Vi beskriver en innovativ strategi som kombinerar kvantifiering av MCs exocytos och fyra (x, y, z, t) dimensions kvantifieringar av tids förfallit tre-dimensionell avbildning av SGS i levande celler med användning av en reportergen för exocytos. Mastceller (MC) är immunceller som är mest känd för sin medverkan i allergiska och inflammatoriska reaktioner som artrit, astma, eosinofil esofagit, kronisk dermatit och anafylaktisk chock 1,2 samt andra patologier inklusive kranskärlssjukdom 3,5 och cancer 3,4. Dessutom MCs spelar viktiga roller i medfödda och adaptiva immuniteten, både i värd försvar mot bakterier och parasiter och genom undertryckande av immunsvar, exempelvis inducera allograft tolerans 5,6. MCs härstammar från benmärgen, utvecklas från CD34 + / CD117 + pluripotenta stamceller 7. Engagerat benmärgs MC gångare släpps ut i blodomloppet och migrera in i de perifera vävnaderna lokalisera främst inom bindväv och epitelytor 8. Mognad och terminal differentiering så småningom uppnås under påverkan of cytokiner inom omgivande miljö 8,9. MCs kan aktiveras av ett allergen (antigen, Ag), vars möte resulterade i generering av immunoglobulin E (IgE) typ antikroppar. MCs aktiveras också av många av deras egna frigjorda mediatorer, som ytterligare förstärker det inflammatoriska svaret. MCs är packade med sekretoriska granuler (SGS) som innehåller immunoregulatoriska mediatorer, däribland vasoaktiva aminer, såsom histamin och serotonin (hos gnagare), proteoglykaner, proteaser, såsom chymas och tryptas, vaskulär endotel tillväxtfaktor och flera cytokiner och kemokiner 8,9. Dessa mediatorer är &quot;redo att gå&quot; och en gång MCs aktiveras genom en lämplig stimulans dessa mediatorer frigörs från cellerna genom reglerat exocytos (degranulering) på några sekunder till minuter 18,19. Svårigheten att genetiskt manipulera primära och odlade MCs har försvårat försöken att klarlägga mekanismerna bakom MC degranulering, vilket förblev dåligt lösas. För att lösa detta problem som vi utvecklat en reporter baserad analys av samtransfektera den mucosal mastcellinjen, råtta basofila leukemi (RBL) -2H3 (nedan kallade RBL) eller benmärg härledda MCs (BMMCs) 30 med en gen av intresse och neuropeptid Y (NPY) smält till mono RFP (mRFP), som en SG reporter. NPY har tidigare visat att rekapitulera beteende endogena SG markörer i andra system. Dessutom, på grund mRFP fluorescens är pH-okänsligt, expression av NPY-mRFP möjliggör visualisering av de sura SGs samt kvantitativ bedömning av exocytos genom att använda 96-brunnars plattor och en fluorescensplattläsare. investigating mast cell secretory granules; from biosynthesis to exocytosis

Mastceller (MC) är sekretoriska celler i immunsystemet som utföra deras fysiologiska och patologiska funktioner genom att släppa förformade och nysyntetiserade allergiska, inflammatoriska och immunregulatoriska mediatorer. MCS medlare påverka flera vävnader och organ som kulminerar i allergiska och immunsvar. Syntesen, lagring och frisättning av MC medlarna är starkt reglerad. De förformade mediatorer är förpackade i cytoplasmiska sekretoriska granuler (SG) som smälter med plasmamembranet och frigör deras innehåll genom reglerad exocytos. Vi presenterar ett protokoll, baserat på samtidigt uttryck av en gen av intresse med en reporter gen som är riktad till SGS och släpps i ett reglerat sätt vid sidan av de endogena SG medlare. Protokollet möjliggör hög upplösning fyrdimensionell konfokal analyser av MC SGS och övervaka deras tidslinje från biogenes till utlöst exocytos. Således använder detta protokoll för screening gener av interest för deras fenotypiska och funktionella påverkan tillåter dechiffrera de molekylära mekanismer som styr biogenes och exocytos av MC SGS och identifiera de tillsynsmyndigheter som berörs. Därmed bör ytterligare insikter i de cellulära mekanismer som står för MCs funktion i hälsa och sjukdom lämnas.

På grund av den låga transfektionseffektiviteten av MCs, är det osannolikt att lämna en inverkan på avläsning av genomsnittlig sekre mätt genom endogena SGS medlare genetiska manipulationer. Ändå, genom att upprätta fullständig samexpression av reportergenen NPY-mRFP och den samtransfekterade plasmiden vid samma celler, övervakning av NPY-mRFP resultat i övervakningen enbart cellpopulationen som uttrycker genen av intresse.
