<p> Cellen werden geïmmobiliseerd en patronen binnen de microgrooved substraten in een microfluïdische apparaat. Het proces van apoptose cellen blootgesteld aan waterstofperoxide werd waargenomen in real-time en geanalyseerd met behulp van Annexine V en propidiumjodide. Zo zou deze microfluïdische apparaat met Microgroove kanalen bruikbaar zijn voor high-throughput screening van geneesmiddelen. </p>

a microfluidic device with groove patterns for studying cellular behavior

<p class="jove_content"> Beschrijven we een microfluïdische apparaat met microgrooved patronen voor het bestuderen van cellulaire gedrag. Deze microfluïdische platform bestaat uit een top vloeibare kanaal en een bodem microgrooved substraat. Te fabriceren van de microgrooved kanalen, was een top poly (dimethylsiloxaan) (PDMS) mal met de indruk van de microfluïdische kanalen op elkaar afgestemd en gebonden aan een microgrooved substraat. Met behulp van dit apparaat, de muis fibroblast cellen werden geïmmobiliseerd en patronen binnen de microgrooved substraten (25, 50, 75 en 100 micrometer breed). Om apoptose in een microfluïdische apparaat studie, was de media die waterstof peroxide, Annexine V en propidiumjodide doorbloed in de vloeibare kanaal voor 2 uur. We vonden dat de cellen blootgesteld aan de oxidatieve stress apoptotische werd. Deze apoptotische cellen werden bevestigd door Annexine V die gebonden aan phosphatidylserine bij de buitenste leaflet van de plasmamembraan tijdens het apoptose-proces. Met behulp van deze microfluïdische apparaat met microgrooved patronen, was het apoptose-proces waargenomen in real-time en geanalyseerd met behulp van een omgekeerde microscoop met een incubatiekamer (37 ° C, 5% CO <sub>2).</sub> Daarom kon dit microfluïdische apparaat opgenomen met microgrooved substraten nuttig zijn voor het bestuderen van de cellulaire gedrag en het uitvoeren van high-throughput screening van geneesmiddelen. </p>
