Keratocytes can become fibroblasts and myofibroblasts during corneal injury and wound healing. We used the in vitro bovine keratocyte repair model system, which involves culturing collagenase-isolated keratocytes in serum-free media and then adding serum or serum plus TGF-beta to the culture media to induce the fibroblast and myofibroblast phenotypes, respectively, to evaluate the synthesis of secreted products by the cells. Serum and serum plus TGF-beta rapidly induced the fibroblast morphology and alpha smooth muscle actin, a marker of myofibroblasts. Keratocytes cultured in serum and serum plus TGF-beta also increased the synthesis of several high molecular weight products (approximately 100kD and larger) and the accumulation of a 43kD protein shown to be osteonectin/SPARC by both sequencing tryptic peptides from the protein and by reaction with antisera to osteonectin/SPARC. Immunohistochemical staining of mouse corneas with antisera to SPARC seven days post-wounding also demonstrated an increased accumulation of SPARC in the regions undergoing repair. These results indicate SPARC accumulation is a marker for stromal repair.

Cheratociti possono diventare fibroblasti e miofibroblasti durante lesione corneale e la guarigione delle ferite. Abbiamo utilizzato il sistema di modello riparazione regolarizza bovina in vitro, che comporta la coltura cheratociti collagenasi-isolato in media serum-free e quindi aggiungendo siero o siero plus TGF-beta per i terreni di coltura per indurre i fenotipi dei fibroblasti e myofibroblast, rispettivamente, per valutare la sintesi dei prodotti secrete dalle cellule. Prolactin (PRL) has long been implicated in Xenopus metamorphosis as an anti-metamorphic and/or juvenilizing hormone. Numerous studies showed that PRL could prevent effects of either endogenous or exogenous thyroid hormone (TH; T(3)). It has been shown that expression of matrix metalloproteinases (MMPs) is induced by TH during Xenopus metamorphosis. Direct in vivo evidence, however, for such anti-TH effects by PRL with respect to MMPs has not been available for the early phase of Xenopus development or metamorphosis. To understand the functional role of PRL, we investigated effects of PRL on Xenopus collagenase-3 (XCL3) and collagenase-4 (XCL4) expression in a cultured Xenopus laevis cell line, XL-177. Northern blot analysis demonstrated that XCL3 and XCL4 expression were not detected in control or T(3)-treated cells, but were differentially induced by PRL in a dose- and time-dependent fashion. Moreover, treatment with IL-1alpha as well as phorbol myristate acetate (PMA), a protein kinase C (PKC) activator, or H8, a protein kinase A (PKA) inhibitor, augmented PRL-induced collagenase expression, suggesting that multiple protein kinase pathways and cytokines may participate in PRL-induced collagenase expression. Interestingly, XCL3 expression could be induced in XL-177 cells by T(3), but only when co-cultured with prometamorphic Xenopus tadpole tails (stage 54/55), suggesting that the tails secrete a required intermediate signaling molecule(s) for T(3)-induced XCL3 expression. Taken together, these data demonstrate that XCL3 and XCL4 can be differentially induced by PRL and T(3) and further suggest that PRL is a candidate regulator of TH-independent collagenase expression during the organ/tissue remodeling which occurs in Xenopus development.

Pour comprendre le r&#xF4;le fonctionnel de la PRL, nous avons &#xE9;tudi&#xE9; les effets de la PRL sur l&#x26;#39;expression de la collag&#xE9;nase Xenopus-3 (XCL3) et de la collag&#xE9;nase-4 (XCL4) dans une ligne de culture cellulaire Xenopus laevis, XL-177. Analyse par Northern blot a d&#xE9;montr&#xE9; que XCL3 et XCL4 expression n&#x26;#39;ont pas &#xE9;t&#xE9; d&#xE9;tect&#xE9;s dans le contr&#xF4;le ou T (3) des cellules trait&#xE9;es, mais &#xE9;taient diff&#xE9;rentiellement induite par la PRL dans une relation dose-et temps-d&#xE9;pendante de la mode. En outre, le traitement par l&#x26;#39;IL-1alpha ainsi que phorbol myristate ac&#xE9;tate (PMA), une prot&#xE9;ine kinase C (PKC) activateur, ou H8, un inhibiteur de la prot&#xE9;ine kinase A (PKA), l&#x26;#39;expression augment&#xE9;e de la collag&#xE9;nase PRL induite, ce qui sugg&#xE8;re que la prot&#xE9;ine kinase multiples voies et les cytokines peuvent participer &#xE0; l&#x26;#39;expression de la collag&#xE9;nase PRL induite. Prolattina (PRL) lungamente &egrave; stato implicato in Xenopus metamorfosi come un ormone anti-metamorphic o juvenilizing. Numerosi studi hanno mostrato che la PRL potrebbe prevenire gli effetti dell&#39;ormone tiroideo di sia endogeni che esogeni (TH; T(3)). &Egrave; stato dimostrato che l&#39;espressione delle metalloproteinasi della matrice (MMP) &egrave; indotta da TH durante la metamorfosi di Xenopus. The cytokine transforming growth factor-beta (TGF-beta) is a key mediator of fibrosis in all organs. Expression of fibrotic markers in repairing cutaneous wounds is reduced in mice lacking Smad3, a downstream cytoplasmic mediator of TGF-beta signaling (Ashcroft et al., 1999, Nat Cell Biol 1(5):260-266). This is correlated with a reduction in inflammation, and thus in the blood elements thought to be a significant source of TGF-beta at the wound site, the principle form being TGF-beta1. Since the major cellular source of TGF-beta in corneal wounds is the epithelium, and the principal isoform is TGF-beta2, we investigated whether Smad3 deficiency has similar anti-fibrotic effects on corneal repair. In contrast to the situation of cutaneous repair, expression of the fibrotic marker, fibronectin, was equivalent in corneal repair tissue of Smad3-/- mice as compared to their +/- littermates, even though expression of a second fibrotic marker not previously examined in cutaneous wounds, alpha-smooth muscle (sm) actin, was reduced. Also unlike in cutaneous wounds, the inflammatory response was unaffected. These differences between corneal and cutaneous repair correlated with the lack of apparent change in the levels of corneal TGF-beta2. There was a significant reduction of alpha-sm actin expression in stromal cell cultures established from Smad3-/- mice as compared to their +/- littermates, but the rate of cell proliferation stimulated by TGF-beta, as well as expression of fibronectin, was unaffected. Therefore, a deficiency in Smad3 has different effects on corneal and cutaneous repair, probably due to the difference in cellular source and principal isoform of the TGF-beta involved.

The healing of a skin wound is a complex process involving many cell lineages. In adult tissues, repair is always accompanied by a robust inflammatory response, which is necessary to counter the potential for infection at any site where the skin barrier is breached. Unlike embryonic tissues that can repair perfectly without a remnant scar at the wound site, adult tissue repair always leads to formation of a fibrotic scar where the wound has healed. In recent years, it has become clear that the wound inflammatory response may be, at least in part, responsible for fibrosis at sites of tissue repair. In this review, we consider the beneficial vs the detrimental functions of inflammatory cells during the repair response and compare data from other tissues, the lung, and liver, where fibrosis and its resolution may be related to a damage-triggered inflammatory response. We also consider how it may be possible to molecularly disentangle the potentially good from the bad influences of inflammatory cells during tissue repair and how fundamental studies in inflammatory cell biology may prove the way forward for development of drug targets in this respect.

La curaci&#xF3;n de una herida en la piel es un proceso complejo que involucra muchos linajes celulares. En los tejidos adultos, la reparaci&#xF3;n va siempre acompa&#xF1;ada de una fuerte respuesta inflamatoria, la cual es necesario para contrarrestar la posibilidad de infecci&#xF3;n en cualquier lugar donde se rompe la barrera de la piel. A diferencia de los tejidos embrionarios que se puede reparar perfectamente sin una cicatriz remanente en el sitio de la herida, la reparaci&#xF3;n de tejidos adultos siempre conduce a la formaci&#xF3;n de una cicatriz fibrosa que la herida ha sanado. En los &#xFA;ltimos a&#xF1;os, ha quedado claro que la respuesta inflamatoria puede ser herida, al menos en parte, responsables de la fibrosis en los sitios de reparaci&#xF3;n de tejidos. En esta revisi&#xF3;n, consideramos que el beneficio frente a las funciones en detrimento de las c&#xE9;lulas inflamatorias durante la respuesta de reparaci&#xF3;n y comparar los datos de otros tejidos, el pulm&#xF3;n y el h&#xED;gado, donde la fibrosis, y su resoluci&#xF3;n puede estar relacionado con un da&#xF1;o provocado la respuesta inflamatoria. Tambi&#xE9;n consideramos c&#xF3;mo puede ser posible separar molecularmente el bien, potencialmente, de las malas influencias de las c&#xE9;lulas inflamatorias en la reparaci&#xF3;n de tejidos y la forma fundamental de los estudios de la biolog&#xED;a de las c&#xE9;lulas inflamatorias puede ser el camino a seguir para el desarrollo de dianas terap&#xE9;uticas en este sentido.
