investigating mast cell secretory granules; from biosynthesis to exocytosis

마스트 셀 (MC)를 형성 미리 새로 합성, 알레르기 염증 및 면역 매개 물질을 방출하여 생리 학적 및 병리학 적 기능을 수행하는 면역 체계의 분비 세포이다. MC들 &#39;매개 알레르기 및 면역 반응에서 남중 여러 조직과 장기에 영향을 미친다. MC 매개체의 합성, 저장 및 방출은 매우 통제된다. 예비 형성된 매개체는 세포막과 융합 세포 외 유출에 의해 규제 대상 콘텐츠를 해제 세포질 분비 과립 (SG)에서 포장된다. 우리는 SG를 대상으로하고 내인성 SG 매개체와 함께 조절 된 방식으로 방출된다 리포터 유전자를 사용하여 원하는 유전자의 공동 발현에 기초하여 프로토콜을 제시한다. 프로토콜은 MC SG를 분석하고 트리거 세포 외 유출에 생합성에서 자신의 타임 라인을 모니터링 높은 해상도 네 차원 공 초점을 수 있습니다. 따라서, 간 유전자의 스크리닝이 프​​로토콜을 이용하여자신의 표현형 및 기능에 미치는 영향에 대한 추정치는 MC SG를의 생합성 및 세포 외 유출을 제어하는​​ 분자 메커니즘을 해독과 관련된 규제를 식별 허용한다. 이에 따라, 건강과 질병에서의 MC 기능을 설명 세포 메커니즘으로 더 통찰력이 제공되어야한다.
