Current methods for analysis of data from studies of protein-protein interactions using fluorescence resonance energy transfer (FRET) emerged from several decades of research using wide-field microscopes and spectrofluorometers to measure fluorescence from individual cells or cell populations. Inherent to most measurements is an averaging of the distributions of FRET efficiencies over large populations of protein complexes, which washes out information regarding the stoichiometry and structure of protein complexes. Although the introduction of laser-scanning microscopes in principle could facilitate quantification of the distributions of FRET efficiencies in live cells, only comparatively recently did this potential fully materialize, through development of spectral- or lifetime-based approaches. To exploit this new opportunity in molecular imaging, it is necessary to further develop theoretical models and methods of data analysis. Using Monte Carlo simulations, we investigated FRET in homogenous and inhomogeneous spatial distributions of molecules. Our results indicate that an analysis based on distributions of FRET efficiencies presents significant advantages over the average-based approach, which include allowing for proper identification of biologically relevant FRET. This study provides insights into the effect of molecular crowding on FRET, and it offers a basis for information extraction from distributions of FRET efficiencies using simulations-based data fitting.

Comparison between whole distribution- and average-based approaches to the determination of fluorescence resonance energy transfer efficiency in ensembles of proteins in living cells.

Les m&#xE9;thodes actuelles d&#x26;#39;analyse des donn&#xE9;es provenant d&#x26;#39;&#xE9;tudes de interactions prot&#xE9;ine-prot&#xE9;ine utilisant la fluorescence de transfert d&#x26;#39;&#xE9;nergie par r&#xE9;sonance (FRET) a &#xE9;merg&#xE9; de plusieurs d&#xE9;cennies de recherche en utilisant l&#x26;#39;&#xE9;chelle microscopes &#xE0; champ et Spectrofluorim&#xE8;tres pour mesurer la fluorescence des cellules individuelles ou des populations cellulaires. Inh&#xE9;rent &#xE0; la plupart des mesures est une moyenne des distributions de rendements de plus de FRET de grandes populations de complexes prot&#xE9;iques, qui baigne l&#x26;#39;information au sujet de la stoechiom&#xE9;trie et la structure des complexes prot&#xE9;iques. Aktuelle Methoden zur Analyse von Daten aus Studien von Protein-Protein Interaktionen mit Fluoreszenz-Resonanz-Energie-Transfer (FRET) entstand aus mehreren Jahrzehnten Forschung mit weiten Feld Mikroskope und berbessert Messen Fluoreszenz von einzelnen Zellen oder Zellen-Populationen. Die meisten Messungen innewohnt eine Mittelwertbildung der Verteilungen der FRET-Effizienz &uuml;ber gro&szlig;e Bev&ouml;lkerungen von Proteinkomplexen, die Informationen &uuml;ber die St&ouml;chiometrie und Struktur von Proteinkomplexen w&auml;scht. Obwohl die Einf&uuml;hrung von Laser-scanning Mikroskope grunds&auml;tzlich erleichtern k&ouml;nnte nur vergleichsweise Quantifizierung der Verteilungen der FRET-Effizienz in lebenden Zellen, vor kurzem hat dieses Potenzial voll zustande, durch die Entwicklung von spektralen oder Lebensdauer-basierte Ans&auml;tze. Um diese neue M&ouml;glichkeit in der molekularen Bildgebung auszunutzen, ist es notwendig, theoretische Modelle und Methoden der Datenanalyse weiterzuentwickeln. Mit Monte-Carlo-Simulationen, untersuchten wir FRET in homogenen und inhomogenen r&auml;umlichen Verteilungen von Molek&uuml;len. Unsere Ergebnisse zeigen, dass eine Analyse auf der Grundlage von Verteilungen von FRET-Effizienz entscheidende Vorteile gegen&uuml;ber dem Durchschnitt basierenden Ansatz, die umfassen pr&auml;sentiert, zulassend korrekte Kennzeichnung der biologisch relevanten FRET. Diese Studie liefert Erkenntnisse &uuml;ber die Wirkung der molekularen Gedr&auml;nge am Bund und bietet eine Grundlage f&uuml;r Informationsextraktion aus Verteilungen der FRET-Effizienz mit Simulationen basierenden Daten Formst&uuml;ck.

Attuali metodi di analisi dei dati provenienti da studi di interazioni proteina-proteina mediante fluorescenza resonance energy transfer (FRET) &egrave; emerso da diversi decenni di ricerca utilizzando microscopi wide-field e spettrofluorimetri per misurare la fluorescenza da singole celle o popolazioni di cella. Inerente alla maggior parte delle misurazioni &egrave; una media delle distribuzioni di FRET efficienze sopra grandi popolazioni dei complessi della proteina, che lava le informazioni riguardanti la stechiometria e la struttura dei complessi della proteina. Sebbene l&#39;introduzione della scansione laser microscopi in linea di principio potrebbero facilitare la quantificazione delle distribuzioni di FRET efficienze in cellule vive, solo relativamente di recente ha fatto questo potenziale completamente materializzare, attraverso lo sviluppo di approcci spettrale o vita-based.
