Developments in the molecular biology and pharmacology of GLU(K5), a subtype of the kainate class of ionotropic glutamate receptors, have enabled insights into the roles of this subunit in synaptic transmission and plasticity. However, little is known about the possible functions of GLU(K5)-containing kainate receptors in pathological conditions. We report here that, in hippocampal slices, selective antagonists of GLU(K5)-containing kainate receptors prevented development of epileptiform activity--evoked by the muscarinic agonist, pilocarpine--and inhibited the activity when it was pre-established. In conscious rats, these GLU(K5) antagonists prevented and interrupted limbic seizures induced by intra-hippocampal pilocarpine perfusion, and attenuated accompanying rises in extracellular L-glutamate and GABA. This anticonvulsant activity occurred without overt side effects. GLU(K5) antagonism also prevented epileptiform activity induced by electrical stimulation, both in vitro and in vivo. Therefore, we propose that subtype-selective GLU(K5) kainate receptor antagonists offer a potential new therapy for epilepsy.

D&eacute;veloppements de la biologie mol&eacute;culaire et de pharmacologie de GLU(K5), un sous-type de la classe ka&iuml;nate des r&eacute;cepteurs ionotropiques du glutamate, ont permis de mieux comprendre les r&ocirc;les de cette sous-unit&eacute; dans la transmission synaptique et plasticit&eacute;. Cependant, on conna&icirc;t mal les fonctions possibles du GLU (K5)-contenant des r&eacute;cepteurs ka&iuml;nate dans des conditions pathologiques. Nous rapportons ici que, dans des tranches d&#39;hippocampe, des antagonistes s&eacute;lectifs de GLU (K5) - contenant le d&eacute;veloppement a emp&ecirc;ch&eacute; les r&eacute;cepteurs ka&iuml;nate d&#39;activit&eacute; &eacute;pileptiforme--&eacute;voqu&eacute;e par l&#39;agoniste muscarinique, pilocarpine--et inhibent l&#39;activit&eacute; quand il a &eacute;t&eacute; pr&eacute;&eacute;tabli. Chez les rats conscients, ces antagonistes GLU(K5) emp&ecirc;chent et interrompu limbiques convulsions induites par la perfusion intra-hippocampique pilocarpine et att&eacute;nu&eacute; accompagnant s&#39;&eacute;l&egrave;ve en extracellulaire L-glutamate et le GABA. Sviluppi nella biologia molecolare e farmacologia di GLU(K5), un sottotipo della classe di recettori ionotropici glutammato Kainato hanno permesso a intuizioni in ruoli di questa subunit&agrave; nella trasmissione sinaptica e plasticit&agrave;. Tuttavia, poco si sa circa le possibili funzioni di GLU (K5)-contenente i recettori Kainato in condizioni patologiche. Abbiamo una relazione qui, in fette hippocampal, antagonisti selettivi di GLU (K5) - contenente sviluppo impediti recettori Kainato dell&#39;attivit&agrave; epileptiform - evocato da agonista muscarinico, pilocarpina - e inibiscono l&#39;attivit&agrave; quando si era prefissato. Avances en la biolog&iacute;a molecular y Farmacolog&iacute;a de GLU(K5), un subtipo de la clase de kainato de receptores ionotr&oacute;picos de glutamato han permitido penetraciones en las funciones de esta subunidad en la transmisi&oacute;n sin&aacute;ptica y plasticidad. Sin embargo, poco se sabe acerca de las posibles funciones de GLU (K5)-que contienen receptores de kainato en condiciones patol&oacute;gicas. Divulgamos aqu&iacute; que, en rodajas hipocampales, antagonistas selectivos de GLU (K5) - que contiene el desarrollo de prevenir los receptores de kainato de actividad epileptiforme--evocaban por el agonista muscar&iacute;nico, pilocarpina--e inhibici&oacute;n la actividad cuando era preestablecido. En ratas conscientes, estos antagonistas GLU(K5) prevenido e interrupci&oacute;n l&iacute;mbicas convulsiones inducidas por perfusi&oacute;n intra-hippocampal pilocarpina y acompa&ntilde;ando atenuada se levanta en medio extracelular L-glutamato y el GABA. Esta actividad anticonvulsivante ocurri&oacute; sin manifiestos efectos secundarios. Glu(K5) antagonismo tambi&eacute;n impidi&oacute; la actividad epileptiforme inducida por estimulaci&oacute;n el&eacute;ctrica, tanto in vitro como in vivo. Por lo tanto, proponemos que subtipo selectivos antagonistas de receptores de kainato GLU(K5) ofrecen una nueva terapia potencial para la epilepsia.

Prolonged use of the dopamine precursor l-DOPA for the treatment of Parkinson's disease commonly results in abnormal involuntary movements, which are termed l-DOPA-induced dyskinesia (LID). Over-activity at corticostriatal synapses onto neurons of the direct and indirect striatal output pathways has been implicated in the development of dyskinesia, but it has proved difficult to investigate the pathways separately due to their morphological similarities. The recent development of bacterial artificial chromosome mice that express green fluorescent protein in either the direct or indirect pathway allows visual identification of the output neurons in each pathway. Here we describe the use of two different strains of these transgenic mice (pure FVB and FVB crossed with C57B1/6) in the development of mouse models of l-DOPA-induced dyskinesia. This model will allow the direct and indirect pathways to be studied individually to delineate the cellular and molecular mechanisms underlying dyskinesias. These studies demonstrate that mouse strain impacts on behavioural responses and l-DOPA sensitivity. Therefore, when generating mouse models of LID, strain must be taken into consideration when choosing the l-DOPA dosing regimen.

L&#x26;#39;utilisation prolong&#xE9;e du pr&#xE9;curseur de la dopamine L-DOPA pour le traitement de la maladie de Parkinson entra&#xEE;ne g&#xE9;n&#xE9;ralement des mouvements anormaux involontaires, qui sont appel&#xE9;s L-DOPA-la dyskin&#xE9;sie induite par (LID). Au cours de l&#x26;#39;activit&#xE9; au niveau des synapses-corticostriatal SUR DES neurones des voies directes et indirectes de sortie du striatum a &#xE9;t&#xE9; impliqu&#xE9; dans le d&#xE9;veloppement de la dyskin&#xE9;sie, mais il s&#x26;#39;est av&#xE9;r&#xE9; difficile d&#x26;#39;enqu&#xEA;ter sur les voies s&#xE9;par&#xE9;ment en raison de leurs similitudes morphologiques. Le d&#xE9;veloppement r&#xE9;cent de souris chromosomes artificiels de bact&#xE9;ries qui expriment la prot&#xE9;ine fluorescente verte soit dans la voie directe ou indirecte permet une identification visuelle des neurones de sortie dans chaque voie. Nous d&#xE9;crivons ici l&#x26;#39;utilisation de deux souches diff&#xE9;rentes de ces souris transg&#xE9;niques (FVB pur et FVB crois&#xE9; avec C57B1 / 6) dans le d&#xE9;veloppement de mod&#xE8;les murins de la L-DOPA-la dyskin&#xE9;sie induite par. Ce mod&#xE8;le permettra aux voies directes et indirectes qui seront &#xE9;tudi&#xE9;s individuellement afin de d&#xE9;limiter les m&#xE9;canismes cellulaires et mol&#xE9;culaires qui sous-tendent les dyskin&#xE9;sies. Ces &#xE9;tudes d&#xE9;montrent que les impacts souche de souris sur les r&#xE9;ponses comportementales et la L-DOPA sensibilit&#xE9;. Over-Activity am Corticostriatal Synapsen auf Neuronen von den direkten und indirekten Striatale Ausgang Wege hat verwickelt wurden, bei der Entwicklung von Dyskinesien, aber es erwies sich schwer, die Bahnen getrennt aufgrund ihrer morphologischen &Auml;hnlichkeiten zu untersuchen. Die aktuelle Entwicklung der bacterial artificial Chromosome-M&auml;use, die gr&uuml;nes fluoreszierendes Protein in beiden Ausdr&uuml;cken erm&ouml;glicht die direkte oder indirekte Pathway visuellen Identifikation von der Ausgabeneuronen in jeder Weg. Hier beschreiben wir die Verwendung von zwei verschiedenen St&auml;mmen von diese transgenen M&auml;usen (reine FVB und FVB gekreuzt mit C57B1/6) bei der Entwicklung von Mausmodelle von l-DOPA-induzierte Dyskinesien. Dieses Modell erlaubt die direkte und indirekte Wege in einzeln untersucht werden, um die zellul&auml;ren und molekularen Mechanismen Dyskinesien abzugrenzen. Diese Studien zeigen, dass die Auswirkungen von Maus-Stamm auf Verhaltensst&ouml;rungen Antworten und l-DOPA-Empfindlichkeit. Daher muss beim Generieren von Mausmodelle des LIDES Belastung ber&uuml;cksichtigt werden bei der Wahl der Dosierung l-DOPA-Therapie.

Erstellung eines Modells von L-DOPA-induzierte Dyskinesien bei zwei verschiedene Maus-St&auml;mme.

Eccessiva attivit&agrave; presso corticostriatal sinapsi sui neuroni di striatal della diretta e indiretta di output percorsi &egrave; stato implicato nello sviluppo di discinesia, ma si &egrave; dimostrato difficile da indagare le vie separatamente a causa della loro somiglianze morfologiche. Il recente sviluppo di topi cromosoma artificiale batterico che esprimono la proteina fluorescente verde in entrambi il pathway diretto o indiretto consente identificazione visiva dei neuroni di output in ogni percorso. Qui descriviamo l&#39;uso di due diversi ceppi di questi topi transgenici (puro FVB e FVB attraversato con C57B1/6) nello sviluppo di modelli murini della discinesia indotta da l-DOPA. Questo modello permetter&agrave; i percorsi diretti e indiretti essere studiato individualmente per delineare i meccanismi cellulari e molecolari alla base di discinesie. Questi studi dimostrano che il ceppo del mouse gli impatti sulle risposte comportamentali e sensibilit&agrave; di l-DOPA. Pertanto, durante la generazione di modelli murini di coperchio, ceppo deve tener conto quando si sceglie il regime di somministrazione di l-DOPA.

This study identifies the risk and protective factors associated with informal caregiving by older (â‰¥70Â years) Mexican Americans and profiles caregiving arrangements. Overall, a greater number of informal caregivers (nÂ =Â 92) were married and female. They also had higher physical functioning and better cognition than non-caregivers (nÂ =Â 1,888) but fewer visited a physician regularly. Informal caregivers also showed an increased risk of depressive symptoms. A third of caregivers spent more than 20Â h/day caregiving and the majority (84%) of care recipients were family members. In order to support the efforts of this disproportionately burdened caregiver group, increased social support and healthcare services are needed.
