Due fotoni di imaging di indicatori di calcio (geneticamente codificate come GCaMP5 7 o R-GECO 8, o coloranti sintetici come OGB o Fluo4) è emersa come un potente metodo di misurazione dell&#39;attività neuronale nel comportarsi topi 1-6. Esso consente la misurazione simultanea dell&#39;attività di centinaia di cellule a potenziale risoluzione azione quasi unico, fino a circa 800 micron sotto la superficie del cervello 9,10. two-photon calcium imaging in mice navigating a virtual reality environment

Negli ultimi anni, l&#39;imaging a due fotoni è diventato uno strumento prezioso in neuroscienze, in quanto consente di misurazione cronica della attività delle cellule geneticamente identificati durante comportamento 1-6. Qui si descrivono metodi per eseguire due fotoni di imaging in topo corteccia mentre l&#39;animale si sposta un ambiente di realtà virtuale. Ci concentriamo sugli aspetti delle procedure sperimentali che sono fondamentali per l&#39;imaging in un animale si comporta in un ambiente virtuale illuminata. I problemi principali che sorgono in questa configurazione sperimentale che noi qui l&#39;indirizzo sono: ridurre al minimo gli artefatti da movimento legati al cervello, riducendo al minimo perdite luce dal sistema di realtà virtuale di proiezione, e minimizzando laser indotto danni ai tessuti. Forniamo anche software di esempio per controllare l&#39;ambiente di realtà virtuale e di fare il monitoraggio allievo. Con queste procedure e risorse dovrebbe essere possibile convertire un microscopio a due fotoni convenzionale per uso nel comportarsi topi.

La qualità dell&#39;immagine in due fotoni Imaging di calcio di popolazioni di cellule marcate con un GECI dipende in gran parte la qualità della finestra protesi cranica.
