Het doel van deze experimenten is om kwantitatief tijdsverloop gegevens over de groei en gen expressie dynamiek van verzwakte S. genereren typhimurium bacteriële kolonies groeien binnen tumoren. We gegenereerde model xenograft tumoren bij muizen door subcutane injectie van een mens eierstokkanker cellijn, OVCAR-8 (NCI DCTD Tumor Repository, Frederick, MD). We getransformeerde verzwakte stammen van S. typhimurium bacteriën (ELH430: SL1344 PhoPQ-1) met een constitutief tot expressie luciferase (luxCDABE) plasmide voor visualisatie 2. Deze stammen specifiek koloniseren tumoren terwijl hoofdzaak niet-virulent blijven de muis 1. Zodra meetbare tumoren werden vastgesteld, werden bacteriën intraveneus geïnjecteerd via de staartader met wisselende dosering. Tumor-gelokaliseerde werd bacteriële genexpressie in real tijd in de loop van 60 uur met eenin vivo imaging-systeem (IVIS). Op elk tijdspunt werden tumoren uitgesneden, gehomogeniseerd en uitgeplaat op bacteriële kolonies correlatie met genexpressiedata kwantificeren. Tezamen zijn deze gegevens levert een kwantitatieve maat van de in vivo expressie en gen dynamiek van bacteriën groeien binnen tumoren.

Met behulp van dit protocol, zijn we in staat om gegevens over de in vivo groei en gen dynamiek expressie van tumor gerichte bacteriën genereren. De algehele werkstroom wordt samengevat in figuur 1.
