Hematopoietic &#x7279;&#x5B9A;&#x6162;&#x75C5;&#x6BD2;&#x8F7D;&#x4F53;&#x89C4;&#x907F;&#x7531;&#x4E8E;&#x5F02;&#x4F4D;&#x86CB;&#x767D;&#x8868;&#x8FBE;&#x7684;&#x7EF4;&#x65AF;&#x79D1;&#x7279;-&#x5965;&#x5C14;&#x5FB7;&#x91CC;&#x5947;&#x7EFC;&#x5408;&#x5F81;&#x7684;&#x7EC6;&#x80DE;&#x6BD2;&#x6027;&#x3002;

Efficient and safe gene modification of hematopoietic stem cells is a requirement for gene therapy of primary immunodeficiencies such as Wiskott-Aldrich syndrome. However, deregulated expression or ectopic expression in the progeny of transduced nonhematopoietic progenitor cells may lead to unwanted toxicity. We therefore analyzed the effect of ectopic expression of Wiskott-Aldrich syndrome protein (WASp) and the potential benefits of hematopoietic-specific lentiviral vectors (driven by the WAS proximal promoter). Overexpression of WASp by constitutive lentiviral vectors is highly toxic in nonhematopoietic cells because it causes dramatic changes in actin localization and polymerization that result in decreased cell viability, as evidenced by a significant growth disadvantage of WASp-overexpressing nonhematopoietic cells and increased cell death. These toxic effects do not affect cells of hematopoietic origin because, remarkably, we found that WASp cannot be readily overexpressed in T cells, even after multiple vector integrations per cell. The adverse cellular effects found after transduction of nonhematopoietic cells with constitutive lentiviral vectors are overcome by the use of transcriptionally targeted lentiviral vectors expressing WASp, which, at the same time, are efficient tools for gene therapy of WAS as demonstrated by their ability to reconstitute cellular defects from WASp-deficient mouse and human cells. We therefore postulate that transcriptionally regulated lentiviral vectors represent a safer and efficient alternative for the development of clinical protocols of WAS gene therapy.

Modification g&eacute;nique efficace et s&eacute;curitaire de cellules souches h&eacute;matopo&iuml;&eacute;tiques est une exigence pour la th&eacute;rapie g&eacute;nique des immunod&eacute;ficiences primaires ex. syndrome de Wiskott - Aldrich. Toutefois, d&eacute;r&eacute;glement&eacute;s ou expression ectopique dans la descendance de cellules prog&eacute;nitrices transduite nonhematopoietic peut-&ecirc;tre conduire &agrave; toxicit&eacute; non d&eacute;sir&eacute;e. Par cons&eacute;quent, nous avons analys&eacute; l&#39;effet de l&#39;expression ectopique de Wiskott - Aldrich, syndrome de prot&eacute;ine (WASp) et les avantages potentiels de h&eacute;matopo&iuml;&eacute;tiques sp&eacute;cifiques des vecteurs LENTIVIRAUX (pilot&eacute;s par le promoteur proximal WAS). La surexpression de la gu&ecirc;pe de vecteurs LENTIVIRAUX constitutives est hautement toxique dans les cellules de nonhematopoietic car il provoque des changements dans la localisation de l&#39;actine et la polym&eacute;risation qui entra&icirc;nent une diminution de viabilit&eacute;, cellulaire comme en t&eacute;moigne un d&eacute;savantage significatif de la croissance des cellules surexprimant la gu&ecirc;pe de nonhematopoietic et de la mort cellulaire accrue. Ces effets toxiques n&#39;affectent pas les cellules d&#39;origine h&eacute;matopo&iuml;&eacute;tique car, fait remarquable, nous avons constat&eacute; que gu&ecirc;pe ne peut pas &ecirc;tre facilement surexprim&eacute;e dans les cellules T, m&ecirc;me apr&egrave;s plusieurs int&eacute;grations de vecteur par cellule. Les effets cellulaires trouvent apr&egrave;s transduction de nonhematopoietic des cellules avec des vecteurs LENTIVIRAUX constitutives sont surmont&eacute;s par l&#39;utilisation de transcriptionnellement cibl&eacute;es vecteurs LENTIVIRAUX exprimant la gu&ecirc;pe, qui, en m&ecirc;me temps, sont des outils efficaces pour la th&eacute;rapie g&eacute;nique de WAS, comme en t&eacute;moigne leur capacit&eacute; &agrave; reconstituer des d&eacute;fauts cellulaires de souris d&eacute;ficientes en gu&ecirc;pe et les cellules humaines. Par cons&eacute;quent, nous postulons que les vecteurs LENTIVIRAUX transcriptionnellement r&eacute;glement&eacute;s repr&eacute;sentent une alternative plus s&ucirc;re et efficace pour l&#39;&eacute;laboration de protocoles cliniques de th&eacute;rapie g&eacute;nique WAS.

Effiziente und sichere genetischer Ver&auml;nderungen von h&auml;matopoetischen Stammzellen ist eine Voraussetzung f&uuml;r Gentherapie der prim&auml;re Immundefekte wie Wiskott - Aldrich-Syndrom. Jedoch kann deregulierten Ausdruck oder ektopische Expression in der Nachkommenschaft transduzierten Nonhematopoietic Vorl&auml;uferzellen zu unerw&uuml;nschten Toxizit&auml;t f&uuml;hren. Wir analysierten daher die Wirkung von ektopische Expression des Proteins Wiskott - Aldrich Syndrom (WASp) und die potenziellen Vorteile der h&auml;matopoetischen-spezifische Lentiviralen Vektoren (angetrieben durch den proximalen WAS-Projekttr&auml;ger). Eine &Uuml;berexpression von WASp durch konstitutive Lentiviralen Vektoren ist hochgiftig in Nonhematopoietic Zellen, weil es dramatische Ver&auml;nderungen in der Aktin-Lokalisierung verursacht und Polymerisation, die verminderte f&uuml;hren Zelle Entwicklungsf&auml;higkeit, wie ein signifikantes Wachstum Nachteil der WASp-&uuml;berexprimierenden Nonhematopoietic Zellen und erh&ouml;hte Zelltod belegt. Diese toxischen Wirkungen betreffen nicht Zellen h&auml;matopoetischen Ursprungs, denn Bemerkenswerterweise fanden wir, dass WASp leicht &uuml;berexprimiert in T-Zellen, auch nach mehreren Vektor-Integrationen pro Zelle nicht. Die zellul&auml;ren Nebenwirkungen gefunden, nachdem Transduktion von Nonhematopoietic Zellen mit konstitutiven Lentiviralen Vektoren werden durch den Einsatz von transcriptionally gezielte Lentiviralen Vektoren WASp, zum Ausdruck zu bringen, die zur gleichen Zeit, effiziente Werkzeuge f&uuml;r Gentherapie von WAS sind wie durch ihre F&auml;higkeit, zellul&auml;re M&auml;ngel von WASp-defizienten Maus und menschlichen Zellen zu rekonstituieren &uuml;berwunden. Wir nehmen daher, dass transcriptionally geregelte Lentivirale Vektoren eine sicherere und effizientere Alternative f&uuml;r die Entwicklung von klinischen Protokollen der Gentherapie WAS dar.

Modificazione genetica efficiente e sicura di cellule staminali ematopoietiche &egrave; un requisito per la terapia genica delle Immunodeficienze primarie quali Wiskott - Aldrich-Syndroms. Tuttavia, deregolamentato espressione o ectopica nella progenie di cellule progenitrici non emopoietico trasdotte possono portare a tossicit&agrave; indesiderate. Abbiamo quindi analizzato l&#39;effetto di espressione ectopica della proteina Wiskott - Aldrich-Syndroms (WASp) e i potenziali benefici di vettori lentivirali ematopoietici specifici (guidati dal promotore prossimale WAS). Sovraespressione di Vespa di vettori lentivirali costitutive &egrave; altamente tossici nelle cellule non emopoietico perch&eacute; provoca cambiamenti drammatici nella localizzazione di actina e polimerizzazione che si traducono in una riduzione delle cellule vitalit&agrave;, come evidenziato da uno svantaggio significativo di crescita delle cellule non emopoietico con sovraesprimono relativa WASp e aumento delle cellule morte. Questi effetti tossici non influenzano le cellule di origine ematopoietica perch&eacute;, incredibilmente, abbiamo trovato che Vespa non pu&ograve; essere prontamente overexpressed in cellule T, anche dopo pi&ugrave; integrazioni di vettore per ogni cella. Gli effetti negativi del cellulari trovano dopo la trasduzione delle cellule non emopoietico con vettori lentivirali costitutive sono superati dall&#39;uso di transcriptionally mirati vettori lentivirali esprimendo WASp, che, allo stesso tempo, sono strumenti efficaci per la terapia genica di WAS, come dimostrato dalla loro capacit&agrave; di ricostituire il cellulari difetti da WASp-carenti mouse e cellule umane. Abbiamo pertanto postulato che vettori lentivirali transcriptionally regolamentati rappresentano un&#39;alternativa pi&ugrave; sicura ed efficace per lo sviluppo di protocolli clinici di terapia genica WAS.

Modificaci&oacute;n g&eacute;nica eficiente y segura de las c&eacute;lulas madre hematopoy&eacute;ticas es un requisito para la terapia g&eacute;nica de inmunodeficiencias primarias como s&iacute;ndrome de Wiskott &ndash; Aldrich. Sin embargo, la expresi&oacute;n liberalizado o expresi&oacute;n ect&oacute;pica en la progenie de c&eacute;lulas progenitoras de nonhematopoietic transduced puede llevar a toxicidad no deseada. Primary immunodeficiencies (PID) are caused by mutations in genes that impair the development or activity of the immune system. Although bone marrow transplants achieve long time restoration in up to 90% of treated patients, morbidity and mortality are still high for some PID and adequate donors are not always available. Gene Therapy (GT) was envisioned as an alternative treatment for PID by inserting the correct gene into the patient's haematopoietic stem cells (HSCs). Up to date, GT for PID has succeeded in 40 of 44 patients treated in four clinical trials. However, five children enrolled in the SCID-X1 clinical trial developed leukaemia-like disease produced by aberrant expression of oncogenes. This phenomenon resulted fatal in one patient and represented a severe setback for gene therapy. Since then, vector development has been a priority in the GT field, by refining existing Murine laeukemia virus (MLV)-based vectors or by developing new ones. This review summarize existing methodologies for PID GT highlighting the importance of animal models in the PID GT success and focusing on new gene transfer vectors to achieve safe, efficient and stable gene modification.

Aunque los trasplantes de m&eacute;dula &oacute;sea alcanzar largo restauraci&oacute;n de tiempo en hasta un 90% de pacientes tratados, morbilidad y mortalidad siguen siendo altos para algunos PID y donantes adecuados no siempre est&aacute;n disponibles. Terapia del gene (GT) fue concebido como un tratamiento alternativo por PID introduciendo el gen correcto en las c&eacute;lulas del paciente madre hematopoy&eacute;ticas (HSCs). Hasta la fecha, GT para PID ha logrado 40 de 44 pacientes tratados en cuatro ensayos cl&iacute;nicos. Sin embargo, cinco ni&ntilde;os matriculados en el ensayo cl&iacute;nico de SCID - X 1 desarrollaron leucemia-como enfermedad producida por la expresi&oacute;n aberrante de oncogenes. Abstract The development of vectors that express a therapeutic transgene efficiently and specifically in hematopoietic cells (HCs) is an important goal for gene therapy of hematological disorders. We have previously shown that a 500-bp fragment from the proximal Was gene promoter in a lentiviral vector (LV) was sufficient to achieve more than 100-fold higher levels of Wiskott-Aldrich syndrome protein in HCs than in nonhematopoietic cells (non-HCs). We show now that this differential was reduced up to 10 times when the enhanced green fluorescent protein gene (eGFP) was expressed instead of Was in the same LV backbone. Insertion of Was cDNA sequences downstream of eGFP in these LVs had a negative effect on transgene expression. This effect varied in different cell types but, overall, Was cDNA sequences increased the hematopoietic specificity of Was promoter-driven LV. We have characterized the minimal fragment required to increase hematopoietic specificity and have demonstrated that the mechanism involves Was promoter regulation and RNA processing. In addition, we have shown that Was cDNA sequences interfere with the enhancer activity of the woodchuck posttranscriptional regulatory element. These results represent the first data showing the role of Was intragenic sequences in gene regulation.

R&eacute;sum&eacute; la mise au point de vecteurs qui expriment un transg&egrave;ne th&eacute;rapeutique efficace et plus pr&eacute;cis&eacute;ment dans les cellules h&eacute;matopo&iuml;&eacute;tiques (HCs) est un objectif important pour la th&eacute;rapie g&eacute;nique des maladies h&eacute;matologiques. Nous avons d&eacute;montr&eacute; pr&eacute;c&eacute;demment qu&#39;un fragment de 500 PB de proximale a &eacute;t&eacute; le promoteur du g&egrave;ne dans un vecteur lentiviral (LV) ne suffisait pas atteindre plus de 100 fois plus &eacute;lev&eacute;s de prot&eacute;ine de Wiskott - Aldrich, syndrome de HCs que dans les cellules nonhematopoietic (non-HCs). Astratto allo sviluppo di vettori che esprimono un transgene terapeutico in modo efficiente e in particolare in cellule ematopoietiche (HCs) &egrave; un obiettivo importante per la terapia genica delle malattie ematologiche. In precedenza abbiamo dimostrato che un frammento di 500 bp dalla prossimale &egrave; stato promotore del gene in un vettore lentivirale (LV) &egrave; stato sufficiente a raggiungere pi&ugrave; di 100 volte pi&ugrave; alti livelli della proteina di Wiskott - Aldrich-Syndroms nell&#39;HCs che nelle cellule non emopoietico (non-HCs). Mostriamo ora che questo differenziale &egrave; stato ridotto fino a 10 volte quando &egrave; stato espresso il gene della proteina fluorescente verde migliorato (eGFP) invece era la stessa spina dorsale LV. Inserimento di sequenze del cDNA a valle di eGFP in questi LVs ha un effetto negativo sull&#39;espressione del transgene. Questo effetto variato tipi differenti delle cellule ma, nel complesso, era sequenze del cDNA aumentata la specificit&agrave; ematopoietica del fu promotore-driven lv Noi abbiamo caratterizzato il minimo frammento necessaria per aumentare la specificit&agrave; ematopoietiche e hanno dimostrato che il meccanismo coinvolge fu promotore regolamento ed elaborazione di RNA. Resumen el desarrollo de vectores que expresan un transgen terap&eacute;utico eficaz y espec&iacute;ficamente en las c&eacute;lulas hematopoy&eacute;ticas (HCs) es un objetivo importante para la terapia g&eacute;nica de desordenes hematol&oacute;gicos. Previamente hemos demostrado que un fragmento de 500-bp de la pr&oacute;xima fue el promotor del gen en un vector lentivirales (LV) fue suficiente para conseguir m&aacute;s de centesimal a mayores niveles de prote&iacute;na del s&iacute;ndrome de Wiskott &ndash; Aldrich en HCs que en las c&eacute;lulas nonhematopoietic (no-HCs). Mostramos ahora que esta diferencia se redujo hasta 10 veces cuando el gen de la prote&iacute;na fluorescente verde mejorada (eGFP) se expres&oacute; en vez de en la misma columna vertebral de LV. Inserci&oacute;n de secuencias de cDNA aguas abajo de eGFP en estos LVs tuvo un efecto negativo en la expresi&oacute;n del transg&eacute;n. Este efecto variada en c&eacute;lulas diferentes tipos pero, total, fue secuencias de cDNA aumentaron la especificidad hematopoy&eacute;tica de fue impulsada por promotor LV Hemos caracterizado el m&iacute;nimo fragmento necesaria para aumentar la especificidad hematopoy&eacute;tico y han demostrado que el mecanismo implica fue promotor regulaci&oacute;n y procesamiento del RNA. Adem&aacute;s, hemos demostrado que era secuencias de cDNA interfiere con la actividad de potenciador del elemento regulador postranscripcional marmota. Dendritic cells (DCs) initiate immune responses as well as tolerance. We showed previously that the neuropeptide vasoactive intestinal peptide (VIP) suppresses innate immune responses, modulates adaptive responses by generating regulatory T cells (Treg) through the induction of tolerogenic DCs (tDCs), and has therapeutic effects in models of autoimmune/inflammatory disorders. Systemic VIP administration is limited by its short biological half-life and by its pleiotropic effects on the cardiovascular system and gastrointestinal tract. Therefore, we used lentiviral vectors to genetically engineer VIP-expressing bone marrow-derived DC (BMDC) and characterized the transduced LentiVIP-DC in terms of phenotype and therapeutic effects in models of experimental autoimmune encephalomyelitis (EAE) and cecal ligation and puncture (CLP) sepsis. LentiVIP-DCs secrete VIP, and resemble tDCs through lack of co-stimulatory molecule upregulation, lack of proinflammatory cytokine secretion, increased interleukin (IL)-10 production, and poor stimulation of allogeneic T cells. A single inoculation of LentiVIP-DC in EAE or CLP mice had therapeutic effects, which correlated with reduced expression of proinflammatory cytokines and increased IL-10 production in spinal cord and peritoneal fluid, respectively. In contrast to systemic VIP administration that requires repeated, high-dose inoculations, local delivery of VIP by LentiVIP-DC may represent a promising therapeutic tool for the treatment of autoimmune diseases and inflammatory disorders.

Lentiviral vectors (LVs) are considered one of the most promising vehicles to efficiently deliver genetic information for basic research and gene therapy approaches. Combining LVs with drug-inducible expression systems should allow tight control of transgene expression with minimal side effect on relevant target cells. A new doxycycline-regulated system based on the original TetR repressor was developed in 1998 as an alternative to the TetR-VP16 chimeras (tTA and rtTA) to avoid secondary effects due to the expression of transactivator domains. However, previously described TetR-based systems required cell cloning and/or antibiotic selection of tetracycline-responsive cells in order to achieve good regulation. In the present manuscript we have constructed a dual Tet-ON system based on two lentiviral vectors, one expressing the TetR through the spleen focus forming virus (SFFV) promoter (STetR) and a second expressing eGFP through the regulatable CMV-TetO promoter (CTetOE). Using these vectors we have demonstrated that the TetR repressor, contrary to the reverse transactivator (rtTA), can be expressed in excess to bind and modulate a high number of TetO operons. We have also showed that this dual vector system can generate regulatable bulk cell lines (expressing high levels of TetR) that are able to modulate transgene expression either by varying doxycycline concentration and/or by varying the amount of CTetOE vector genomes per cell. Based on these results we have developed a new all-in-one lentiviral vector (CEST) driving the expression of TetR through the SFFV promoter and the expression of eGFP through the doxycycline-responsive CMV-TetO operon. This vector efficiently produced Tet-ON regulatable immortalized (293T) and primary (human mesenchymal stem cells and human primary fibroblasts) cells. Bulk doxycycline-responsive cell lines express high levels of the transgene with low amount of doxycycline and are phenotypically indistinct from its parental cells.

Vecteurs LENTIVIRAUX (LVs) sont consid&eacute;r&eacute;s comme un des v&eacute;hicules plus prometteurs pour d&eacute;livrer efficacement l&#39;information g&eacute;n&eacute;tique pour des approches fondamentales de th&eacute;rapie g&eacute;nique et de la recherche. Combinant LVs avec syst&egrave;mes d&#39;expression inductible par la drogue devrait permettre un contr&ocirc;le serr&eacute; de transgene expression avec des effets secondaires minimes sur les cellules cibles concern&eacute;s. Un nouveau syst&egrave;me de doxycycline r&eacute;glement&eacute;s bas&eacute; sur le r&eacute;presseur TetR original a &eacute;t&eacute; d&eacute;velopp&eacute; en 1998 comme une alternative pour les chim&egrave;res de Tite-VP16 (tTA et rtTA) afin d&#39;&eacute;viter des effets secondaires en raison de l&#39;expression des domaines transactivateur. Cependant, d&eacute;crit pr&eacute;c&eacute;demment clonage de syst&egrave;mes ax&eacute;s sur la Tite cellulaire requis et/ou antibiotique s&eacute;lection de cellules sensibles &agrave; la t&eacute;tracyclines pour atteindre la bonne r&eacute;glementation. Dans le pr&eacute;sent manuscrit, nous avons construit un syst&egrave;me dual de Tet-ON bas&eacute; sur deux vecteurs LENTIVIRAUX, on exprime la Tite gr&acirc;ce &agrave; la mise au point de rate formant le promoteur du virus (SFFV) (STetR) et un deuxi&egrave;me eGFP exprimant par le biais du promoteur de CMV-TetO r&eacute;glementable (CTetOE). &Agrave; l&#39;aide de ces vecteurs, nous avons d&eacute;montr&eacute; que le r&eacute;presseur de Tite, contrairement &agrave; l&#39;inverse transactivateur (rtTA), peut &ecirc;tre exprim&eacute; en exc&egrave;s de lier et de moduler un nombre &eacute;lev&eacute; de TetO op&eacute;rons. Nous avons &eacute;galement montr&eacute; que ce syst&egrave;me de double vecteur peut g&eacute;n&eacute;rer des lign&eacute;es de cellules r&eacute;glementable en vrac (exprimant des niveaux &eacute;lev&eacute;s de Tite) qui sont capables de moduler l&#39;expression du transg&egrave;ne soit en faisant varier la concentration de la doxycycline et/ou en faisant varier la quantit&eacute; de CTetOE vector g&eacute;nomes par cellule. Selon ces r&eacute;sultats, nous avons d&eacute;velopp&eacute; un nouveau tout-en-un g&egrave;nes vecteur (CEST) conduisant &agrave; l&#39;expression de Tite par le promoteur SFFV et l&#39;expression d&#39;eGFP par le biais de l&#39;op&eacute;ron CMV-TetO doxycycline-sensible. Ce vecteur produit efficacement Tet-ON r&eacute;glementable 293 immortalis&eacute; (t) et les cellules primaires (cellules souches m&eacute;senchymateuses humaines et les fibroblastes humains primaires). Des lign&eacute;es de cellules sensibles &agrave; la doxycycline en vrac expriment des niveaux &eacute;lev&eacute;s du transg&egrave;ne avec faible quantit&eacute; de doxycycline et sont ph&eacute;notypiquement indistinctes de ses cellules parentales.

D&eacute;veloppement d&#39;un syst&egrave;me tout-en-un vecteur lentiviral bas&eacute; sur la Tite original pour la g&eacute;n&eacute;ration simple de lign&eacute;es cellulaires de Tet-ON.

Lentivirale Vektoren (LVs) gelten als eines der vielversprechendsten Fahrzeuge effizient genetischen Informationen f&uuml;r grundlegende Forschung und gen-Therapie-Ans&auml;tze zu liefern. Kombination von LVs mit Droge-inducible Expressionssysteme sollten strenge Kontrolle &uuml;ber Transgenexpression mit minimaler Nebeneffekt auf relevanten Zielzellen erm&ouml;glichen. Ein neues Doxycyclin regulierte System, basierend auf der urspr&uuml;nglichen TetR-Hemmer entwickelte sich im Jahr 1998 als Alternative zu den TetR-VP16 Chim&auml;ren (tTA und RtTA) Nebenwirkungen aufgrund der Ausdruck Transactivator Domains zu vermeiden. Jedoch zuvor beschriebenen TetR-basierte Systeme erforderlich Zelle Klonen und/oder Antibiotika Auswahl von Tetracyclin-responsive Zellen, um gute Regelung zu erreichen. In der vorliegenden Handschrift haben wir ein duales Tet-ON-System basiert auf zwei Lentiviralen Vektoren, ein Ausdruck der TetR durch die Milz-Fokus bilden Virus (SFFV) Projekttr&auml;ger (STetR) und einen zweiten Ausdruck eGFP durch dosierbare CMV-TetO Projekttr&auml;ger (CTetOE) errichtet. Mit diesen Vektoren haben wir bewiesen, dass der Verdr&auml;nger TetR entgegen den umgekehrten Transactivator (RtTA), im &Uuml;berma&szlig; zu binden und eine hohe Anzahl von TetO Operone modulieren ausgedr&uuml;ckt werden kann. Wir haben auch gezeigt, dass dieses dualen Vektor-System kann dosierbare Massen-Zelllinien generieren (hohe TetR auszudr&uuml;cken), die Transgenexpression modulieren k&ouml;nnen entweder durch Variation von Doxycyclin Konzentration bzw. durch Variation der Menge an CTetOE Genome pro Zelle Vektor. Basierend auf diesen Ergebnissen haben wir einen neuen all-in-One Lentiviralen Vektor (MESZ) fahren, der Ausdruck der TetR durch den Projekttr&auml;ger SFFV und der Ausdruck der eGFP durch die Doxycyclin-responsive CMV-TetO-Operon entwickelt. Dieser Vektor effizient erzeugt Tet-ON dosierbare verewigt (293T) und Prim&auml;rzellen (humaner mesenchymaler Stammzellen und menschlichen prim&auml;re Fibroblasten). Lose Doxycyclin-responsive Zelllinien express hohe Transgene mit niedrigen Betrag von Doxycyclin und aus seiner elterlichen Zellen ph&auml;notypisch nicht fokussiert sind.

Entwicklung eines all-in-One Lentiviralen Vektor-Systems auf der Grundlage der urspr&uuml;nglichen TetR f&uuml;r die einfache Generierung von Tet-ON-Zelllinien.

Vettori lentivirali (LVs) sono considerati uno dei veicoli pi&ugrave; promettenti per fornire in modo efficiente informazioni genetiche per base approcci di terapia genica e di ricerca. Combinando LVs con sistemi di espressione droga-inducible dovrebbe consentire stretto controllo dell&#39;espressione del transgene con effetti collaterali minimi su cellule bersaglio pertinenti. Un nuovo sistema regolamentato doxiciclina basato sul repressor TetR originale &egrave; stato sviluppato nel 1998 come alternativa per le chimere TetR-VP16 (tTA e rtTA) per evitare effetti secondari dovuti l&#39;espressione dei domini transactivator. Tuttavia, in precedenza descritto TetR-ha basato i sistemi richiesti cell clonazione o selezione antibiotica delle cellule tetraciclina-sensible a reagire al fine di conseguire il buon regolamento. Nel manoscritto presente noi abbiamo costruito un sistema duale di Tet-ON basato su due vettori lentivirali, uno che esprime il TetR attraverso la messa a fuoco della milza formando una seconda eGFP che esprimono attraverso il promotore CMV-TetO regolabile (CTetOE) e virus (SFFV) promotore (STetR). Utilizzando questi vettori abbiamo dimostrato che il repressore TetR, contrariamente a quanto il transactivator inversa (rtTA), pu&ograve; essere espresso in eccesso per associare e modulano un numero elevato di TetO operoni. Noi abbiamo anche mostrato che questo sistema di doppio vettore pu&ograve; generare linee cellulari bulk regolabile (che esprimono alti livelli di TetR) che sono in grado di modulare l&#39;espressione del transgene sia variando la concentrazione di doxiciclina e/o variando la quantit&agrave; di CTetOE vector genomi per cella. Sulla base di questi risultati abbiamo sviluppato un nuovo all-in-one lentivirali vector (CEST) guida l&#39;espressione del TetR attraverso il promotore SFFV e l&#39;espressione di eGFP attraverso l&#39;operone CMV-TetO doxiciclina-sensibli a reagire. Questo vettore prodotto efficientemente Tet-ON regolabile immortalata (293T) e celle primarie (cellule staminali mesenchimali e fibroblasti umani primari). Cellula doxiciclina-sensibli a reagire alla rinfusa esprimono alti livelli di transgene con bassa quantit&agrave; di doxiciclina e &egrave; fenotipicamente indistinte dalle sue cellule parentali.

Vectores lentivirales (LVs) se consideran uno de los veh&iacute;culos m&aacute;s prometedores para administrar informaci&oacute;n gen&eacute;tica para enfoques b&aacute;sicos de la terapia de la investigaci&oacute;n y gene. Combinando LVs con sistemas de expresi&oacute;n inducible por la droga debe permitir un control estricto de la expresi&oacute;n del transg&eacute;n con m&iacute;nimo efecto secundario en las c&eacute;lulas blanco relevante. Un nuevo sistema de doxiciclina-regulado basado en el represor Tet original fue desarrollado en 1998 como una alternativa a las quimeras de Tet-VP16 (tTA y rtTA) para evitar efectos secundarios debido a la expresi&oacute;n de dominios transactivator. Sin embargo, previamente descrito sistemas basados en Tet celular requiere clonaci&oacute;n o selecci&oacute;n antibi&oacute;tico de tetraciclinas responden c&eacute;lulas para lograr buena regulaci&oacute;n. En el presente manuscrito hemos construido un sistema dual de Tet-ON basado en dos vectores lentivirales, uno expresar el Tet concentr&aacute;ndose bazo formando promotor de virus (SFFV) (STetR) y un segundo eGFP expresar a trav&eacute;s de la promotora de CMV-TetO regulable (CTetOE). Usando estos vectores hemos demostrado que el represor del Tet, contrario a la inversa transactivator (rtTA), puede expresarse en exceso para atar y modulan un alto n&uacute;mero de operones de TetO. Tambi&eacute;n hemos demostrado que este sistema de doble vector puede generar l&iacute;neas de c&eacute;lulas de la regulable a granel (expresando altos niveles de Tet) que son capaces de modular la expresi&oacute;n del transg&eacute;n ya sea variando la concentraci&oacute;n de doxiciclina o variando la cantidad de CTetOE vector genomas por la c&eacute;lula. Basado en estos resultados hemos desarrollado nuevo all-in-one lentivirales vector (CEST) conduce a la expresi&oacute;n del Tet a trav&eacute;s de la promotora de la SFFV y la expresi&oacute;n de eGFP por el oper&oacute;n de CMV-TetO doxiciclina-sensible. Este vector producido eficientemente Tet-ON inmortalizadas regulable (293T) y c&eacute;lulas primarias (c&eacute;lulas madre mesenquimales humanas y fibroblastos humanos primarios). L&iacute;neas de c&eacute;lulas sensibles a la doxiciclina a granel expresan altos niveles del transg&eacute;n con baja cantidad de doxiciclina y son fenot&iacute;picamente indistintas de sus c&eacute;lulas parentales.
