Monstervoorbereiding is een belangrijke stap in een analytisch proces. Vooral het verwijderen van matrixeffecten, vermindering van storingen en de verrijking van het analyt nodig om nauwkeurige resultaten te bereiken en lage detectiegrenzen. Hormoonverstorende stoffen (EDC&#39;s) zijn van bijzonder belang vanwege hun actie op de levende organismen, zelfs wanneer aanwezig op zeer lage niveaus in het milieu. Het natuurlijke hormoon 17β-estradiol is aanwezig op de EU watervervuiling watch list en gevoelig te worden toegevoegd aan de lijst van prioritaire stoffen die vallen onder de Europese Kaderrichtlijn Water. Vaste fase-extractie (SPE) is gewoonlijk voor de analyse van kleine verontreinigingen in het water, zowel chemisch 1-5 (chromatografie, massaspectrometrie) en immunologische 6-9 detectiemethoden. De laatste kreeg belang op het gebied van toezicht op het milieu, zoals immunoassays zijn verkrijgbaar in grote verscheidenheid van formaten, zijn specifiek voor de tarGebruik analyt, en bereiken lage aantoonbaarheidsgrenzen. 6, 7, 10, 11 Diverse enzymgekoppelde immunosorbent assays (ELISA) zijn commercieel verkrijgbaar en in staat om meerdere monsters tegelijkertijd te analyseren op een multi-well plaat. De procedure bestaat uit achtereenvolgende reactiestappen die enkele uren kan duren. Het uiteindelijke reactieproduct kan optisch worden gedetecteerd om de concentratie van het doelwitmolecuul op basis van een ijkcurve te bepalen. Klassieke SPE procedures omvatten sorbens pre-conditioning, monstername, wassen, elutie en indampen van het eluens. Het oplosmiddel gebruikt voor het verdunnen van het extract wordt gekozen afhankelijk van de detectiemethode. Voor immunologische werkwijzen, de hoeveelheid organisch oplosmiddel beïnvloedt sterk de gevoeligheid van de werkwijze. 12 Naast het herstel en de pre-concentratie uitvoeringen, de werkwijze moet ook eenvoudig en kostenefficiënt. Automatisering van de procedure helpt tot-mens-gerelateerde fouten te verminderen. In onze vorige werk 13 we ons prototype voor geautomatiseerde SPE geïntroduceerd, en onze methode werd toegepast op de analyse van het natuurlijke hormoon 17β-estradiol in zee watermonsters. Met de huidige video willen we de technische voordelen van onze methode vergeleken met de traditionele off-line en on-line SPE, en de bijzondere compatibiliteit met detectie door immuno-reacties te markeren. We beschrijven het protocol toegepast watermonsters voor de detectie van 17β-oestradiol. SPE wordt uitgevoerd met octadecyl-silicagel (C18) sorbent fase en elutie wordt uitgevoerd met verdunde methanol.

Een nieuwe methode voor vastefase-extractie (SPE) milieu watermonsters voorgesteld. De ontwikkelde prototype is kostenefficiënt en gebruiksvriendelijk, en maakt het mogelijk om een ​​snelle, geautomatiseerde en eenvoudige SPE voeren. De pre-geconcentreerde oplossing is geschikt voor analyse door immunoassay, met een laag gehalte organisch oplosmiddel.
