Dette arbeidet er en metabolsk ingeniør / syntetisk biologi prosjekt der planteceller er konstruert for å uttrykke en reporter protein i flere organeller, men med bare en enkelt DNA-konstruksjon. En tilnærming for å målrette proteiner til mer enn ett sted innebærer kloning flere genetiske eksemplarer, hvert inneholdende en annen lokalisering peptid. Hvert eksemplar må være introdusert av suksessive retransformation, eller alternativt, ved tilbakekryssing enkelt forvandler en. Dette innebærer ytterligere kloning, og er begrenset av en lokaliseringssignal per terminus. En annen måte til å lokalisere et protein til flere steder er gjennom alternativ spleising 2-5. Ettersom disse nettstedene blir bedre beskrevet, de åpner opp muligheter for engineering. Plant metabolske ingeniører står overfor en utfordring når de uttrykker et protein i flere organeller. For et protein som er til å være lokalisert til flere organelle, må ingeniøren å klone det samme genet flere ganger, hver med en egen signalsekvens dirigere det til organeller av interesse. For et enkelt gen i tre organeller, er dette bare tre gener. Men for en seks-genet metabolismevei, utvides dette til 18 gener, en betydelig kloning innsats. Kombinere flere lokaliserings sekvenser i en enkelt, alternativt-skjøtes genet tegnreduserer ificantly dette arbeidet. For eksempel, re-engineering fotorespirasjon 7,8 og isoprenoid syntese 9,10 innebærer både kloroplaster og peroxisomes. I vårt tilfelle tok vi nytte av skjøtesteder som observert i en naturlig Arabidopsis thaliana system beskrevet tidligere seks.
