The plasma membrane is made up of lipids and proteins, and serves as an active interface between the cell and its environment. Many plasma-membrane proteins are laterally segregated in the plane of the membrane, but the underlying mechanisms remain controversial. Here we investigate the distribution and dynamics of a representative set of plasma-membrane-associated proteins in yeast cells. These proteins were distributed non-homogeneously in patterns ranging from distinct patches to nearly continuous networks, and these patterns were in turn strongly influenced by the lipid composition of the plasma membrane. Most proteins segregated into distinct domains. However, proteins with similar or identical transmembrane sequences (TMSs) showed a marked tendency to co-localize. Indeed we could predictably relocate proteins by swapping their TMSs. Finally, we found that the domain association of plasma-membrane proteins has an impact on their function. Our results are consistent with self-organization of biological membranes into a patchwork of coexistingÂ domains.

La membrane plasmique est constitu&eacute;e de lipides et de prot&eacute;ines et sert d&#39;interface active entre la cellule et son environnement. Plusieurs prot&eacute;ines de la membrane plasmique sont s&eacute;par&eacute;s lat&eacute;ralement dans le plan de la membrane, mais les m&eacute;canismes sous-jacents demeurent controvers&eacute;es. Ici, nous &eacute;tudions la distribution et la dynamique d&#39;un ensemble repr&eacute;sentatif de prot&eacute;ines associ&eacute;es &agrave; la membrane plasmatique dans les cellules de levure. Ces prot&eacute;ines ont &eacute;t&eacute; distribu&eacute;es de fa&ccedil;on non homog&egrave;ne dans les mod&egrave;les allant des correctifs distincts aux r&eacute;seaux presque continus, et ces mod&egrave;les ont &eacute;t&eacute; &agrave; leur tour fortement influenc&eacute;es par la composition lipidique de la membrane plasmique. Die Plasmamembran besteht aus Lipiden und Proteinen, und dient als aktive Schnittstelle zwischen der Zelle und ihrer Umgebung. Viele Plasma-Membranproteine sind seitlich in der Ebene der Membran getrennt, aber die zugrunde liegenden Mechanismen bleiben umstritten. Hier untersuchen wir die Verteilung und die Dynamik einer repr&auml;sentativen Gruppe von Plasma-Membran-assoziierte Proteine in Hefezellen. Diese Proteine wurden in Muster, die von unterschiedlichen Flecken bis hin zu nahezu kontinuierliche Netzwerke nicht homogen verteilt, und diese Muster waren wiederum beeinflusst durch die Lipid-Zusammensetzung der Plasmamembran. Die meisten Proteine getrennt in unterschiedlichen Dom&auml;nen. Proteine mit &auml;hnlichen oder identischen Transmembran-Sequenzen (TMSs) zeigte eine deutliche Tendenz zu co-localize. Tats&auml;chlich konnten wir wie geplant Proteine zu verlagern, durch tauschen ihre TMSs. Schlie&szlig;lich fanden wir, dass die Dom&auml;nenzuordnung Plasmamembran Proteine einen Einfluss auf ihre Funktion hat. Unsere Ergebnisse sind konsistent mit Selbstorganisation von biologischen Membranen in einem Flickwerk nebeneinander existierende Domains.

La membrana del plasma &egrave; costituita da lipidi e proteine e serve come interfaccia attiva tra la cellula e il suo ambiente. Molte proteine di membrana del plasma sono segregate lateralmente nel piano della membrana, ma i meccanismi sottostanti rimangono controversi. Qui indaghiamo la distribuzione e la dinamica di un insieme rappresentativo di plasma-membrana-associated proteins in cellule di lievito. Queste proteine sono state distribuite in modo non omogeneo nei modelli che vanno dalla patch distinte reti quasi continuo, e questi modelli erano a sua volta fortemente influenzati dalla composizione lipidica della membrana plasmatica. La maggior parte delle proteine segregati in domini distinti. Tuttavia, le proteine con sequenze transmembrane simili o identici (TMSs) ha mostrato una marcata tendenza a co-localize. Infatti noi potremmo delocalizzare prevedibilmente proteine scambiando loro TMSs. Infine, abbiamo trovato che l&#39;associazione del dominio delle proteine di membrana del plasma ha un impatto sulla loro funzione. La membrana plasm&aacute;tica est&aacute; compuesta por l&iacute;pidos y prote&iacute;nas y sirve como una interfaz activa entre la c&eacute;lula y su ambiente. Muchas prote&iacute;nas de la membrana plasm&aacute;tica est&aacute;n segregadas lateralmente en el plano de la membrana, pero los mecanismos subyacentes siguen siendo pol&eacute;micos. Aqu&iacute; investigamos la distribuci&oacute;n y la din&aacute;mica de un conjunto representativo de prote&iacute;nas asociadas a plasma-membrana en c&eacute;lulas de levadura. Estas prote&iacute;nas se distribuyeron no homog&eacute;neo en patrones desde distintos parches a redes casi continuas, y estos patrones fueron a su vez influenciados por la composici&oacute;n lip&iacute;dica de la membrana plasm&aacute;tica. Mayor&iacute;a de las prote&iacute;nas segregadas en dominios distintos. Sin embargo, las prote&iacute;nas con similares o id&eacute;nticas secuencias transmembrana (TMSs) mostraron una marcada tendencia a co-localizan. De hecho como era de esperarse podr&iacute;amos reubicar prote&iacute;nas mediante el canje de sus TMSs. Finalmente, se encontr&oacute; que la Asociaci&oacute;n de dominio de las prote&iacute;nas de la membrana plasm&aacute;tica tiene un impacto en su funci&oacute;n. Nuestros resultados son consistentes con la autoorganizaci&oacute;n de las membranas biol&oacute;gicas en un mosaico de dominios coexistentes.
