The acinar cell culture plays a very important role in research of pancreatic pathophysiology. The aim of this study was to establish a long-term culture of human (foetal) pancreatic acinar cells in standardized nutrient media with supplements. Acinar cells were prepared from pancreatic tissues obtained from aborted foetus (> or =35 weeks) with no prior pancreatic complications by collagenase digestion and cultured using different media and supplements. The purity and phenotype of acinar cells was confirmed by various staining techniques and FACS. The acinar cell proliferation was determined at different time intervals by Bromo-deoxyuridine (BrdU) incorporation, and metabolic enzyme activity was analysed. The acini could be cultured and maintained in Ham's F-12 K/M199 media in the presence of 5% BSA, 0.1 mg/ml STI, 10 ng/ml EGF, and 10% FCS with the same morphological appearance as that of freshly prepared for 12 days with maximum viability of 80-85% and formation of monolayer without extracellular matrix. A significant BrdU incorporation of acinar cells in primary culture was observed which was maximum (105%) at day four. Higher amylase and lipase activity was seen in freshly isolated acinar cells which decreased with time of the culture. The established human pancreatic acinar cell culture may act as an excellent model to study exocrine dysfunction or pancreatitis in response to acinar cell injury.

La culture de cellules acineuses joue un r&ocirc;le tr&egrave;s important dans la recherche de physiopathologie du pancr&eacute;as. Le but de cette &eacute;tude &eacute;tait d&#39;&eacute;tablir une culture &agrave; long terme des cellules acineuses du pancr&eacute;as humains &agrave; (foetus) dans des milieux nutritifs normalis&eacute;s avec suppl&eacute;ments. Les cellules acineuses sont pr&eacute;par&eacute;s &agrave; partir des tissus pancr&eacute;atiques provenant de f&oelig;tus avort&eacute;s (&gt; ou = 35 semaines) avec pas de complications pancr&eacute;atiques pr&eacute;alables par digestion de collag&eacute;nase et cultiv&eacute;es &agrave; l&#39;aide de diff&eacute;rents m&eacute;dias et suppl&eacute;ments. La puret&eacute; et le ph&eacute;notype des cellules acineuses a &eacute;t&eacute; confirm&eacute;e par diff&eacute;rentes techniques de coloration et FACS. La prolif&eacute;ration des cellules acineuses &eacute;tait d&eacute;termin&eacute;e &agrave; diff&eacute;rents intervalles de temps par l&#39;incorporation de la Bromo-deoxyuridine (BrdU) et l&#39;activit&eacute; enzymatique m&eacute;tabolique a &eacute;t&eacute; analys&eacute;e. Les acini pourraient &ecirc;tre cultiv&eacute;s et entretenus dans les m&eacute;dias F-12 K/M199 Ham en pr&eacute;sence de 5 % de BSA, STI de 0,1 mg/ml et 10 ng/ml EGF FCS de 10 % avec le m&ecirc;me aspect morphologique que celle de fra&icirc;chement pr&eacute;par&eacute;e pendant 12 jours avec viabilit&eacute; maximale de 80 &agrave; 85 % et de la formation de la monocouche sans matrice extracellulaire. Nous observons une incorporation de BrdU importante des cellules acineuses en culture primaire qui &eacute;tait maximale (105 %) au quatri&egrave;me jour. Activit&eacute; amylase et lipase plus &eacute;lev&eacute;e a &eacute;t&eacute; observ&eacute;e dans les cellules acineuses fra&icirc;chement isol&eacute;es qui d&eacute;cro&icirc;t avec le temps de la culture. La culture &eacute;tablie des cellules acineuses du pancr&eacute;as humain peut agir comme un excellent mod&egrave;le pour &eacute;tudier la dysfonction exocrine ou pancr&eacute;atite en r&eacute;ponse &agrave; une l&eacute;sion des cellules acineuses.

Die Kultur Azinus Zellen spielt eine sehr wichtige Rolle in der Forschung des Pankreas Pathophysiologie. Ziel dieser Studie war eine Langzeitkultur menschlicher (fetale) Pankreas Azinus Zellen in Standard-N&auml;hrstoff-Medien mit Erg&auml;nzungen zu etablieren. Azinus Zellen wurden vorbereitet von pankreatischen Gewebe aus abgetriebenen F&ouml;ten gewonnen (&gt; oder = 35 Wochen) ohne vorherige pankreatischen Komplikationen von Collagenase-Verdauung und kultivierten mithilfe verschiedener Medien und Erg&auml;nzungen. Die Reinheit und Ph&auml;notyp Azinus Zellen wurde durch die verschiedenen befleckenden Techniken und FACS best&auml;tigt. Der Azinus Zellproliferation wurde in verschiedenen Zeitintervallen durch Bromo-Deoxyuridine (BrdU) Aufnahme bestimmt, und metabolische Enzymaktivit&auml;t analysiert. Die Acini k&ouml;nnte diese kultiviert und gepflegt in Ham&#39;s F-12 K/M199 Medien in Anwesenheit von 5 % BSA, 0,1 mg/ml STI, 10 ng/ml EGF und 10 % FCS mit dem gleichen morphologischen erscheinen wie der frisch zubereitete f&uuml;r 12 Tage mit maximaler Tragf&auml;higkeit von 80-85 % und Bildung der Monolage ohne extrazellul&auml;ren Matrix. Eine bedeutende BrdU-Aufnahme der Azinus Zellen in der Prim&auml;rkultur wurde beobachtet, die maximale (105 %) am Tag vier war. H&ouml;here Amylase und Lipase-Aktivit&auml;t wurde in frisch isolierten Azinus Zellen gesehen, die mit der Zeit der Kultur gesunken. Die etablierten menschlichen Pankreas Azinus Zellkultur kann als ein hervorragendes Modell exokrine Funktionsst&ouml;rung oder Pankreatitis als Reaktion auf Verletzung der Azinus Zellen wirken.

La coltura delle cellule acinose gioca un ruolo molto importante nella ricerca di Fisiopatologia del pancreas. Lo scopo di questo studio era di stabilire una cultura a lungo termine delle cellule acinose del pancreas (fetale) umane in terreno nutritivo standardizzato con integratori. Le cellule acinose sono state preparate dai tessuti del pancreas ottenuti dal feto abortito (&amp;gt; o = 35 settimane) senza complicazioni del pancreas preventiva di digestione collagenasi e colta utilizzando diversi mezzi e integratori. La purezza e il fenotipo delle cellule acinose &egrave; stato confermato da varie tecniche di colorazione e FACS. La proliferazione delle cellule acinose &egrave; stata determinata a intervalli di tempo diversi dall&#39;incorporazione di Bromo-deoxyuridine (BrdU), ed &egrave; stata analizzata l&#39;attivit&agrave; dell&#39;enzima metabolico. Gli acini potrebbero essere colta e mantenuti in media F-12 K/M199 Ham in presenza di 5% BSA, 0.1 mg/ml STI, EGF di 10 ng/ml e 10% FCS con lo stesso aspetto morfologico come quella di preparati per 12 giorni con validit&agrave; massima di 80-85% e formazione di monostrato senza matrice extracellulare. Un&#39;incorporazione di BrdU significativo delle cellule acinose in coltura primaria &egrave; stato osservato che era massimo (105%) a quattro giorni. Maggiore attivit&agrave; di amilasi e lipasi &egrave; stato visto nelle cellule acinose appena isolate che &egrave; diminuito con il tempo della cultura. La coltura delle cellule acinose del pancreas umano stabilito pu&ograve; agire come un eccellente modello per studiare esocrina o pancreatite in risposta alla lesione delle cellule acinose.

El cultivo de c&eacute;lulas acinares juega un papel muy importante en la investigaci&oacute;n de Fisiopatolog&iacute;a pancre&aacute;tica. El objetivo de este estudio fue establecer una cultura a largo plazo de las c&eacute;lulas acinares pancre&aacute;ticas (fetales) humanas en medios nutritivos estandarizados con suplementos. Las c&eacute;lulas acinares se prepararon de tejidos pancre&aacute;ticos de feto abortado (&gt; = 35 semanas) sin complicaciones pancre&aacute;ticas previo por la digesti&oacute;n de la colagenasa y cultivadas utilizando diferentes medios y suplementos. La pureza y el fenotipo de las c&eacute;lulas acinares fue confirmada por diversas t&eacute;cnicas de tinci&oacute;n y FACS. La proliferaci&oacute;n de c&eacute;lulas acinares se determin&oacute; en distintos intervalos de tiempo por la incorporaci&oacute;n de Bromo-deoxiuridina (BrdU), y se analiz&oacute; la actividad metab&oacute;lica de la enzima. Los acinos podr&iacute;an ser cultivados y mantenidos en medios de Ham F-12 K/M199 en presencia de 5% BSA, its de 0.1 mg/ml, 10 ng/ml EGF y FCS de 10% con el mismo aspecto morfol&oacute;gico que reci&eacute;n preparada durante 12 d&iacute;as con la m&aacute;xima viabilidad de 80-85% y la formaci&oacute;n de monocapa sin matriz extracelular. Se observ&oacute; una importante incorporaci&oacute;n de BrdU de c&eacute;lulas acinares en cultivo primario que era m&aacute;ximo (105%) en la cuarta jornada. Se observ&oacute; una mayor actividad de amilasa y lipasa en c&eacute;lulas acinares reci&eacute;n aisladas que disminuyeron con el tiempo de la cultura. La cultura de c&eacute;lula acinar pancre&aacute;tica humana establecido puede actuar como un excelente modelo para el estudio de disfunci&oacute;n exocrina o pancreatitis en respuesta a la lesi&oacute;n de c&eacute;lulas acinares.

Chemokines have been known to play a critical role in pathogenesis of chronic pancreatitis and acinar cell death. However, the role played by one of the CXC chemokines: CXCL10 in regulation of acinar cell death has remained unexplored. Hence, this study was designed to assess the role of CXCL10 promoting apoptosis in ex vivo cultured acinar cells. Primary human pancreatic acinar cell cultures were established and exposed to varying doses of CXCL10 for different time intervals. Apoptotic induction was evaluated by both qualitative as well as quantitative analyses. The involvement of upstream apoptotic regulators like pJNK, p38 and Bax was established on the basis of their increased expression of CXCL10. The change of Psim by 50% was observed in the presence of CXCL10 in treated acinar cells along with enhanced expression of Cytochrome C, apaf-1 and caspase 9/3 activation. In addition, ATP depletion was also noticed in CXCL10 stimulated acinar cells. CXCL10 induces cell death in human cultured pancreatic cells leading to apoptosis and DNA fragmentation via CXCR3 signalling. These signalling mechanisms may play an important role in parenchymal cell loss and injury in pancreatitis.

Embryonic stem (ES) cell technology may serve as a platform for the discovery of drugs to treat diseases such as diabetes. However, because of difficulties in establishing reliable ES cell differentiation methods and in creating cost-effective plating conditions for the high-throughput format, screening for molecules that regulate pancreatic beta cells and their immediate progenitors has been limited. A relatively simple and inexpensive differentiation protocol that allows efficient generation of insulin-expressing cells from murine ES cells was previously established in our laboratories. In this report, this system is characterized in greater detail to map developmental cell stages for future screening experiments. Our results show that sequential activation of multiple gene markers for undifferentiated ES cells, epiblast, definitive endoderm, foregut, and pancreatic lineages was found to follow the sequence of events that mimics pancreatic ontogeny. Cells that expressed enhanced green fluorescent protein, driven by pancreatic and duodenal homeobox 1 or insulin 1 promoter, correctly expressed known beta cell lineage markers. Overexpression of Sox17, an endoderm fate-determining transcription factor, at a very early stage of differentiation (days 2-3) enhanced pancreatic gene expression. Overexpression of neurogenin3, an endocrine progenitor cell marker, induced glucagon expression at stages when pancreatic and duodenal homeobox 1 message was present (days 10-16). Forced expression (between days 16 and 25) of MafA, a pancreatic maturation factor, resulted in enhanced expression of insulin genes, glucose transporter 2 and glucokinase, and glucose-responsive insulin secretion. Day 20 cells implanted in vivo resulted in pancreatic-like cells. Together, our differentiation assay recapitulates the proceedings and behaviors of pancreatic development and will be valuable for future screening of beta cell effectors.

Madre embrionarias (ES) la tecnolog&#xED;a de c&#xE9;lulas puede servir como una plataforma para el descubrimiento de medicamentos para tratar enfermedades como la diabetes. Sin embargo, debido a las dificultades en el establecimiento de m&#xE9;todos fiables ES diferenciaci&#xF3;n celular y en la creaci&#xF3;n de condiciones efectivas de costo-planchas para el formato de alto rendimiento, la detecci&#xF3;n de las mol&#xE9;culas que regulan las c&#xE9;lulas beta del p&#xE1;ncreas y sus progenitores inmediatos ha sido limitada. Una diferenciaci&#xF3;n relativamente simple y barato protocolo que permite la generaci&#xF3;n eficiente de la insulina-c&#xE9;lulas que expresan a partir de c&#xE9;lulas embrionarias murinas se estableci&#xF3; previamente en nuestros laboratorios. En este informe, este sistema se caracteriza en mayor detalle para asignar las etapas de desarrollo de c&#xE9;lulas para los experimentos de cribado en el futuro. Nuestros resultados muestran que la activaci&#xF3;n secuencial de m&#xFA;ltiples marcadores gen&#xE9;ticos de las c&#xE9;lulas madre embrionarias no diferenciadas, epiblasto, endodermo definitivo, intestino anterior, y los linajes de p&#xE1;ncreas se encontr&#xF3; a seguir la secuencia de eventos que imita la ontogenia de p&#xE1;ncreas. Las c&#xE9;lulas que expresaban la prote&#xED;na verde fluorescente mejorado, impulsado por el p&#xE1;ncreas y el duodeno homeobox 1 o promotor de la insulina 1, expresado correctamente conocidos marcadores beta linaje de c&#xE9;lulas. La sobreexpresi&#xF3;n de Sox17, un endodermo destino, la determinaci&#xF3;n de factor de transcripci&#xF3;n, en una fase muy temprana de la diferenciaci&#xF3;n (d&#xED;as 2-3) una mayor expresi&#xF3;n de genes de p&#xE1;ncreas.
