When the process of cell-fate determination is examined at single-cell resolution, it is often observed that individual cells undergo different fates even when subject to identical conditions. This "noisy" phenotype is usually attributed to the inherent stochasticity of chemical reactions in the cell. Here we demonstrate how the observed single-cell heterogeneity can be explained by a cascade of decisions occurring at the subcellular level. We follow the postinfection decision in bacteriophage lambda at single-virus resolution, and show that a choice between lysis and lysogeny is first made at the level of the individual virus. The decisions by all viruses infecting a single cell are then integrated in a precise (noise-free) way, such that only a unanimous vote by all viruses leads to the establishment of lysogeny. By detecting and integrating over the subcellular "hidden variables," we are able to predict the level of noise measured at the single-cell level.

Wenn der Prozess der Zelle-Schicksal Bestimmung bei einzelligen Aufl&ouml;sung untersucht wird, wird es oft beobachtet, dass einzelne Zellen unterschiedliche Schicksale sogar unter identischen Bedingungen unterzogen werden. Diese &quot;laut&quot; Ph&auml;notyp wird in der Regel die inh&auml;rente Stochasticity chemischer Reaktionen in der Zelle zugeschrieben. Hier zeigen wir, wie die beobachtete einzellige Heterogenit&auml;t durch eine Kaskade von Entscheidungen, die auf der subzellul&auml;ren Ebene erkl&auml;rt werden kann. Wir folgen die Infektion Entscheidung in Bakteriophage Lambda bei Einzel-Virus Aufl&ouml;sung und zeigen, dass eine Wahl zwischen Zerst&ouml;rung und Lysogeny zun&auml;chst auf der Ebene des einzelnen Virus gemacht wird. Viral infection begins with the binding of a virus to a specific target on the surface of the host cell, followed by viral genome delivery into the host and a continuation of the infection process. Before binding occurs, the virus must first find its receptor by a process whose details are largely unknown. We applied high-resolution fluorescence microscopy and single-particle tracking to elucidate the target-finding process in bacteriophage Î» as it infects an Escherichia coli cell. By monitoring the motion of individual viruses through the early stages of infection, we identified a unique spatial focusing process that allows a virus to arrive from its initial random landing site to its destination at the cell pole. The search process is governed by the interaction between the virus and the LamB receptors, and by the spatial organization of the receptor network on the cell surface. Our findings allowed us to develop a theoretical model for the target-finding process that reproduces the key features observed in experiment. We discuss the possible implications of our findings for the process of viral receptor-finding in higher systems.

Single-virus tracking reveals a spatial receptor-dependent search mechanism.

Infection virale commence par la liaison d&#x26;#39;un virus &#xE0; une cible sp&#xE9;cifique sur la surface de la cellule h&#xF4;te, suivie par la d&#xE9;livrance du g&#xE9;nome viral dans l&#x26;#39;h&#xF4;te et une poursuite du processus de l&#x26;#39;infection. Avant la liaison se produit, le virus doit d&#x26;#39;abord trouver son r&#xE9;cepteur par un processus dont les d&#xE9;tails sont largement inconnues. Nous avons appliqu&#xE9; &#xE0; haute r&#xE9;solution microscopie par fluorescence et une particule de suivi pour &#xE9;lucider la cible processus de recherche dans le bact&#xE9;riophage &#x3BB; il infecte une cellule d&#x26;#39;Escherichia coli. Nos r&#xE9;sultats nous ont permis de mettre au point un mod&#xE8;le th&#xE9;orique pour le processus cible-constat qui reproduit les principales caract&#xE9;ristiques observ&#xE9;es dans l&#x26;#39;exp&#xE9;rience. Infezione virale comincia con l&#39;associazione di un virus ad un target specifico sulla superficie della cellula ospite, seguita dalla consegna del genoma virale in host e una continuazione del processo di infezione. Prima si verifica l&#39;associazione, il virus deve prima trovare il recettore di un processo in cui i dettagli sono in gran parte sconosciuti. Abbiamo applicato la microscopia di fluorescenza ad alta risoluzione e single-particle tracking per spiegare il processo di ricerca di destinazione nel batteriofago &lambda; come infetta una cellula di Escherichia coli. Monitorando il movimento dei singoli virus attraverso le prime fasi dell&#39;infezione, abbiamo individuato un processo unico di messa a fuoco spaziale che permette un virus arrivare dal suo sito di atterraggio casuale iniziale fino alla sua destinazione presso il polo della cellula. Il processo di ricerca &egrave; regolato dall&#39;interazione tra i virus e i recettori di agnello e di organizzazione spaziale della rete del recettore sulla superficie cellulare. I nostri risultati ci ha permesso di sviluppare un modello teorico per il processo di destinazione-constatazione che riproduce le caratteristiche chiave osservate nell&#39;esperimento. Discuteremo le possibili implicazioni dei nostri risultati per il processo di ricerca del recettore virale nei sistemi superiori.

La infecci&#xF3;n viral comienza con la uni&#xF3;n de un virus a una diana espec&#xED;fica en la superficie de la c&#xE9;lula hu&#xE9;sped, seguida por la entrega genoma viral en el hu&#xE9;sped y una continuaci&#xF3;n del proceso de infecci&#xF3;n. Antes de que se produce el enlace, el primer virus debe encontrar su receptor por un proceso cuyos detalles son desconocidos. Hemos aplicado de alta resoluci&#xF3;n microscop&#xED;a de fluorescencia y el seguimiento de una sola part&#xED;cula de dilucidar el objetivo proceso de investigaci&#xF3;n en el bacteri&#xF3;fago &#x3BB;, ya que infecta a una c&#xE9;lula de Escherichia coli. Al controlar el movimiento de los virus individuales a trav&#xE9;s de las etapas tempranas de la infecci&#xF3;n, hemos identificado un &#xFA;nico proceso espacial de enfoque que permite un virus para llegar desde su sitio inicial de aterrizaje aleatoria a su destino en el polo de la c&#xE9;lula. El proceso de b&#xFA;squeda se rige por la interacci&#xF3;n entre el virus y los receptores de cordero, y por la organizaci&#xF3;n espacial de la red de receptores en la superficie celular. Nuestros resultados nos ha permitido desarrollar un modelo te&#xF3;rico para el proceso de meta-hallazgo que reproduce las principales caracter&#xED;sticas observadas en el experimento. Se discuten las posibles implicaciones de nuestros resultados para el proceso de la replicaci&#xF3;n viral del receptor de investigaci&#xF3;n en sistemas de mayor.
