hESCs er en ideell plattform for å studere ulike celletyper og holder bemerkelsesverdig potensial for klinisk oversettelse. Vi bruker en definert, spin EB tilnærming å skille hESC / iPSCs til blodkreft stamceller. Humane embryonale stamceller (hESCs) og induserte pluripotente stamceller (iPSCs) er udifferensierte, pluripotente celler i stand til ubegrenset selvfornyelse og multi-avstamning differensiering. hESCs har blitt differensiert i modne og funksjonelle undergrupper av hver bakterie lag, herunder celler av hematopoietisk system 1-3. Natural killer (NK) celler er lymfocytter i det medfødte immunsystemet som kan utledes fra hESCs ved dannelsen av embryoid organer (EBS) 4,5 eller co-kultur med stromal cellelinjer 1,2,6-8. NK-celler har anti-viral og anti-tumor egenskaper, og har potensial for å være effektive mot et bredt spekter av maligniteter, ettersom de ikke krever forutgående antigenstimulering å utføre sine effektorfunksjoner. Dermed hESC-avledet NK-celler er en attraktiv kilde til celler for immunterapi. Nå, ved å konstruere våre hESCs å uttrykke ildflue luciferase 13,14 kan vi følge fordelingen og trafficking av NK-celler til K562 kreftceller som uttrykker den nylig preget fluorescerende protein, turboFP650 15. Vi har valgt denne dual reporter system for å følge samtidig de to cellepopulasjoner in vivo (figur 1). Mest dual bildebehandling modeller har vært dual-luciferase systemer, men disse systemene kan være teknisk utfordrende på grunn av levering krav coelenterazine, underlaget kreves for uttrykk for de fleste Renilla og Gaussia luciferase reportere 16-18. Denne bekymringen har oppmuntret utviklingen av langt-røde fluoriserende proteiner, som gir mulighet for bedre vev penetrans og høyere spesifikk signal i forhold til bakgrunnen 15,19. development, expansion, and in vivo monitoring of human nk cells from human embryonic stem cells (hescs) and induced pluripotent stem cells (ipscs)

Vi presenterer en metode for å utlede natural killer (NK) celler fra udifferensiert hESCs og iPSCs ved hjelp av en mater-fri tilnærming. Denne fremgangsmåte gir opphav til høye nivåer av NK-celler etter 4 ukers kultur og kan gjennomgå ytterligere to-log utvidelse med kunstige antigenpresenterende celler. hESC-og iPSC-derived NK-celler utviklet i dette systemet har en moden fenotype og funksjon. Fremstillingen av et stort antall genetisk modifiserbare NK-celler er anvendelig for både enkle mekanistiske så vel som anti-tumor-studier. Ekspresjon av ildflue luciferase i hESC-avledede NK-celler tillater en ikke-invasiv metode for å følge NK-celle innpodingen, distribusjon og funksjon. Vi beskriver også en dual avbildning-ordning som gjør det mulig separat overvåking av to forskjellige cellepopulasjoner til mer utpreget karakterisere deres interaksjoner in vivo. Denne metoden for avledning, ekspansjon, og dual in vivo avbildning gir en pålitelig metode for å fremstille NK-celler og deres evaluasjon som er nødvendig for å forbedre dagens NK-celle adoptive terapi.

Generering av blodkreft stamceller ved hjelp av spin EB tilnærming gir mulighet for optimal NK celle utvikling fra hESCs og iPSCs. Som vist i figur 2, dag 11 spin EBS inneholde høye prosenter av stamceller uttrykker CD34, CD45, CD43 og CD31. Høye nivåer av CD34 og CD45 tillater direkte overføring til NK forhold uten behov for sortering eller støtte stromaceller. Hvis det er suboptimal spin EB differensiering, er det anbefalt at stromale celler slik som EL08-1D2 er brukt i de sekundære NK forhold....
