent une association de l&#x26;#39;infection IPF et gammaherpesvirus, nous avons infect&#xE9; des jeunes (2-3 mois) et &#xE2;g&#xE9;s (&#x2265; 18 mois) chez les souris C57BL / 6 avec le gammaherpesvirus murin 68 (MHV68). Aigu&#xEB; MHV68 infection chez les souris a entra&#xEE;n&#xE9; le vieillissement et la fibrose s&#xE9;v&#xE8;re pneumonie par rapport aux animaux jeunes. Basierend auf Studien, die eine Vereinigung von IPF und Gammaherpesvirus Infektion empfehlen, infizierten wir jung (2-3 Mo) und alt (18 Mo) C57BL/6 M&auml;usen mit dem murinen Gammaherpesvirus 68 (MHV68). Akute Infektion der MHV68 in alternden M&auml;usen f&uuml;hrte zu schweren Pneumonie und Fibrose im Vergleich mit Jungtieren. Progressive klinische Verschlechterung und Lunge Fibrose in der sp&auml;ten chronischen Phase der Infektion wurde ausschlie&szlig;lich in alten M&auml;use mit Diminution Tidalvolumen beobachtet. Infizierte alternden M&auml;usen zeigte h&ouml;heren Ausdruck von TGF-&beta; w&auml;hrend der akuten Phase der Infektion. Au&szlig;erdem infiziert alternde M&auml;use zeigte Erh&ouml;hung von Pro-inflammatorische Zytokine und der Fibrozyt Rekrutierung Chemokin CXCL12 in Bronchoalveol&auml;re Lavage. Analysen der lytische Infektion und Virus-Reaktivierung zeigen, dass die alten M&auml;use konnten chronischen Infektion zu kontrollieren und Anti-Virus-Immunantworten zu entlocken. Diese Ergebnisse zeigen, dass die Alterung Lunge anf&auml;lliger f&uuml;r Verletzungen und Fibrose ER Stress, Apoptose des Typs II Lunge Epithelzellen und Aktivierung der Pro-fibrotische Bahnen zugeordnet ist.

L&#39;incidenza di fibrosi polmonare idiopatica (FPI) aumenta con l&#39;et&agrave;. I meccanismi che sottendono il rischio et&agrave;-dipendente per IPF sono sconosciuti. Sulla base di studi che suggeriscono un&#39;associazione tra infezione IPF e gammaherpesvirus siamo infettati giovani (2-3 mo) e vecchi (&ge;18 mo) C57BL/6 topi con la murino gammaherpesvirus 68 (MHV68). Infezione acuta MHV68 in topi di invecchiamento provocato grave polmonite e fibrosi rispetto ad animali giovani. La incidencia de la fibrosis pulmonar idiop&#xE1;tica (IPF) aumenta con la edad. Los mecanismos que subyacen en el riesgo dependiente de la edad para la FPI es desconocida. Con base en estudios que sugieren una asociaci&#xF3;n de la infecci&#xF3;n por el GIB y el gammaherpesvirus, estamos infectados joven (2-3 meses) y adultos (&#x2265; 18 meses) en ratones C57BL / 6 con la gammaherpesvirus murino 68 (MHV68). MHV68 aguda infecci&#xF3;n en los ratones del envejecimiento como resultado neumonitis severa y fibrosis en comparaci&#xF3;n con los animales j&#xF3;venes. Deterioro cl&#xED;nico progresivo y la fibrosis pulmonar en la fase tard&#xED;a de la infecci&#xF3;n cr&#xF3;nica se observ&#xF3; exclusivamente en ratones viejos con disminuci&#xF3;n del volumen corriente. Ratones infectados envejecimiento mostraron mayor expresi&#xF3;n de TGF-&#x3B2; durante la fase aguda de la infecci&#xF3;n. Adem&#xE1;s, el envejecimiento de los ratones infectados mostraron elevaci&#xF3;n de citoquinas pro-inflamatorias y la contrataci&#xF3;n de quimioquinas CXCL12 fibrocito en el lavado broncoalveolar. Los an&#xE1;lisis de infecci&#xF3;n por el virus l&#xED;tico y la reactivaci&#xF3;n del virus indican que los ratones viejos eran capaces de controlar la infecci&#xF3;n cr&#xF3;nica y provocar anti-virus respuestas inmunes. Sin embargo, la edad, los ratones infectados mostraron un incremento significativo en las respuestas de apoptosis in situ determinados por ensayo de TUNEL, los niveles de caspasa-3 y la expresi&#xF3;n de la mol&#xE9;cula pro-BIM apoptotitc. La apoptosis de las c&#xE9;lulas epiteliales del tipo II del pulm&#xF3;n en los pulmones de envejecimiento estuvo acompa&#xF1;ado por la regulaci&#xF3;n positiva de la tensi&#xF3;n ER Bip marcador y el empalme de XBP1. Estos resultados indican que el envejecimiento del pulm&#xF3;n es m&#xE1;s susceptible a las lesiones y la fibrosis asociada con el estr&#xE9;s ER, la apoptosis de las c&#xE9;lulas de tipo II del pulm&#xF3;n epiteliales y activaci&#xF3;n de pro-fibr&#xF3;ticos v&#xED;as.
