Dit artikel presenteert de integratie van een spectrale focusmodule en een dual-output pulslaser, waardoor snelle hyperspectrale beeldvorming van gouden nanodeeltjes en kankercellen mogelijk wordt. Dit werk is bedoeld om de details van multimodale niet-lineaire optische technieken te demonstreren op een standaard laserscanmicroscoop.
Het onderzoeken van gouden nanodeeltjes (AuNP's) in levende systemen is essentieel om de interactie tussen AuNP's en biologische weefsels te onthullen. Bovendien, door niet-lineaire optische signalen zoals gestimuleerde Raman-verstrooiing (SRS), twee-foton geëxciteerde fluorescentie (TPEF) en transiënte absorptie (TA) in een beeldvormingsplatform te integreren, kan het worden gebruikt om biomoleculair contrast van cellulaire structuren en AuNP's op een multimodale manier te onthullen. Dit artikel presenteert een multimodale niet-lineaire optische microscopie en past deze toe om chemisch specifieke beeldvorming van AuNP's in kankercellen uit te voeren. Dit beeldvormingsplatform biedt een nieuwe benadering voor het ontwikkelen van efficiënter gefunctionaliseerde AuNP's en het bepalen of ze zich in vasculatuur rond de tumor, pericellulaire of cellulaire ruimtes bevinden.
Gouden nanodeeltjes (AuNP's) hebben een groot potentieel getoond als biocompatibele beeldvormingsondes, bijvoorbeeld als effectieve oppervlakte-verbeterde Raman-spectroscopie (SERS) substraten in verschillende biomedische toepassingen. Belangrijke toepassingen zijn gebieden zoals biosensing, bioimaging, oppervlakte-verbeterde spectroscopieën en fotothermische therapie voor kankerbehandeling1. 1. Het lasersysteem inschakelen
De Spectral Focusing Timing and Recombination Unit (SF-TRU) module wordt geïntroduceerd tussen de dual-output femtoseconde laser en de gemodificeerde laserscanmicroscoop. Het instelbare ultrasnelle lasersysteem dat in deze studie wordt gebruikt, heeft twee uitgangspoorten die één straal leveren op een vaste golflengte van 1.045 nm en de andere bundel die instelbaar is in het bereik van 680-1.300 nm. Deze studie heeft de combinatie van SF-TRU-module en ultrasnel lasersysteem met dubbele uitgang gepresenteerd en zijn toepassingen voor multimodale microspectroscopie gedemonstreerd.
