in vivo visualization of synaptic vesicles within drosophila larval segmental axons

Belysa mekanismerna för axonal transport har visat sig vara mycket viktigt för att avgöra hur fel i långväga transporter påverkar olika neurologiska sjukdomar. Defekter i denna viktiga process kan ha skadliga effekter på neuronal funktion och utveckling. Vi har utvecklat en dissektion protokoll som utformats för att utsätta<em&gt; Drosophila</em&gt; Larver segmentella nerver för att visa axonal transport i realtid. Vi har anpassat detta protokoll för live avbildning från en publicerad av Hurd och Saxton (1996) används för immunolocalizatin av larver segmentell nerver. Noggrann dissektion och rätt förutsättningar buffert är kritiska för att maximera livslängden på dissekeras larver. När ordentligt gjort, har dissekerat larver visat robust vesikler transporter i 2-3 timmar under fysiologiska förhållanden. Vi använder UAS-GAL4 metod<sup&gt; 1</sup&gt; Att uttrycka GFP-märkta APP eller synaptotagmin blåsor inom ett enda axon eller många axoner i larvernas segmentella nerver med hjälp av olika neuronala GAL4 förare. Andra fluorescerande taggade markörer, exempelvis mitokondrierna (MitoTracker) eller lysosomer (LysoTracker), kan även appliceras på larverna innan visning. GFP-vesikler rörelse och partikel rörelser kan visas samtidigt med separata våglängder.
