hESCs zijn een ideaal platform om verschillende celtypes te bestuderen en te houden opmerkelijke potentieel voor klinische vertaling. Menselijke embryonale stamcellen (hESC &#39;s) en geïnduceerde pluripotente stamcellen (iPSCs) zijn ongedifferentieerde, pluripotente cellen die in staat onbeperkt zelf-vernieuwing en multi-lijn differentiatie. hESCs zijn succesvol onderscheiden in volwassen en functionele subsets van iedere kiem lagen, waaronder cellen van het hematopoietische systeem 1-3. Natural killer (NK) cellen zijn lymfocyten van het aangeboren immuunsysteem, dat kan worden afgeleid uit hESCs door de vorming van embryoid instanties (EBS) 4,5 of co-cultuur met stromale cellijnen 1,2,6-8. NK-cellen bezitten anti-virale en anti-tumor vermogens en hebben het potentieel om effectief te zijn tegen een breed spectrum van tumoren, omdat zij niet gevalideerd antigeenstimulatie hun effectorfuncties uitvoeren. Aldus hESC-afgeleide NK cellen een aantrekkelijke bron van cellen voor immunotherapie. Daarnaast is de afleiding van NK-cellen uit hESCs biedt een genetisch vatbaar systeem om normale ontwikkeling studeren &lt;em&gt; in vitro. De mogelijkheid om efficiënt genoeg leiden NK cellen die in staat om patiënten blijft een belangrijke hinderpaal voor klinische toepassing en, in mindere mate, een beperking voor uitgebreide preklinische in vivo studies van NK cel ontwikkeling en anti-kanker functies. Hier gebruiken we een spin EB benadering van hematopoietische voorlopercellen ontlenen hESCs 4,5,10. Na 11 dagen de spin EBS worden overgebracht naar NK celkweek met of zonder feeders voor 28 dagen. Na 4 weken in NK cel kweekmedium, NK-cellen zijn transferred aan co-kweek met K562 cellen gemodificeerd om expressie membraangebonden interleukine 21 (IL-21), die als kunstmatige antigeenpresenterende cellen (aAPCs). Aanpassen van een protocol voor de uitbreiding van perifere bloed NK-cellen met behulp van deze kunstmatige APC&#39;s 11,12, zijn we in staat om NK-cellen uit te breiden 2-logs met behoud van een rijp fenotype en cytotoxische capaciteiten. Dit proces van ontwikkeling en uitbreiding voldoende hESC-afgeleide cellen NK voor uitgebreide in vivo karakterisering. Voor in vivo studies, zijn we in staat om niet-invasief bewaken lange termijn innesteling en kinetiek van geïnjecteerde vuurvliegluciferase uiten (Fluc +), hESC-afgeleide NK-cellen met behulp van bioluminescentie beeldvorming. Bovendien zijn wij in staat om NK-cel interacties te volgen met tumorcellen met een dual, bioluminescente of fluorescerende beeldvorming regeling. Een eerdere studie van onze groep gebruikt bioluminescentie beeldvorming in een anti-tumor model om tumorprogressie en cle volgenarance van Fluc + K562-cellen in vivo 7. Nu, door de engineering van onze hESCs aan uitdrukkelijke vuurvliegluciferase 13,14 wij kunnen de biologische verdeling van en handel in NK-cellen te volgen naar K562 tumorcellen dat de onlangs gekenmerkt fluorescerend eiwit, turboFP650 15 drukken. We hebben dit dubbele reporter systeem gekozen om gelijktijdig volgen twee celpopulaties in vivo (figuur 1). De meeste dual imaging modellen zijn dual-luciferase systemen geweest, maar deze systemen kunnen technisch gezien een uitdaging vanwege de levering eisen van coelenterazine, de ondergrond die nodig zijn voor de expressie van de meeste Renilla en Gaussia luciferase reporters 16-18. Fluorescerende reporters hebben toegestaan ​​eenvoudige controle van veel cellijnen en constructies in vitro, maar heeft beperkt succes voor in vivo beeldvorming moest vanwege de overlap tussen weefsel en bont autofluorescentie en de emissie spectra van vele commonly gebruikte fluorescerende verslaggevers waaronder GFP, DsRed en tdTomato 15,19. development, expansion, and in vivo monitoring of human nk cells from human embryonic stem cells (hescs) and induced pluripotent stem cells (ipscs)

We stellen een werkwijze voor het afleiden van natuurlijke killer (NK) cellen ongedifferentieerd hESCs en iPSCs met een feeder-free benadering. Deze methode leidt tot hoge niveaus van NK-cellen na 4 weken kweek en verder log2 uitbreiding met kunstmatige antigeenpresenterende cellen ondergaan. hESC-en IPSC-afgeleide NK-cellen ontwikkeld in dit systeem hebben een volwassen fenotype en functie. De productie van grote hoeveelheden van genetisch beïnvloedbare NK-cellen kan zowel voor de mechanistische basis als anti-tumor studies. Expressie van vuurvliegluciferase in hESC-afgeleide NK-cellen kan een niet-invasieve benadering van NK cellen aanslaan, distributie, en de functie volgt. We hebben ook een dual-imaging regeling die het mogelijk maakt afzonderlijke bewaking van twee verschillende celpopulaties hun interacties duidelijker karakteriseren in vivo te beschrijven. Deze methode van afleiding, expansie, en dual in vivo beeldvorming biedt een betrouwbare aanpak voor de productie van NK-cellen en hun evaluatieatie die nodig zijn om de huidige NK cel adoptie therapieën te verbeteren is.
