Spiders spin high performance threads that have diverse mechanical properties for specific biological applications. To better understand the molecular mechanism by which spiders anchor their threads to a solid support, we solubilized the attachment discs from black widow spiders and performed in-solution tryptic digests followed by MS/MS analysis to identify novel peptides derived from glue silks. Combining matrix-assisted laser desorption ionization tandem time-of-flight mass spectrometry and cDNA library screening, we isolated a novel member of the silk gene family called pysp1 and demonstrate that its protein product is assembled into the attachment disc silks. Alignment of the PySp1 amino acid sequence to other fibroins revealed conservation in the non-repetitive C-terminal region of the silk family. MS/MS analysis also confirmed the presence of MaSp1 and MaSp2, two important components of dragline silks, anchored within the attachment disc materials. Characterization of the ultrastructure of attachment discs using scanning electron microscopy studies support the localization of PySp1 to small diameter fibers embedded in a glue-like cement, which network with large diameter dragline silk threads, producing a strong, adhesive material. Consistent with elevated PySp1 mRNA levels detected in the pyriform gland, MS analysis of the luminal contents extracted from the pyriform gland after tryptic digestion support the assertion that PySp1 represents one of the major constituents manufactured in the pyriform gland. Taken together, our data demonstrate that PySp1 is spun into attachment disc silks to help affix dragline fibers to substrates, a critical function during spider web construction for prey capture and locomotion.

Pour mieux comprendre le m&#xE9;canisme mol&#xE9;culaire par lequel les araign&#xE9;es ancrer leurs discussions sur un support solide, nous solubilis&#xE9; les disques de fixation de la veuve noire et r&#xE9;alis&#xE9;e en solution digestion trypsique suivie par MS / MS pour identifier de nouveaux peptides d&#xE9;riv&#xE9;s de la soie de colle. La combinaison de laser assist&#xE9;e par matrice d&#x26;#39;ionisation d&#xE9;sorption spectrom&#xE9;trie de masse tandem temps de vol et de criblage de banques d&#x26;#39;ADNc, nous avons isol&#xE9; un nouveau membre de la famille des g&#xE8;nes de la soie appel&#xE9; pysp1 et de d&#xE9;montrer que son produit prot&#xE9;ique est assembl&#xE9; dans les soies de disque de fixation. Alignement de la s&#xE9;quence d&#x26;#39;acides amin&#xE9;s &#xE0; PySp1 fibroins autres r&#xE9;v&#xE9;l&#xE9; la conservation dans le non-r&#xE9;p&#xE9;titif r&#xE9;gion C-terminale de la famille de la soie. MS / MS analyse a &#xE9;galement confirm&#xE9; la pr&#xE9;sence de MASP1 et MaSp2, deux composantes importantes de la soie dragline, ancr&#xE9;e dans les mat&#xE9;riaux de fixation de disque. Caract&#xE9;risation de l&#x26;#39;ultrastructure de disques de fixation &#xE0; l&#x26;#39;aide de balayage &#xE9;tudes en microscopie &#xE9;lectronique en charge la localisation de PySp1 aux fibres de petit diam&#xE8;tre noy&#xE9;s dans un ciment colle comme, quel r&#xE9;seau avec des fils de gros diam&#xE8;tre en soie dragline, la production d&#x26;#39;un fort, un mat&#xE9;riau adh&#xE9;sif. Conform&#xE9;ment &#xE0; &#xE9;lev&#xE9;es PySp1 les niveaux d&#x26;#39;ARNm d&#xE9;tect&#xE9;s dans la glande piriforme, l&#x26;#39;analyse MS du contenu luminal extraite de la glande piriforme soutien apr&#xE8;s digestion trypsique l&#x26;#39;affirmation selon laquelle PySp1 repr&#xE9;sente l&#x26;#39;un des constituants majeurs fabriqu&#xE9;s dans la glande piriforme. Pris dans leur ensemble, nos donn&#xE9;es montrent que PySp1 est fil&#xE9; en soie disque de fixation pour aider fibres dragline apposer sur des substrats, une fonction critique lors de la construction de toile d&#x26;#39;araign&#xE9;e pour la capture des proies et la locomotion.

Matrix assisted Laser Desorption Ionisation Tandem Time-of-Flight mass Spektrometrie und cDNA Bibliothek Screening kombinierend wir isoliert eine neuartige Mitglied der Seide-gen-Familie namens pysp1 und nachweisen, dass sein Protein-Produkt in der Anlage-Scheibe-Seide gefertigt wird. Ausrichtung der PySp1 Aminos&auml;ure-Sequenz in anderen Fibroins offenbart Erhaltung der nicht wiederholende C-terminalen Region aus der Familie der Seide. MS/MS-Analyse best&auml;tigte auch das Vorhandensein von MaSp1 und MaSp2, zwei wichtige Komponenten von Kratzbaggers Seide, in die Anlage CD Materialien verankert. Charakterisierung der Ultrastruktur der Anlage-Disks, die mit Scan Elektronenmikroskopie Studien unterst&uuml;tzen die Lokalisierung von PySp1 zu kleinem Durchmesser Fasern eingebettet in ein Kleber-wie Zement, die Vernetzung mit gro&szlig;em Durchmesser Kratzbaggers Seidenf&auml;den, produziert ein starkes, selbstklebend-Material. Einklang mit erh&ouml;hten PySp1-mRNA Niveaus in der birnenf&ouml;rmig Dr&uuml;se, erkannt und MS Inhaltsanalyse des luminalen aus extrahiert die birnenf&ouml;rmig Dr&uuml;se nach tryptic Verdauung Unterst&uuml;tzung die Behauptung, dass PySp1 eine der wichtigsten Bestandteile in die birnenf&ouml;rmig Dr&uuml;se hergestellt darstellt. Zusammengenommen zeigen unsere Daten, dass PySp1 in Anlage CD Seide zu helfen Kratzbaggers Fasern Substrate, eine kritische Funktion w&auml;hrend Spinne Netzaufbau f&uuml;r Beute erfassen und Fortbewegung anbringen gesponnen ist.

Combinando matrix-assisted laser desorption ionizzazione tandem a tempo di volo massa spettrometria e cDNA biblioteca screening, abbiamo isolato un romanzo membro della famiglia di seta gene chiamato pysp1 e dimostrare che il suo prodotto proteico &egrave; assemblato nelle sete disco allegato. Allineamento di sequenza aminoacidica PySp1 per altri fibroins rivelato conservazione nella regione C-terminale non ripetitiva della famiglia seta. MS/MS analisi confermata anche la presenza di MaSp1 e MaSp2, due componenti importanti di sete della gru a cingoli, ancorate all&#39;interno dei materiali del disco allegato. Caratterizzazione di ultrastruttura dei dischi allegati utilizzando gli studi di microscopia elettronica a scansione supporta la localizzazione di PySp1 alle fibre di piccolo diametro incorporato in un cemento simile a colla, che in rete con grande diametro della gru a cingoli filetti di seta, producendo un materiale forte, adesivo. Coerente con mRNA di PySp1 elevati livelli rilevati nella ghiandola piriforme, MS analisi del contenuto del lume estratta dalla ghiandola piriforme dopo supporto digestione trittica l&#39;asserzione che PySp1 rappresenta uno dei principali costituenti fabbricati nella ghiandola piriforme. Para comprender mejor el mecanismo molecular por el que las ara&#xF1;as anclar sus hilos a un soporte s&#xF3;lido, que solubilizan los discos de fijaci&#xF3;n de las ara&#xF1;as viuda negra y se realiz&#xF3; en soluci&#xF3;n tr&#xED;ptico digeridos seguido por MS / MS an&#xE1;lisis para identificar nuevos p&#xE9;ptidos derivados de las sedas de cola. La combinaci&#xF3;n de matriz asistida por l&#xE1;ser de ionizaci&#xF3;n de desorci&#xF3;n t&#xE1;ndem tiempo de vuelo espectrometr&#xED;a de masas y selecci&#xF3;n de la biblioteca de ADNc, se aisl&#xF3; un nuevo miembro de la familia de genes de seda llamado pysp1 y demostrar que su producto proteico se ensambla en las sedas de fijaci&#xF3;n de disco. En conjunto, nuestros datos demuestran que PySp1 se hila en sedas de discos de fijaci&#xF3;n para ayudar a las fibras de adherir a la red de arrastre de sustratos, una funci&#xF3;n cr&#xED;tica en la construcci&#xF3;n de tela de ara&#xF1;a para la captura de la presa y la locomoci&#xF3;n.
