Las células madre embrionarias humanas (hESCs) y células madre pluripotentes inducidas (iPSCs), son células indiferenciadas pluripotentes capaces de auto-renovación y multi-linaje diferenciación ilimitada. hESCs se han diferenciado con éxito en subconjuntos maduras y funcionales de cada capa germinal, incluyendo las células del sistema hematopoyético 1-3. Las células asesinas naturales (NK) son los linfocitos del sistema inmune innato que se puede derivar de hESCs por la formación de cuerpos embrioides (EB) de 4,5 o co-cultivo con líneas de células del estroma 1,2,6-8. Se presenta un método para derivar las células asesinas naturales (NK) de hESCs indiferenciadas y iPSCs utilizando un enfoque de enlace de forma gratuita. Este método da lugar a altos niveles de células NK después de 4 semanas la cultura y puede someterse a una mayor expansión 2-registro con las células presentadoras de antígeno artificiales. células NK y células madre IPSC derivado desarrollados en este sistema tienen un fenotipo y función maduro.
