Daarnaast MCs spelen een belangrijke rol in aangeboren en adaptieve immuniteit, zowel in de afweer tegen bacteriën en parasieten en door onderdrukking van immuunreacties, bijvoorbeeld het induceren allotransplantaat tolerantie 5,6. investigating mast cell secretory granules; from biosynthesis to exocytosis

Mestcellen (MC) zijn secretoire cellen van het immuunsysteem die de fysiologische en pathologische functies vervullen door het vrijgeven van voorgevormde en nieuw gesynthetiseerde allergische, ontstekings- en immuunregulerende mediatoren. MCs &#39;mediators beïnvloeden meerdere weefsels en organen culminerend in allergische en immuunreacties. De synthese, opslag en afgifte van de MC mediators zijn sterk gereguleerd. De voorgevormde bemiddelaars zijn verpakt in cytoplasmatische secretoire korrels (SG) die fuseren met de plasmamembraan en de inhoud ervan vrij te maken door gereguleerde exocytose. We presenteren een protocol gebaseerd op de co-expressie van een gen van belang met een reportergen dat is gericht op de SG en opgenomen in een reguleerbare wijze naast de endogene SG mediators. Het protocol maakt hoge resolutie vierdimensionale confocale analyses van de MC SG&#39;s en het toezicht op hun tijdlijn van biogenese naar aanleiding exocytose. Dus, door dit protocol voor screening van genen interest hun fenotypische en functionele effect maakt ontcijferen moleculaire mechanismen die de biogenese en exocytose MC SG bepalen en identificeren van de betrokken regulatoren. Daarbij moet verder inzicht in de cellulaire mechanismen die verantwoordelijk zijn voor MCs functie in gezondheid en ziekte worden.

Vanwege de lage transfectie efficiëntie van MC, genetische manipulaties waarschijnlijk geen invloed op uitlezingen van gemiddeld secretie gemeten door endogene mediators SG verlaten. Niettemin, door het instellen volledige co-expressie van het reportergen NPY-mRFP en de gecotransfecteerde plasmide in dezelfde cellen, controle van NPY-mRFP resultaten bewaken uitsluitend de celpopulatie die het gen van belang tot expressie brengt.
