Presentiamo un protocollo per visualizzare direttamente le attività metaboliche nelle cellule regolate da aminoacidi utilizzando la microscopia a diffusione Raman stimolata (DO-SRS) con ossido di deuterio (acqua pesante D2O), che è integrata con la microscopia a fluorescenza di eccitazione a due fotoni (2PEF).
Gli amminoacidi aromatici essenziali (AAA) sono elementi costitutivi per sintetizzare nuove biomasse nelle cellule e sostenere le normali funzioni biologiche. Ad esempio, un'abbondante fornitura di AAA è importante per le cellule tumorali per mantenere la loro rapida crescita e divisione. Con questo, c'è una crescente domanda di un approccio di imaging altamente specifico e non invasivo con una preparazione minima del campione per visualizzare direttamente come le cellule sfruttano gli AAA per il loro metabolismo in situ. Qui, sviluppiamo una piattaforma di imaging ottico che combina il sondaggio all'ossido di deuterio (D2O) con lo scattering Raman stimolato (DO-SRS) e integra DO-SRS con fluorescenza di eccitazione a due fotoni (2PEF) in un singolo microscopio per visualizzare direttamente le attività metaboliche delle cellule HeLa sotto regolazione AAA. Collettivamente, la piattaforma DO-SRS fornisce un'elevata risoluzione spaziale e specificità di proteine e lipidi di nuova sintesi in singole unità cellulari HeLa. Inoltre, la modalità 2PEF può rilevare segnali di autofluorescenza di nicotinammide adenina dinucleotide (NADH) e flavina in modo label-free. Il sistema di imaging qui descritto è compatibile sia con modelli in vitro che in vivo , che è flessibile per vari esperimenti. Il flusso di lavoro generale di questo protocollo include la coltura cellulare, la preparazione dei terreni di coltura, la sincronizzazione cellulare, la fissazione cellulare e l'imaging dei campioni con le modalità DO-SRS e 2PEF.
Essendo aminoacidi aromatici essenziali (AAA), la fenilalanina (Phe) e il triptofano (Tryp) possono essere assorbiti dal corpo umano per sintetizzare nuove molecole per sostenere le normali funzioni biologiche1. Phe è necessario per la sintesi di proteine, melanina e tirosina, mentre Tryp è necessario per la sintesi di melatonina, serotonina e niacina 2,3. 1. Preparazione dei supporti

Integrated SRS Laser System | Applied Physics & Electronics, Inc. | picoEMERALD | picoEMERALD provides an output pulse at 1031 nm with 6-ps pulse width and 80-MHz repetition rate, which serves as the Stokes beam. The frequency doubled beam at 532 nm is used to synchronously seed a picosecond optical parametric oscillator (OPO) to produce a mode-locked pulse train with five~6 ps pulse width (the idler beam of the OPO is blocked with an,interferometric filter). The output wavelength of the OPO is tunable from 720–950 nm, which serves as the pump beam. The intensity of the 1031 nm Stokes beam is modulated sinusoidally by a built-in EOM at 8 MHz with a modulation depth of more than 90%. The pump beam is spatially overlapped with the Stokes beam by using a dichroic mirror inside picoEMERALD. The temporal overlap between pump and Stokes pulses are achieved with a built-in delay stage and optimized by the SRS signal of pure D2O at the microscope. |
