Ce travail est un projet d&#39;ingénierie / de la biologie synthétique métabolique dans lequel les cellules végétales sont modifiées pour exprimer une protéine rapporteur dans plusieurs organites mais avec une seule construction d&#39;ADN. Une approche pour cibler des protéines à plus d&#39;un emplacement de clonage implique de multiples copies génétiques, chacune contenant un peptide de localisation différente. Chaque copie doit être introduite par retransformation successifs, ou encore, par rétrocroisement transformations simples 1. Il s&#39;agit de clonage supplémentaire, et est limitée par une balise de localisation par terminus. Une autre façon de localiser une protéine à plusieurs endroits à travers l&#39;épissage alternatif 2-5. ARN est transcrit à partir d&#39;un seul gène, mais différentes copies de la transcription sont traitées différemment, souvent dans plus d&#39;un titre par cellule. Cela peut entraîner plus d&#39;un ARN messager dans la cellule transcrit à partir d&#39;un gène unique. Ces différents messengeARN R peut coder pour différentes isoformes de la même protéine, ou dans le cas d&#39;un décalage du cadre, une protéine différente. Les constructions ont été conçues dans le travail précédent 11, et ont été clones en utilisant Gibson assemblage, un procédé de clonage indépendant de la ligature qui ne nécessite pas d&#39;enzymes de restriction. Les plasmides résultants ont été introduits dans les cellules épidermiques des feuilles de Nicotiana benthamiana avec un protocole Gene Gun modifié. Enfin, les feuilles transformées ont été observées par microscopie confocale.

L&#39;effort de conception est le résultat d&#39;une planification importante. Roman de ce projet est l&#39;utilisation de l&#39;épissage alternatif de créer un pré-ARNm qui est traduit en protéines exprimées de façon différentielle. Ces protéines sont exprimées dans différents organelles, dans ce cas, le chloroplaste, des peroxysomes et / ou cytosol.
