Die Molecular Devices &quot;FlexStation 3 ist ein Tischgerät Multi-Mode Mikroplatten-Reader mit einer automatisierten Fluoreszenz-Messung in Multi-Well-Platten. Es ist ideal für mittel-bis High-Throughput-Screens in einem akademischen Umfeld. Es verfügt über einen integrierten Fluid-Transfer-Modul mit einer Multi-Channel-Pipette und die Maschine liest eine Spalte in einer Zeit, zur Fluoreszenz Veränderungen einer Vielzahl von fluoreszierenden Reagenzien-Monitor ausgestattet. HEK293-Zellen transient oder stabil exprimieren menschliches TRPA1 Kanäle, in 96-well-Platten kultiviert, sind mit einem Ca 2 +-sensitiven Fluoreszenzfarbstoff Fluo-4, und Echtzeit-Fluoreszenz-Veränderungen in diesen Zellen geladen werden, vor und während der Anwendung gemessen ein TRPA1 Agonist mit dem FLEX-Modus des FlexStation 3. Die Wirkung eines mutmaßlichen TRPA1 Antagonisten wurde ebenfalls untersucht. Die Daten stammen aus der SoftMax Pro-Software übertragen, um Konzentrations-Wirkungs-Beziehungen von TRPA1 Aktivatoren und Inhibitoren konstruieren.
