Commencez par mesurer la concentration de cardiomyocytes ventriculaires du rat néonatal à l’aide d’un hémocytomètre. Diluer la solution de suspension cellulaire à 3 000 cellules par millilitre, avec de l’alpha MEM contenant 10 % de FBS inactivé par la chaleur. Distribuez 100 microlitres de cardiomyocytes ventriculaires dilués dans chaque puits des plaques non adhésives à 96 puits de la cellule.
Centrifugez les cellules pendant cinq minutes à 100 G.Et cultivez-les pendant deux à trois jours dans l’incubateur de culture cellulaire, avec 5% de dioxyde de carbone à 37 degrés Celsius. Après l’incubation, aspirer le milieu de culture des boules de cardiomyocytes de rat ensemencées dans la plaque non adhésive à 96 puits. Ensuite, transférez les boules cardiomyocytaires de chaque puits dans un tube de 15 millilitres.
Centrifuger les billes cardiomyocytaires et aspirer le surnageant. Ensuite, lavez-les à l’aide d’un tampon ADS. Pour faciliter le suivi des cellules cardiomyocytaires chez le rat après la prise de greffe, colorer les cellules avec PKH26.
Avant de centrifuger les cellules, suivez le développement des boules de cardiomyocytes bidim en les observant régulièrement au microscope après l’ensemencement. Commencez à préparer l’appareil d’injection principal pour une injection de balle de cardiomyocytes néonatals à l’aide d’une aiguille de calibre 29 et de 50 millimètres de long munie d’une seringue. Connectez une aiguille de seringue de transfert d’énergie d’un millilitre de calibre 18 à un tube mince en polypropylène à faible capacité d’expansion lumineuse.
Aspirez l’eau à l’aide d’une seringue et retirez toutes les bulles de la seringue et des tubes connectés. Répétez l’opération pour une autre aiguille de calibre 18. Ensuite, connectez l’autre côté du tube de transfert d’énergie à l’aiguille de calibre 18 de la même seringue d’un millilitre.
Confirmez la réussite de la transition de puissance en poussant un piston de seringue vers le bas et un autre piston de seringue vers le haut. Une fois cela fait, placez la seringue dans un pousse-seringue. Ensuite, configurez le système de refroidissement en fixant le tube de cuivre étroitement enroulé à la cellule contenant une partie de la seringue d’injection de cellule.
Laisser 10 millimètres d’excédent de tuyau aux deux extrémités. Connectez le tube en cuivre au tube en plastique flexible avant de connecter les autres extrémités du tube en plastique à une pompe externe. Remplissez la conduite avec de l’eau de refroidissement et plongez l’excès de tube de cuivre dans de la glace pilée pour refroidir l’eau.
Ce système de refroidissement maintient la solution de suspension cellulaire dans le cylindre à environ 2 degrés Celsius.
